Berberine is an isoquinoline alkaloid with a long history of use in Chinese medicine. It has been isolated from a variety of plants, such as Berberis aquifolium and Berberis aristata, and has a wide spectrum of clinical applications such as in bacterial diarrhea, intestinal parasitic infection, cardiovascular diseases, inflammation and hypercholesterolemia. Accumulating evidence has shown the possibility of berberine being used as a drug for diabetes. However, the molecular basis for this action is yet to be elucidated. We report here the molecular mechanism of berberine and some progresses made in the drug discovery projects based on berberine and its analogues as novel mitochondrial function modulators.

A novel copper-catalyzed trifluoromethylation-initiated 1,2-aryl migration in α,α-diaryl allylic alcohols has been disclosed, which allowed for the synthesis of a variety of α-aryl β-trifluoromethyl ketones that are difficult to prepare by other means. Notably, the preferential migratory aptitude of electron-poor aryl group over electron-rich one on unsymmetrical substrates supports the radical mechanism, which was further confirmed by DFT calculations.

A doubling piece Schiff base is reported for the colorimetric and fluorometric dual-channel sensing of Hg2+ ions by taking advantage of the direct cleavage of Schiff base with Hg2+. This work provides a novel approach for the selective recognition of mercury ions. Notably, the color changes are very significant and all the recognition and recycling processes can be observed by the naked eye.

Highly active esterification catalysts that show selectivity towards carboxylic acids and alcohols are needed for the laboratory synthesis of complicated molecules and industrial production of pharmaceuticals and their intermediates with diverse functionalities[1,2]. N-Butyl-2,4-dinitro-anilinium p-toluenesulfonate(1) was found to be a very active esterification catalyst that promotes condensation of equal mole amount of carboxylic acids and alcohols under mild conditions. This catalyst is also highly selective towards carboxylic acid and alcohol substrates under ambient temperature.
