目的 :研究中药制剂过程中不同因素对人参皂苷Ro（简称Ro）胶束化的影响,从而为增强中药难溶性药物的溶解度提供指导。方法 :采用介观动力学方法（MesoDyn）,考察了Ro的临界胶束浓度（CMC）及聚集形态随温度、难溶性药物柴胡皂苷a（简称Sa）浓度及体系盐强度的变化。通过对有序参数及珠子密度分布的分析,探讨介观尺度下不同因素对聚集体相行为的影响机制。结果 :Ro与水的二元混合体系中,当Ro浓度低于1.29 vol%时,体系呈均相态,溶液中尚无胶束形成,有序参数值很小;而当浓度达到1.29 vol%时,体系开始发生相分离而出现球状聚集体,有序参数发生明显变化,故Ro在水溶液中的CMC约为1.29 vol%。在298K～373K温度范围内,Ro的CMC随温度升高呈近似线性增大;固定Ro浓度为2.0 vol%,随温度升高,体系发生相分离所用的时间延长,表明温度的升高不利于胶束的形成。通过对2.0 vol%Ro体系中不同珠子密度分布的分析发现,随着温度升高,中心亲水珠子的密度增大,亲水区域增大,表明温度升高有利于亲水分子向囊泡中心扩散,中心含水量也略有增加。在Ro、Sa、水三元混合体系中,随着Sa浓度的增加,Ro的CMC减小,且Sa浓度越大,CMC降低的程度越大;随着Sa浓度增加,Ro发生相分离所需的时间越短,且呈幂函数降低,说明Sa有利于促进Ro胶束的形成。考察不同盐强度下Ro的CMC,结果显示当随着盐强度的增加,Ro的CMC略有下降,但并不显著,当离子强度大于0.2mol·L时,CMC基本不变,考虑可能与Ro疏水链较短有关。结论 :介观动力学方法（Mesodyn）可用于表面活性剂相分离过程及其与化合物间的相互作用的研究。

近红外光谱技术不仅广泛应用于食品、农业和石油化工等领域,近年来也广泛应用于制药工业中,以控制各种原料药、中间体和成品的质量。为进一步推广并规范近红外光谱技术在制药领域的应用,欧洲医药管理局颁布了《欧盟制药工业近红外光诺技术应用、申报和变更资料要求指南(草案)》(以下简称《指南》),围绕近红外光谱分析方法的建立、校正、验证、维护和应用等,提出了一系列指导原则和要求。本文对此予以详细介绍,以期为我国近红外光谱技术在制药领域中的研究与应用提供参考。《指南》具体从适用范围、法律依据、近红外光谱分析方法建立与完善的一般要求、定性分析方法与定量分析方法中的具体要求及近红外光谱分析方法的变更与维护等方面进行详细的论述,还介绍了对近红外光谱法进行申报与变更时所需呈递的资料要求。近红外光谱法的建立与完善是一个动态过程,主要包括数据采集、模型校正、模型验证和模型维护与更新等方面。数据采集时主要从样本集的组成、样本数量、样品的制备装载方式及光谱采集的影响因素等方面进行考虑,以确保得到稳健的校正模型。对于定量分析,通常需要三个样本集(校正集、测试集、验证集)用以建立、优化和验证校正模型。其验证内容主要包括专属性、线性、准确度、精密度、稳健性和检出限与定量限。近红外定性分析方法主要用于鉴定、判别与合格检查三个方面,在方法建立时仅需校正集与独立验证集。其验证内容包括专属性与稳健性。对于影响近红外光谱分析方法性能的任何变更,都需对近红外模型进行重新校正和/或验证,以证明模型的适用性。

A simple and specific UPLC-Q-TOF-MS method with automated data analysis(MetaboLynx) has been developed and applied for the primary investigation of metabolism pathway of active Xiang-Fu-Si-Wu Decoction fraction（XFSW-8）.Through the online ESI-MSE analysis and automated data analysis(MetaboLynx), a total of fifteen constituents have been identified or tentatively characterized from the nine tested samples after oral administration of *** parent compounds and nine metabolites were identified in blood and eight *** / demethylates sulfation,glucuronidation,sulfation and glucuronidation conjugates were the major metabolic pathways of alkaloids in XFSW-8 in *** present study provided important information about the metabolism of alkaloids in Xiang-Fu-Si-Wu Decoction which will be helpful for fully understanding the mechanism of action of this Chinese medicine.

目的 :提出基于虚拟基因敲除的疾病分子网络关键节点辨识方法,以疾病相关分子网络为载体,通过考察网络中节点虚拟敲除前后对于其他节点的节点影响力值产生的影响,来判定被敲除节点的关键性,从而在宏观和微观两个层面上对关键节点进行研究。方法 :以构建的阿尔茨海默病疾病分子网络为研究载体,针对网络中疾病的已知治疗靶点进行虚拟敲除,通过考察网络中靶点敲除后对其他节点的节点影响力值产生的影响(影响的范围及影响的幅度)来验证被敲除靶点的关键性,并在后续研究中针对其他的非靶点节点进行虚拟敲除,通过与靶点敲除后产生影响的比较,从而发现可能的关键节点作为药物的新治疗靶点。结果 :对阿尔茨海默病的主要治疗靶点如GSK3-beta(3β-糖原合成酶)、APP(Aβ淀粉样A4蛋白)、CDK5(细胞周期蛋白依赖性激酶5)等进行了虚拟敲除,结果显示这些靶点的敲除对网络中其他节点的节点影响力值产生很大影响,其他节点的节点影响力值均有显著下降。随机选择处于网络中心的非靶点进行敲除,如beta-catenin(β连环蛋白)、Ras蛋白的敲除,分别对24个节点和29个节点的节点影响力值产生了影响,且节点影响力值下降幅度均极小。虚拟敲除过程中阿尔茨海默病已知靶点的虚拟敲除,使其他节点的节点影响力值均有显著下降,而随机节点的敲除无此现象。结论 :靶点与非靶点的敲除对其他节点的节点影响力值是不同的,且靶点敲除后的影响面和影响程度比非靶点的敲除影响更明显,证实了靶点在分子网络中的重要性,有利于发现新的靶点。本文所建立的方法能够验证已知靶点的重要性并筛选出新的关键节点,方法具有可行性和可靠性。

As one of the most commonly used medicinal plants,ginseng has been an attractive model herb for study.A wide range of analytical methods has been used to characterize its ***,less effort has been devoted to the rare ginseng saponins,especially their isomers and sugar *** this study,we used segmental monitoring and diagnostic ion filtering to characterize ginseng saponins by rapid liquid chromatography with time-of-flight mass spectrometry(LC-TOF-MS).By using selected diagnostic ions,specific groups of ginseng saponins were readily extracted from the complicated matrix.20(R) and 20(S) stereo-saponins were differentiated using the peak abundance ratio of[M-H20+H]+ to[M-2H20+H]+.The fragmentation behavior of ginsenosides was first reported in negative ion mode by MS/MS with high-energy collision-induced dissociation, producing rules to determine sugar numbers,positions and *** the rules,we identified and compared the nontarget ginseng saponin profiling of raw and steamed American ginseng roots and *** characterized 70 saponins in the *** strategy can be extended to screen and characterize other rare ginseng saponins and their metabolites

Shaofu Zhuyu decoction,a famous effective traditional Chinese formula,is commonly utilized for treating blood stasis syndrome of gynecology diseases in clinic for a long *** this paper,we compared the pharmacokinetics profile of three active aromatic acids in normal and dysmenorrhea rats plasma following oral administration of SFZYD and its ethanol *** rats were randomly divided to six groups and given the same dose of 2.1 mg/kg SFZYD and *** samples were collected at different time (0,0.083,0.25,0.5,1,2,4,6,8,12,24 h) after administration,the concentration of three aromatic acid components were then determined by ultra-performance liquid chromatography/time-of-flight mass spectrometry (UHPLC-QTOF/MS) and main pharmacokinetic parameters were estimated by DAS 2.0 *** aromatic acids were absorbed better in SFZYD than SFE,which was prepared of same amount of crude *** group has a better absorption than normal group which was benefit for therapy efficacy.

A strategy based on chemical markers' fishing and knockout has been proposed for holistic activity and interaction evaluation of the bioactive components in herbal medicines(HMs).It was devised to screen bioactive-compound group that represents the efficacy of HM,estimate the bioactivity contribution of each component and elucidate the interactions of *** strategy was accomplished through the following steps:(1) screen out the chemical markers(target peaks) in a HM fingerprint using online two-dimensional turbulent flow chromatography/liquid chromatography-mass spectrometry technique,(2) fish target peaks and knockout any interested peak,and(3) evaluate the bioactivities of fishing and knockout portions. After comparison of the bioactivities of samples containing different target peaks,the efficacy of target-peak group, bioactivity contribution of each compound,and the interactions of multi-components are *** Acetylcholinesterase(AChE) and Bulbs of Lycoris radiata(***.) Herb.(BLR) as the experimental materials, four target peaks were screened out as the AChE *** target peaks' fishing and knockout,combined with activity evaluation,we observed that the bioactivity of the four-peak mixture is similar with the global bioactivity of BLR extract,and there are significant suppressive actions among these four target *** results indicate that this proposed strategy is a useful approach for holistic screening of bioactive-compound group and elucidation of the multi-component interactions in HM.
