目的:探讨核苷类药物替比夫定对肝癌细胞株HepG2肿瘤干细胞标志分子CD133和甲胎蛋白(AFP)的影响。方法:本研究以HepG2细胞为实验细胞,实验分为两组:实验组(与替比夫定共育组)和对照组(单纯细胞组)。实验组加入含替比夫定(10umol/L)培养... 详细信息

目的:探讨核苷类药物替比夫定对肝癌细胞株HepG2肿瘤干细胞标志分子CD133和甲胎蛋白(AFP)的影响。方法:本研究以HepG2细胞为实验细胞,实验分为两组:实验组(与替比夫定共育组)和对照组(单纯细胞组)。实验组加入含替比夫定(10umol/L)培养液与细胞共育,对照组只加不含替比夫定细胞培养液培养。隔天更换细胞培养液。培养细胞至80-90%铺满培养瓶时,收集细胞。1)4×10～5细胞/ml被用于流式细胞仪双染法检测HepG2细胞表面CD133的表达:2)1×10～7细胞/ml被用于化学发光免疫检测细胞内AFP含量;3)1×10～7细胞/ml被用于提取RNA,等量的RNA分别被用于实时荧光定量聚合酶链反应(Real-Time PCR)检测细胞内CD133和AFP的表达。每组细胞三个培养瓶,重复3次,将各次实验数值取均数,用于统计学处理。结果:1)流式细胞仪检测:替比夫定组和对照组的CD133阳性细胞的表达率分别为0.83%±0.02%和1.9%±0.04%;2)化学发光免疫分析法测得两组细胞内AFP的数值分别为:149.07±3.57(μg/L)和180.32±3.92(μg/L);3)定量PCR检测,替比夫定组和对照组HepG2细胞CD133 mRNA和AFP mRNA的表达分别为:0.609±0.057和0.956±0.132,1.061±0.034和2.507±1.379。统计学t检验处理各实验数据,CD133阳性细胞率、AFP值、及CD133 mRNA和AFP mRNA的表达,替比夫定组和对照组间均存在显著性差异(p<0.05),替比夫定与HepG2细胞共育后CD133和AFP的表达均减少。结论:替比夫定抑制HepG2细胞的肿瘤干细胞标志分子CD133的表达,降低AFP合成,这可能是核苷类药物治疗肝炎延缓肝癌发生的分子生物学机制之一。

Background Acute fatty liver of pregnancy (AFLP) can lead to fatal liver damage that occur in the third trimester or early postpartum *** effect of traditional therapy is not very ideal for facilitating hepatic *** safety and effect ofnon-bioartificial liver (NBAL) in application for treating AFLP are lacking systematic *** Analysis of the timing and efficacy of non-bioartificial liver (NBAL) in the treatment of acute fatty liver of pregnancy (AFLP).Methods We observed the clinical data of 5 cases of AFLP treated with NBAL from 2007 to 2012 including symptoms, physical signs, adverse effects, and compared all relevant laboratory test results before and after *** (1) Side effects during NBAL were slight and well-tolerated, and no patients interrupted treatment because of negligible side effects;Clinical symptoms, signs, and liver and renal functions can be ameliorated significantly after NBAL;(2) In the 5 cases treated, 4 were cured, 1 succeeded in bridging to transplantation;(3) In the 4 cases cured, the sooner a AFLP patient received NBAL the better the curative *** Treatment of AFLP by NBAL is safe and effective, and early application of NBAL in AFLP patients can effectively treat and reverse progress of the disease.

Objective: To explore the relationship between nonalcoholic fatty liver and insulin ***: 102 patients were divided into two groups: NAFLD group(n=52) and without NAFLD group(n=50), BMI, WHR, TC, TG, CRP, HDL-C, LDL-C, ALT, Cr, FPG, FINS and HOMA-IR were compared between the two groups, and logistic regression was ***: In NAFLD group, the level ofBMI(26.7±2.3 vs 22.4±2.5, P<0.01), WHR(0.94± 0.06 vs 0.83 ± 0.05, P<0.01), TG(2.4 + 0.6 vs 1.8 ± 0.6, P<0.01), ALT(37.3 ± 8.3 vs 28.1±7.2, P<0.05), FPG(6.2± 1.4 vs 5.2±0.7, P<0.01), FINS(23.6± 13.6 vs 8.6±3.5, P<0.01)、 HOMA-IR(6.7 ± 4.7 vs 2.0± 1.6, P<0.01) were higher than the control *** the logistic regression BMI(P<0.01), WHR(P< 0.01), TG(P<0.01), ALT(P <0.05) and HOMA-IR (P<0.01) were independent *** l us ions: BMI, WHR, TG, ALT and HOMA-IR were independent factors of NAFLD.

Backgrounds: Glypican-3(GPC-3) has been reported as one of the most promising serum markers for hepatocellular carcinoma (HCC), while several studies have conflicting results for the diagnostic accuracy between GPC-3 and alpha-fetoprotein (AFP).Methods : Studies that explored the diagnostic value of GPC-3 and AFP in HCC were searched in MEDLINE, EMBASE, PUBMED, the Cochrane Library and Chinese BioMedical Literature Database (CBM).Sensitivity, specificity and other measures about the accuracy of serum GPC-3 and AFP in the diagnosis of HCC were pooled using random-effects *** receiver operating characteristic curve (sROC) analysis was used to summarize the overall test *** l ts : Ten studies were included in our *** pooled sensitivity for AFP and GPC-3 are: AFP 51.9% (95%confidence interval (CI): 0.47-0.56), GPC-3 59.2% (95%CI:0.55-0.63);while the pooled specificity for AFP and GPC-3 are: AFP 94% (95%CI:92.1% to 95.6%), GPC-3 84.8% (95%CI:82% to 87.3%).The pooled diagnostic odds ratio (DOR): AFP 23.4 (95%CI: 10.3-53.2), GPC-3 17.99 (95%CI:5.4-60.4).Area under sROC for AFP and GPC-3 are the same as 0.81 (95%CI: 0.77-0.84).Conclusions: GPC-3 is comparable to AFP as a serum marker for the diagnosis of HCC, combination of AFP and GPC-3 can elevate the sensitivity of diagnosis.

Alcoholic liver disease (ALD) is associated with a spectrum of liver injury ranging from steatosis and steatohepatitis to fibrosis and *** response to gut-derived lipopolysaccharide (LPS), activation of Kupffer cells plays a key role in the development and progression of ALD by secreting a variety of pro-inflammatory ***, inhibition of macrophage-activation would have therapeutic benefits for alleviating the progression of *** (Sal), one of main bioactive components isolated from Rhodiola Sachalinensis, has been reported to suppress LPS-induced inflammatory response, but the underlying mechanisms in macrophages remain poorly *** this study, we investigate the anti-inflammatory effects of Salidroside and the possible mechanisms in LPS-stimulated phrobol 12-myristate 13-acetate (PMA)-differentiated THP-1 macrophage *** results showed that Sal markedly decreased the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2(COX2), interleukin-lbeta (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) at both mRNA and protein levels, and there was dose-effect relationship between the three Sal pretreated *** studies revealed that Sal strongly suppressed NF-κB activation and down-regulated the phosphorylation of ERK,p38 and *** present study demonstrated that Sal could suppress the production of iNOS, COX2, IL-1β, IL-6 and TNF-α in LPS-stimulated PMA-differetiated THP-1 cells by inhibiting NF-rB activation and MAPK signal pathway phosphorylation.

Background and aims Hepatitis B related acute-on-chronic liver failure (HBV-ACLF) has a poor prognosis with very high *** in nucleoside analogues treated patients with HBV-ACLF is *** study aimed to evaluate the efficacy and safety of Entecavir, Lamivudine and Telbivudine in treating patients with HBV-ACLF and to validate TPPM model in these *** In this retrospective study, we enrolled 283 patients with HBV-ACLF (100 treated with Entecavir, 98 treated with Lamivudine and 85 treated with Telbivudine).There were no significant differences in baseline clinical and virological characteristics between patients treated with Entecavir, Telbivudine or *** There were no significant differences in the 4 and 12-week survival rates of Entecavir, Telbivudine and Lamivudine-treated patients (79.00%, 81.18%, and 86.73%, respectively at 4 weeks and 67.00%, 65.88%, and 73.47%, respectively at 12 weeks).Patients in all three groups achieved an improvement of MELD *** the Hosmor and Lemeshow test, the validation of TPPM for HBV-ACLF demonstrated a good degree of fit with disease *** on this unique group of patients, the TPPM with an AUC of 0.787 was superior to MELD which had an AUC of 0.736 in the prediction of 12-weeks *** had an AUC of 0.733 and MELD had an AUC of 0.672 in the prediction of 4-weeks *** a cutoff of 0.22 for 12-weeks mortality prediction by TPPM, the positive predictive value was 49.66%, with a negative predictive value of 89.55%.Conclusion Nucleotide analogs including Entecavir, Lamivudine and Telbivudine treatment prevented disease progression and increased the survival of patients with *** of the established TPPM scoring system in this study confirmed its superior predictive value for HBV-ACLF patients when compared with MELD.

Increased KCNJ15 expression has been found in patients with hepatitis B virus (HBV)-induced Acute-on-chronic liver failure (HBV-ACLF) in our previous gene microarray ***, the mechanism underlying the enhanced expression of KCNJ15 in patients with HBV-ACLF remains *** this study, we further investigate the up-regulated KCNJ15 and its possible regulatory mechanisms in HBV induced liver *** expression of KCNJ15 was studied in patients with HBV-ACLF on both mRNA and protein levels in peripheral blood mononuclear cells (PBMC) and liver tissue, while the function of KCNJ15 was further determined in Jurkat cells in vitro by transfection of a human KCNJ15 expression *** results showed that KCNJ15 was over-expressed on PBMCs and hepatic infiltrating lymphocytes from HBV-ACLF ***, KCNJ15 had an abundant expression in peripheral and hepatic CD4+T cells in patients with HBV-ACLE In the supematant of KCNJ15 over-expressed Jurkat cells, increased IL-17A secretion was observed, which was involved with elevated intracellular calcium *** data suggested KCNJ15 may contribute to liver injury through IL-17A production in HBV induced liver ***, we also provide a potential susceptible gene for patients with HBV-ACLF.
