This monograph is concerned with mathematical aspects of compartmental an alysis. In particular, linear models are closely analyzed since they are fully justifiable as an investigative tool in tracer experiments. The objective of the monograph is to bring the reader up to date on some of the current mathematical prob lems of interest in compartmental analysis. This is accomplished by reviewing mathematical developments in the literature, especially over the last 10-15 years, and by presenting some new thoughts and directions for future mathematical research. These notes started as a series of lectures that I gave while visiting with the Division of Applied ~1athematics, Brown University, 1979, and have developed in to this collection of articles aimed at the reader with a beginning graduate level background in mathematics. The text can be used as a self-paced reading course. With this in mind, exercises have been appropriately placed throughout the notes. As an aid in reading the material, the e~d of a proof is indicated by ~. Sub section titles are utilized to make it easier for the reader to skim over detailed material on a first reading and make the entire manuscript somewhat more accessible, especially to nonmathematicians in the biosciences. In February 1985 a small international meeting of scientists took place at the recreation resort of the Polish Academy of Sci ences in Mogilany, near Cracow, Poland. The initiative for holding the workshop came from a working meeting on mathematical immunology and related topics at the International Institute for Applied Sys tems Analysis in Laxenburg, Austria, in November 1983. In addition to representatives of IIASA, delegates of the IIASA National Member Organizations (NMO) of Czechoslovakia, Italy, and the soviet Union took part in that working meeting. The participants came to the conclusion that IIASA could play an important role in facilitating the development of research in this field. The first step that they recommended to I IASA was to organize a workshop on mathematical immunology. The purpose of the workshop was to review the progress that has been made in applying mathematics to problems in immunology and to explore ways in which further progress might be achieved, especially by more efficient interactions between scientists working in mathematical and experimental immunology. Some National Member Organizations contributed to the success of the workshop by nominat ing further participants working in this or related fields. For instance, thanks to a suggestion of the British NMO, the meeting also included analyses of the interactions between the immune state of a population and epidemiological phenomena. There were 33 participants at Mogilany from 11 countries, namely Canada, Czechoslovakia, Federal Republic of Germany, Hungary, Japan, Netherlands, Poland, Sweden, united Kingdom, USA, and USSR.

These notes have grown from a series of seminars given at Leeds between 1972 and 1975. They represent an attempt to gather together the different kinds of model which have been proposed to account for the stochastic activity of neurones, and to provide an introduction to this area of mathematical biology. A striking feature of the electrical activity of the nervous system is that it appears stochastic: this is apparent at all levels of recording, ranging from intracellular recordings to the electroencephalogram. The chapters start with fluctuations in membrane potential, proceed through single unit and synaptic activity and end with the behaviour of large aggregates of neurones: L have chgaen this seque~~e\/~~';uggest that the interesting behaviourr~f :the nervous system - its individuality, variability and dynamic forms - may in part result from the stochastic behaviour of its components. How weIl can we model experimental observations of the peripheral auditory system'? What theoretical predictions can we make that might be tested'? It was with these questions in mind that we organized the 1985 Mechanics of Hearing Workshop, to bring together auditory researchers to compare models with experimental observations. Tbe workshop forum was inspired by the very successful 1983 Mechanics of Hearing Workshop in Delft [1]. Boston University was chosen as the site of our meeting because of the Boston area's role as a center for hearing research in this country. We made a special effort at this meeting to attract students from around the world, because without students this field will not progress. Financial support for the workshop was provided in part by grant BNS- 8412878 from the National Science Foundation. Modeling is a traditional strategy in science and plays an important role in the scientific method. Models are the bridge between theory and experiment. Tbey test the assumptions made in experimental designs. They are built on experimental results, and they may be used to test hypotheses and predict experimental results. Tbe latter is the scientific method at its best. Cochlear function is very complicated. For this reason, models play animportant role. One goal of modeling is to gain understanding, but the necessary mathematical tools are often formidably complex. An ex am pie of this is found in cochlear macromechanics.

The 18 research articles of this volume discuss the major themes that have emerged from mathematical and statistical research in the epidemiology of HIV. The opening paper reviews important recent contributions. Five sections follow: Statistical Methodology and Forecasting, Infectivity and the HIV, Heterogeneity and HIV Transmission Dynamics, Social Dynamics and AIDS, and The Immune System and The HIV. In each, leading experts in AIDS epidemiology present the recent results. Some address the role of variable infectivity, heterogeneous mixing, and long periods of infectiousness in the dynamics of HIV; others concentrate on parameter estimation and short-term forecasting. The last section looks at the interaction between the HIV and the immune system.

The past forty years have been the stage for the maturation of mathematical biolo~ as a scientific field. The foundations laid by the pioneers of the field during the first half of this century have been combined with advances in ap plied mathematics and the computational sciences to create a vibrant area of scientific research with established research journals, professional societies, deep subspecialty areas, and graduate education programs. Mathematical biology is by its very nature cross-disciplinary, and research papers appear in mathemat ics, biology and other scientific journals, as well as in the specialty journals devoted to mathematical and theoretical biology. Multiple author papers are common, and so are collaborations between individuals who have academic bases in different traditional departments. Those who seek to keep abreast of current trends and problems need to interact with research workers from a much broader spectrum of fields than is common in the traditional mono-culture disciplines. Consequently, it is beneficial to have occasions which bring together significant numbers of workers in this field in a forum that encourages the exchange of ideas and which leads to a timely publication of the work that is presented. Such an occasion occurred during January 13 to 16, 1990 when almost two hun dred research workers participated in an international conference on Differential Equations and Applications to Biology and Population Dynamics which was held in Claremont.

These notes are based on (i) a series of lectures that I gave at the 14th Biennial Seminar of the Canadian Mathematical Congress held at the University of Western Ontario August 12-24, 1973 and (li) some of my lectures in a modeling course that I have cotaught in the Division of Bio-Medical Sciences at Brown during the past several years. An earlier version of these notes appeared in the Center for Dynamical Systems lectures notes series (CDS LN 73-1, November 1973). I have in this revised and extended version of those earlier notes incorporated a number of changes based both on classroom experience and on my research efforts with several colleagues during the intervening period. The narrow viewpoint of the present notes (use of optimization and control theory in biomedical problems) reflects more the scope of the CMC lectures given in August, 1973 than the scope of my own interests. Indeed, my real interests have included the modeling process itself as well as the contributions made by investiga tors who employ the techniques and ideas of control theory, systems analysis, dif ferential equations, and stochastic processes. Some of these contributions have quite naturally involved application of optimal control theory. But in my opinion many of the interesting efforts being made in modeling in the biomedical sciences encompass much more than the use of control theory.

The articles of these proceedings arise from a NSF-CBMS regional conference on the mathematical modeling of the hearing process, that was held at Rensselaer Polytechnic Institute in the summer of 1980. To put the a=ticles in perspective, it is best to briefly review the history of suc~ modeling. It has proceeded, more or less, in three stages. The first was initiated by Herman Helmholtz in the 1880's, whose theories dominated the subject for years. However, because of his lack of accurate experimental data and his heuristic arguments it became apparent that his models needed revision. Accordingly, based on the experimental observations of von Bekesy, the "long wave" theories were developed in the 1950's by investigators such as Zwislocki, Peterson, and Bogert. However, as the ex?eri~ents became more refined (such as Rhode's ~wssbauer Measurements) even these models came into question. This has brought on a flurry of 'activity in recent years into how to extend the models to account for these more recent eXT. lerimental observations. One approach is through a device co~monly refered to as a second filter (see Allen's article) and another is through a more elaborate hydroelastic model (see Chadwick's article). In conjunction with this latter approach, there has been some recent work on developing a low frequency model of the cochlea (see Holmes' article).

This book illustrates some of the ways physics and mathematics have been, and are being, used to elucidate the underlying mechan isms of passive ion movement through biological membranes in general, and the membranes of excltable cells in particular. I have made no effort to be comprehensive in my introduction of biological material and the reader interested in a brief account of single cell electro physlology from a physically-oriented biologists viewpoint will find the chapters by Woodbury (1965) an excellent introduction. Part I is introductory in nature, exploring the basic electrical properties of inexcitable and excitable cell plasma membranes. Cable theory is utilized to illustrate the function of the non-decrementing action potential as a signaling mechanism for the long range trans mission of information in the nervous system, and to gain some in sight into the gross behaviour of neurons. The detailed analysis of Hodgkin and Huxley on the squid giant axon membrane ionic conductance properties is reviewed briefly, and some facets of membrane behaviour that have been revealed since the appearance of their work are dis cussed. Part II examines the foundations of electrodiffusion theory, and the use of that theory in trying to develop quantitative expla nationsof the observed membrane properties of excitable cells, in particular the squid giant axon. In addition, an ad hoc formulation of electrodiffusion theory including active transport is presented to illustrate the qualitative nature of cellular homeostasis with respect to intracellular ionic concentrations and membrane potential, and cellular responses to prolonged stimUlation.
