目的:利用煤焦沥青烟提取物(Coal Tar Pitch Extracts,CTPE)构建体外永生化支气管上皮细胞(Immortalized Human Bronchial Epithelial Cells,BEAS-2B)恶性转化模型,通过RNAi技术研究ENST00000501520可能的靶向基因,对其发挥生物学作用的机制进行进一步研究,从LncRNAs水平探讨细胞恶性转化的发生发展机制。方法:CNC(Codingnon-coding Gene Co-expression Network)分析预测ENST00000501520可能的靶向基因,Western-blot检测ENST00000501520干扰后相关靶基因的蛋白表达变化。结果:根据Pearson系数选择CLTB、TAP2、RIPK2、SOCS3和邻近编码基因SBK1进行进一步研究。ENST00000501520干扰后,选择mRNA表达水平下降百分率超过50%的SBK1、SOCS3进行蛋白水平检测,结果显示相比较于CTPE30组,siRNA组细胞中SBK1、SOCS3蛋白水平明显下降(均P<0.001)。结论:CPTE所致恶性转化BEAS-2B细胞中ENST00000501520的表达量明显上调,干扰ENST00000501520后,相关联的SBK1、SOCS3基因蛋白表达量明显减少,说明ENST00000501520与SBK1、SOCS3存在正向相关关系。
