水稻瘤矮病毒(rice gall dwarf virus,RGDV)属于呼肠孤病毒科(Reoviridae)植物呼肠孤病毒属(Phytoreovirus)。RGDV的主要传播介体是电光叶蝉(Recilia dorsalis),介体叶蝉一旦带毒,可终身带毒并经卵传毒。目前RGDV在其主要介体电光叶蝉体内的侵染和扩散过程还不清楚,本研究通过免疫荧光标记和电镜技术观察了RGDV侵染电光叶蝉介体不同时间后,病毒在其消化道系统的定位和侵染循回过程。研究发现电光叶蝉饲毒后3天,病毒在电光叶蝉的滤室处复制组装成子代病毒粒体,表明电光叶蝉的滤室上皮细胞是RGDV的初侵染点。饲毒后5天,子代病毒穿过基底膜扩散到滤室外层的肌肉组织,进而侵染中肠和后肠,同时病毒释放到血淋巴细胞。饲毒后15天,RGDV侵染电光叶蝉的唾液腺。由此明确RGDV在电光叶蝉介体的侵染和扩散过程为:病毒首先侵入滤室上皮细胞,经复制后穿过基底膜扩散到滤室外层的肌肉组织,沿着环肌和纵肌侵染整个中肠和后肠并释放到血淋巴细胞,最后侵染唾液腺。本研究首次直观地阐述了RGDV在电光叶蝉消化系统的侵染和扩散过程,为解析病毒和介体侵染屏障的互作机制奠定基础。
核盘菌引起的菌核病是分布最广、危害最严重的油料和蔬菜等作物病害之一,但其致病机制的研究还有待深入。根据生物信息学预测,我们从核盘菌中鉴定了500个可能的分泌蛋白,经过数据库逐一比对,发现其中,40%的蛋白含未知结构域;糖苷酶约占24%,可能参与细胞壁降解;其次为蛋白代谢相关蛋白约占9%;脂类代谢蛋白约占5%;Domain of unknown function占约6%;金属氧化酶和转录因子各约占1.2%。此外还有少部分蛋白可能参与核酸代谢、次生代谢等。在此基础上,本研究将候选分泌蛋白基因克隆到诱导型表达载体pINDEX3中,借助农杆菌介导的本氏烟草瞬时表达系统,筛选效应蛋白。经核盘菌接种筛选后发现,4个LysM蛋白SsLysM1-SsLysM4处理的本氏草对核盘菌较为敏感,相对感病;台盼蓝染色发现,经SsLyMs处理的烟草中核盘菌的菌丝较为舒展。目前许多真菌LysM家族基因编码的效应子蛋白在抑制植物的免疫反应中起着重要作用(如番茄叶霉Ecp6,稻瘟病菌Moslp1蛋白),但DAB染色发现,SsLysMs并未改变其处理叶片中ROS积累情况,说明核盘菌LysM蛋白很可能通过别的途径抑制烟草防御反应。此外,荧光定量检测发现,核盘菌SsLysM编码基因在侵染早期(6h)得到提高,而后期(24h)则呈现下降的趋势,说明SsLysM可能在发病早期起作用并先抑制宿主的防御反应。
