Severely debilitating and life-threatening (SDLT) diseases and advanced cancer share many similarities including shortened life expectancy and/or decreased quality of life. Therefore,the nonclinical program required to support the development of SDLT therapeutics should follow the principles outlined in ICHS9–the current internationally accepted nonclinical guidance for the development of therapeutics to treat advanced cancers. In this presentation, we will highlight the advantages of applying the principles outlined in ICHS9 for the development of therapeutics to treat SDLT diseases such as Congestive Heart Failure.

健康风险评估的一个重要过程是危害识别,而危害识别的重要手段,即毒理学数据资料的收集与整合,是通过进行毒理学数据的系统综述来实现的。本摘要即以对苯丙(a)蒽(Benz(a)anthracene,BaA)的健康风险评估为目的,对BaA进行毒性危害识别并进行研究展望。苯丙(a)蒽(Benz(a)anthracene, BaA)是一种四环结构的多环芳烃(Polycyclic Aromatic Hydrocarbons, PAH)类物质,不完全燃烧的副产物,与其它的PAHs以混合物的形式存在。BaA可通过口腔、鼻腔及皮肤等多种途径暴露于人类,对人的主要危害是致癌性。国际癌症研究机构(International Agency for Research on Cancer,IARC)根据其对致癌物的分类标准评价BaA为2B类,对人可能致癌。目前尚无充足数据支持剂量外推斜率因子(Slope Factor)的计算来完成动物致癌系数与人体致癌系数之间的推导。BaA的毒理学数据较为丰富,但多集中在上世纪80年代。考虑到BaA多与其它PAHs以混合物的形式存在,联合毒性或者在毒性效应中的贡献率等可能是未来研究的一个重要方向。另外,BaA的毒性分子机制研究也比较缺乏,这对于致癌性风险评估时进行毒理学数据人群相关性分析,特别是动物致癌系数与人体致癌系数之间的换算以及斜率因子的计算等方面均比较受限。国内学者可在近年来雾霾频发的基础上,分析其中的BaA含量并对其(致癌)毒性作用及分子机制进行深入地研究与分析,为指导特定地区大气细颗粒物的健康风险评估提供可靠的实验数据。
