Haemonchus contortus,the blood-feeding gastrointestinal parasite,is a major constraint on ruminant health and production *** occurrences of antiparasitic-resistant strains exacerbate the control of ***,the development of vaccines against *** is an alternative *** order to gain further understanding of host immune responses to ***,we examined the gene expression profiles of sheep PBMCs stimulated with *** extracts(Hc Ag).Differential expression analysis revealed that the expression of a substantial number of genes changed significantly after *** analysis of the genes concerning germline-encoded receptors revealed increased transcription of CLEC2 L and NLRP3 after Hc Ag *** for the genes involved in signaling cascades,we found that ATF3,a negative regulatory transcription factor in TLR and CLR pathways,increased significantly,which may account for the mild expression profile of inflammatory ***,we examined which component of *** extracts may play a key role in regulating the expression of immune-related genes of sheep *** found that stimulation with proteinase K-treated Hc Ag could also increase the expression of CLEC2 L,NLRP3,ATF3 and most effector molecules of sheep PBMCs,but not by the stimulation with periodate-treated Hc *** real-time PCR also confirmed this expression ***,we proposed that carbohydrate-containing molecules in *** extracts could stimulate sheep PBMCs to upregulate CLEC2 L and NLRP3,then signaling through ATF3 to regulate the expression of cytokines and *** research may give a hint to identify vaccine candidates against *** infection.

Experimental Cerebral Malaria(ECM)-induced neuroinflammatory response is associated with activation of brain resident microglia and accumulation of circulating macrophage infiltration,which play a crucial role in nerve damage and neurological ***,most researches haven't distinguished these two kinds of diffirent genetic macrophages,it is poorly understood whether and how resident microglia activation contributes to neuroinflammation in intraparenchymal area of cerebral *** order to find the evidence which microglia involvement in ECM,we detected the microglia inflammatory marker in different regions of brain of ECM mouse *** order to remove the influence of peripheral macrophages on brain parenchyma,we took the brain tissue following perfusion after P.b ANKA infection,while the blood-brain barrier was still *** to the m RNA levels examined by q PCR,we found that M1 polarization markers including Ccr2,Cx3 cr1,Cx3 cl1 and Itgam increased in different degrees,especially Ccr2 was markedly elevated in olfactory,cerebellum,brain stem and ***-inflammatory factor TNF-a and IL-6 raised in most area of brain,meanwhile anti-inflammatory molecules Fizz1 significantly reduced in all *** addition,there was statistically significant increase in neuron specific NOS1 and glial cell marker GFAP in *** data shown that before the blood-brain barrier injuried and the peripheral macrophages entered the parenchyma,microglia was the main part of macrophage causing inflammatory action in the parenchymal *** it is meaningfull to understand the role of microglia in the nervous pathological damage of cerebral malaria,in order to reduce the death and the sequelae of nervous system.

Eimeria intestinalis is one of the most pathogenic species of rabbit coccidia and the infection rate of *** is very high in *** two species should be taken into account to develop live oocyst vaccines against rabbit *** this study,we immunized rex rabbits with precocious lines of ***(Ei P) and ***(Em P).The dose for group 1 is 500 Ei P and 500 Em P;while the dose for group 2 is 500 Ei P and 1000 Em *** rabbits were challenged with 1×10～4 *** wild-type oocysts and 1×104 *** wild-type oocysts on day 14 post *** in group 1 and group 2 showed no clinical syndrome or loss of body weight *** challenge,the body weight gain of rabbits in both group 1 and 2 was similar with that of the control,which is about(22.5+4.9) g per *** it was higher than that of the unimmunized and challenged rabbits(P<0.01),which is about only –(21.4+3.6) g per ***,oocyst output of the two species in group 1 and 2 was both less than 8.0×10～6,which was significantly less than that of the unimmunized and challenged rabbits(2.5×10～8)(P<0.01).These results demonstrated that immunization with *** and *** could provide sufficient protection for rex rabbits against homologous challenge and there was no significant difference between the two doses of Em P *** the combination of 500 Ei P and 500 Em P oocysts was more *** study indicates that development of attenuated live vaccines for rabbit coccidiosis is hopeful.

目的:等足类(Isopoda)是甲壳动物中一个形态多样分布广泛的类群,目前已报道的等足类物种数超过10300种。寄生是等足类重要的分类依据,几乎所有的寄生类群都隶属于缩头水虱亚目(Cymothoida)。目前利用分子生物学手段来分析等足类系统发育进化关系的研究还比较少,报道的完整的等足类线粒体基因只有3种,尚无寄生等足类物种的线粒体基因的报道。日本鱼怪(Ichthyoxenus japonensis)和中华急游水虱(Tachaea chinensis)为寄生等足类,都隶属于缩头水虱亚目,日本鱼怪寄生于鱼类的体腔,中华急游水虱寄生于沼虾的体表,均影响宿主的生长和繁殖。本研究旨在克隆中华急游水虱和日本鱼怪线粒体基因组全长,分析其基因组成、基因重排和r RNA二级结构,在基因组水平上分析这2种寄生等足类的系统进化关系。方法:通过PCR扩增,测序获得日本鱼怪和中华急游水虱线粒体基因组全序列。参考相近物种序列进行基因注释。系统进化分析:利用Prot Test3.4和modelgenerator软件选取最适替代模型。运用软件Mr Bayes 3.2.6和raxml GUI1.5b1构建系统发育树。对等足类线粒体基因排列进行比较,分析其基因重排和系统进化的关系。利用mfold预测r RNA的二级结构和控制区的二级结构元件。结果:日本鱼怪和中华急游水水虱的线粒体全长分别为15440 bp和14616 bp,均包含13个蛋白编码基因,2个r RNA基因、1个主非编码区和21个t RNA。日本鱼怪线粒体基因缺失trn I,中华急游水虱线粒体缺失trn S1。等足类线粒体基因存在大量重排现象,中华急游水虱和日本鱼怪线粒体基因的排列不同,且二者与已有等足类物种线粒体的基因排列均不同,基因重排主要发生控制区附近的基因。trn A位于nad3和trn V之间是缩头水虱亚目的三个已有线粒体所共有的排列,在其他的等足类中均未被发现。比较三种缩头水虱亚目物种线粒体的12S和16S r RNA的二级结构,发现其保守序列不均等分布。在12S r RNA三个结构域中结构域III最为保守(42.2%),16S r RNA的五个结构域中结构域V最为保守(52.4%)。系统发育分析显示,氨基酸序列和核苷酸序列所构建的系统发育树中等足目的物种均以较高的支持率聚为一支,中华急游水虱与日本鱼怪亲缘关系较近在基部聚为一支,与其他的等足类物种形成姐妹群,但是两者并未与自由生活的缩头水虱亚目浪漂水虱科的Eurydice pulchra聚为一支,这与经典的形态分类不一致,但基于线粒体基因组的系统分析支持寄生是等足类重要的分类依据。

The eggs produced by the mature female worms cause the main pathological damages of host and the spread of the *** single-sex infected female worms can't develop thoroughly to sex mature to produce normal *** this study,iTRAQ-Based proteomics was used to investigate the difference of proteins between single-sex infected and bisexual infected female *** proteomic analyses revealed that 1540 of 3468 detected proteins were significantly differentially expressed between paired and unpaired female worms(p-Value<0.05,fold>1.2).There are 719 proteins(46.7%) upregulated expressed while the remaining 821 proteins(53.3%) downregulated expressed respectively in paired female worms compared with unpaired female *** analysis showed that on the biological process paired female worms express more proteins related to reproductive process,reproduction and immune system process,whereas more proteins related to locomotion,biological adhesion and signaling were expressed in unpaired *** the molecular function,paired females express more proteins related to structural molecule,enzyme regulator activity,antioxidant activity,electron carrier ***,unpaired females express more proteins related to transporter activity and oxidative *** study identify the DEPs between single-sex and bisexual female worms,further experiment on DEPs will provides valuable theoretical basis to reveal the molecular biology mechanism of sexual reproductive development and supports vaccine candidates or new diagnostic reagents for schistosomiasis.

Neospora caninum is an intracellular protozoan parasite which can cause abortion and stillbirth in *** morphology of this parasite is similar to Toxoplasma ***,it is yet to know whether Tibetan sheep can be infected with ***.A total of 2187 serum samples were collected from Tibetan sheep in the major production areas of Luqu,Maqu,Tianzhu in Gansu province,and Nyingchi in southeast Tibet,*** samples were analyzed for the presence of antibodies to *** using an enzyme-linked immunoassay(ELISA).ELISA-positive samples were confirmed by an indirect fluorescent antibody test(IFAT).The *** seroprevalence ranged from 4.40%(95% CI 1.42–7.37) to 9.36%(95% CI 7.52–11.20) among different regions,and seroprevalence in male animals(10.82%)(95% CI 8.41–13.23) was significantly higher than in female animals(7.43%)(OR =1.51,95% CI 6.12–8.73)(P < 0.01).This is the first report of *** seroprevalence in Tibetan sheep in China,which provides baseline data for the prevention and control of *** infection in Tibetan sheep in China.

Trichinella spp.,one of the most widespread parasitic nematodes,live in muscle tissue of a wide variety of vertebrate and cause humans *** infection mainly occurs by ingestion of undercooked,infected *** simple and accurate diagnosis of Trichinella *** in pigs is crucial to prevention and control of human *** the present study,a simple,portable,rapid and accurate diagnostic method was developed using the combination of recombinase polymerase amplification and a lateral flow strip(LF-RPA) to detect Trichinella spiralis and Trichinella pseudospiralis *** LF-RPA assay targeting the Trichinella *** small-subunit ribosomal RNA(rrn S) gene could detect 100 fg DNA of examined Trichinella strains,which was appropriately 10 times more sensitive than the conventional PCR *** LF-RPA assay could be finished within 10-25 min at temperatures wide ranging from 25 °C to 45 °C and showed no cross-reaction with other parasites and wild *** addition,the LF-RPA assay was worked better with high concentration of PCR inhibitor than conventional PCR *** diagnostic validity of LF-RPA assay was evaluated by detection of experimentally infected pigs and also compared the results of that from a conventional PCR showing excellently diagnostic agreement with the conventional PCR with 100% specificity and high *** together,the developed LF-RPA assay is a simple,specific,sensitive,rapid and convenient method for the diagnosis of Trichinella infection in pigs.
