The protozoan parasite Toxoplasma gondii is capable of infecting all warm blooded animals and *** are three infectious stages of *** in its intermediate and definitive hosts: tachyzoites,tissue cysts or bradyzoites,and *** tachyzoites in liquid nitrogen is used to maintain *** strains in *** this study,we tried freezing tissue ***/c mice infected with 200 tachyzoites of Toxoplasma gondii Pru strain were subjected to tissue cysts isolation from the brain between 4 and 8 weeks post *** cysts were purified using gradient density centrifugation with human lymphocytes separation *** the tissue cysts were treated with Trypsin-EDTA to disrupt the cyst wall and frozen in liquid nitrogen with a medium containing 90% fetal bovine serum and 10% *** viability of the freezed tissue cysts or bradyzoites was confirmed by observing the invasion of bradyzoites released from the cysts in Vero *** successful freezing of tissue cysts or bradyzoites provides more choice for manipulating *** in laboratory.

组织滴虫病是由火鸡组织滴虫引起的一种禽类原虫病。长期以来,该病的防治主要依靠药物。近年来,因公共卫生要求,药物已受到限制或禁止,使得该病防治陷入困境,流行严重,迫切需要研制安全的新药或疫苗。如何评价药物或疫苗的控制效果,建立动物感染模型并明确其致病机理是关键。江苏地区黄羽肉鸡饲养范围广,数量多且为地面平养,组织滴虫病发病率较高,本研究选用该品种通过人工感染,对组织滴虫病致病机理进行了探究。将从田间分离的强毒虫株JSYZ-A株虫体通过泄殖腔感染15日龄苏禽黄羽肉鸡,在感染后7-31d内,每3d扑杀5只感染鸡,采集不同组织脏器的样品和非抗凝血,组织样品进行虫体18S r RNA DNA PCR检测和组织病理学检查,血液样品进行生化指标检测,同时进行称重。虫体动态分布结果表明,在感染后7d起,黄羽肉鸡的盲肠和肝脏中可检测到虫体,且两个脏器中的虫体维持时间最长;心脏、十二指肠、空肠、脾脏、肺脏、法氏囊、腺胃、直肠中,偶尔能够检测到虫体;胰脏、脑组织中未检测到虫体。组织肝脏和盲肠病变程度在10-16d时最高,19d后病变程度减轻。PCR检测的结果与组织病理学结果基本一致。火鸡组织滴虫主要靶器官为肝脏和盲肠,但其并非单一感染盲肠和肝脏,而是会侵染机体的各个组织。同时,火鸡组织滴虫的感染会导致感染鸡体重明显减轻,机体的一系列生化指标出现紊乱,如谷丙转氨酶、谷草转氨酶、谷氨酰胺转移酶、球蛋白、尿素浓度的上升,葡萄糖、甘油三酯浓度的下降。本研究揭示了病原在体内的动态分布和血液生化指标变化规律,为后续更深入开展组织滴虫病的致病机理研究、药物的研发和疫苗的研制提供了有价值的参考。
