Effects of butachlor,bensuifuron-methyl and dimethoate on the growth,photosynthesis and photoinhibition of the edible cyanobacterium Ge-Xian-Mi were examined in order to gain insight into the relationship between its productivity reduction and the abusive use of pesticides in the *** inhibition of growth was found in the presence of 4-6 fold field concentration of butachlor and very high concentrations of bensulfuron-methyl and *** stimulation of photosynthesis was observed over a limited range of low concentrations of these three *** found that PSII and PSI were respectively the inhibitory sites of 150 μM butachlor and 150 μM ***,the inhibitory site of 2000 μM dimethoate seems to be situated at the terminal of whole chain or dark *** colonies exposed to 150 μM butachlor were more sensitive to high light than the control cells and those exposed to bensulfuron-methyl,dimethoate or low butachlor *** light induced rapid recovery of photoinhibited colonies was observed for the control,10 μM butachlor,bensulfuron-methyl and dimethoate ***, the maximal PSII photochemical efficiency of photoinhibited colonies treated with 150 μM butachlor was maintained at a relatively stable value in low *** findings suggested that the abusive utilization of butachlor might be an important factor limiting the productivity of Ge-Xian-Mi in the field.

Arthrospira(Spirulina)(Setchell&Gardner)is an important cyanobacterium not only in its nutritional potential but in its special biological *** unbiased fosmid library of Arthrospira maxima FACHB438 that contains 4300 clones was *** size distribution of insert fragments is from 15.5 to 48.9 kb and the average size is 37.6 *** recombination frequency is 100%.Therefore the library is 29.9 equivalents to the Arthrospira genome size of 5.4 Mb.A total of 719 sample clones were randomly chosen from the library and 602 available sequences, which consisted of 307,547 bases,covering 5.70% of the whole *** codon usage of *** was not strongly *** content at the first position of codons(46.9%)was higher than the second(39.8%)and the third(45.5%)positions. GC content of the genome was 43.6%.Of these sequences,287(47.7%)showed high similarities to known genes,63(10.5%)to hypothetical genes and the remaining 252 (41.8%)had no significant *** assigned genes were classified into 22 categories with respect to different biological ***,the high presence of 25 sequences(4.2%)encoding reverse transcriptase indicates the RT gene may have multiple copies in the *** genome and might play an important role in the evolutionary history and metabolic *** addition,the sequences encoding the ATP-binding cassette transport system and the two-component signal transduction system were the second and third most frequent genes,*** genomic features provide some clues as to the mechanisms by which this organism adapts to the high concentration of bicarbonate and to the high pH environment.

Trchalose is a non-reducing disaccharide of two glucose units that functions as a stress protection metabolite in the stabilization of biological structures under stress tolerance in various *** biosynthesis of trehalosc is catalyzed by a two-step process that involves trehalosc-6-phosphate synthasc(TPS)and trchalosc-6-phosphate phosphatase(TPP).In this study,the full-length cDNA sequence(3219 bp)of the trehalose-6-phosphate synthase gene of Porphyra yezoensis(PyTPS)was isolated by RACE(rapid amplification of eDNA ends)-PCR. PyTPS encodes the TPS protein of 908 amino acids with a calculated molecular mass of 101786 Daltons,consisting of a TPS domain at the N-terminus and a putative TPP domain at the *** putative TPS sequence was deposited in GenBank(NCBI)with the accession numbers AY729671(mRNA)and AAW27916(protein).The homology alignment for the TPS proteins from five different organisms indicates that the most closely related sequences to PyTPS were those from OsTPS and AtTPS,whereas the most distant sequence to PyTPS was the one from EcOtsAB.

The impacts of solar PAR and UV radiation(UVR)on the growth,photosynthetic performance and morphology of the cyanobacterium Anabaena ***7120 were investigated under three different solar radiation treatments:exposures to a)constant low PAR(photosynthetic active radiation,400-700 nm),b)natural levels of solar radiation with and c)without UV radiation(290 -400nm).The photochemical efficiency was reduced by about 40% and 90% at day one when the cells were exposed to solar PAR or PAR+UVR,respectively,and recovered at a faster pace under the treatment without UVR in the following *** UVR inhibited the growth up to 40%,reduced trichome length by up to 49% and depressed the differentiation of heterocyst. Negligible concentrations of UV-absorbing compounds were found even in the presence of UVR. During the first 2 d of exposure to natural levels of PAR,carotenoid concentrations increased but no prolonged increase was *** formation was enhanced under elevated PAR levels with stimulated quantum yield and growth after an initial *** concentrations of carotenoids and a two-fold increase in the carotenoid to chlorophyll a ratio provided protection from the high levels of solar *** radiation treatments with UVR the relatively greater decrease in chlorophyll a concentrations compared with the increase in carotenoids was responsible for the higher carotenoid:chlorophyll a *** formation was-disrupted in the presence of solar ***,the longer term impact of heterocyst disruption to the survival of Anabaena *** further study.

In this experiment we studied the affects of the temperature,light and salinity to the growth of the marine diatoms Melosira sulcata,as well as its basic mechanism of life *** results showed that the optimum growth temperature and light intensity for Melosira sulcata was 25℃ and *** the salinity range of 16‰ and 32‰,the algae could grow normally,and the opt salinity is 24‰, but when the salinity the below 16‰ or above 32‰,the growth of Melosira sulcata was *** investigating the growth rates of Melosira suleata under different temperature and illumination,we got the suitable conditions to cultivate and study the life cycle of this marine long-chained *** result showed that the condition (20℃ 45001ux,32‰)was the suitable condition for spores' germination among all the conditions in the *** first the vegetative cells began to shrink along the longitudinal direction and increase in width,when its diameter reached 34.51ma the spores formed consequently,often after having been cultured approximately for 3 *** are two styles of spores' germination:one is the"blooming"in which spores were released from parent cell almost at the same time,the other is budding in which the spores germinate into a new cell one by one and develop into a new chain *** spores and fertilization wasn't observed,the spores might be auxospores or asexual *** in the term of asexual reproduction,the whole life cycle of Melosira suleata was completed in seven days at normal conditions.

Sequence-related amplified polymorphism(SRAP)and target region amplification polymorphism(TRAP)axe two newly developed PCR-based molecular marker techniques aimed for the amplification of open reading frames(ORFs).SRAP markers were distributed arbitrarily across the genomes;while TRAP markers may be correlated to the targeted EST sequence,from which the fixed primer originated to some *** and TRAP analytic system were set up and successfully used in diversity detection and germplasms identification of Porphyra lines in this study *** dendrograms and the DNA fingerprints of sixteen Porphyra lines were constructed according to the result of SRAP and TRAP *** polymorphic markers were all above 98%.The dendrograms from two marker techniques were accordant with the conventional taxonomy *** Porphyra lines were divided into two major groups at the 0.68 and 0.71 similarity level respectively in SRAP and TRAP analysis respectively,one group contained only P haitanensis lines and the other group contained P yezoensis and P oligospermatangia *** SRAP-DNA and TRAP-DNA fingerprints were constructed using eleven and twelve amplified fragments from two *** the DNA fingerprints,each of the 16 Porphyra lines has its unique fingerprinting pattern and can be easily distinguished from each *** PGI-SRAP and PGI-TRAP(Porphyra germplasm identification developed by SRAP and TRAP method)developed by DNA fingerprints could be used to germplasm identification of these Porphyra lines.

The photosynthetic acclimation of Haematococcus pluvialis Flotow to elevated irradiance and its photoprotective mechanism were *** light caused a remarkable increment in carotenoids content per *** and volumetric chlorophyll contents were significantly increased after four days of high light *** photosynthesis of high light treated cells was decreased but their dark respiration was increased during the accumulation of secondary carotenoids(SC).The inactivation of reaction centers was observed in high light treated cells,and their normalized complementary area and turn-over number were higher than those under low *** PS II activity of red cells from high light was decreased by 17% compared with green cells from low light but their PSI activity was significantly *** K-step could not be observed in the fluorescence transients of red cells indicating that the oxygen-evolving complex was not affected during SC *** could protect itself against strong irradiance through the D1 protein repair cycle and the xanthophyll *** D1 protein repair cycle was the most important protective mechanism in *** and its operation could alleviate the photoinhibition by 49% in green cells'and 53～55% in red *** xanthophylls cycle could contribute to protect green cells subjected to strong irradiance but its roles could be neglected in red *** Fv/Fo values were decreased respectively by 45% in green cells and 32～34% in red cells after 2.5 hours of photoinhibitory ***,this could not necessarily indicate that the accumulated SC in red cells might play a photoprotective role.

We carded out experiments to evaluate the impacts of UV radiation（UVR, 280-400nm）on photosynthetic carbon fixation of phytoplankton assemblages from coastal waters of the South China Sea during March 2004 - Sep *** surface samples were taken and exposed to different radiation treatments using sharp cut-off filters,receiving full spectrum of solar radiation（PAB,280-700nm）,PA（320-700nm） that filtered out of UV-B（280-320nm）,and P（400-700nm）that cut off both UV-A and UV-B（320-400nm）.Solar UVR significantly inhibited photosynthetic carbon fixation rates of phytoplankton assemblages during spring and winter,with UV-A accounting for most of the observed inhibition;little UVR-induced inhibition was observed during *** view of the photosynthetic performance among different weather conditions,phytoplankton cells appeared to be able to utilize UVR under low levels of solar irradiances on the cloudy *** mean values of assimilation number（ASN）were 2.83（spring）7.62（summer）,and 4.35μg C（μg chl a）h （winter）under PAR,and were 1.57,5.70,2.71μg C（μg chl a）hunder PAR+UVR, *** a concentration ranged from 0.99-1.12 in the spring, 1.12-16.2 in the summer and 0.72-1.35 μg/L in the *** period showed the highest ASN and the lowest UVR-related *** the summer period, higher temperature might have led to faster enzymatic reactions for the repair of UVR-induced damages,and stratification（less vertical mixing）could result in higher abundance of the species tolerating high levels of PAR and UVR.

蓝藻中的念珠藻对极端生态环境具有显著适应能力,而且还有较高的经济价值,研究其抗逆及适应的分子机制不但有助于揭示生命适应极端环境的生存机制,而且为念珠藻的资源保护和开发利用提供理论依据和技术参数。本文从分子水平对生长在阴湿环境的地木耳(Nostoc commune)、生长在淡水中的葛仙米(***)和生长在干旱地区的发菜(***)三种念珠藻的超氧化物歧化酶基因(SOD)分别进行克隆并分别在大肠杆菌中进行重组表达以及对重组蛋白进行纯化和活性测定,发现这三种念珠藻的 SOD 基因的 DNA 序列和翻译蛋白的氨基酸序列的同源性分别高达97%和100%。纯化后的重组蛋白的都具有 SOD 酶活性,比活力均为2550U/mg,并且在胁迫条件下,三种重组蛋白的酶活性无显著差异。但是这三种念珠藻的蛋白粗提液中的 SOD 酶活性在胁迫环境下却表现出明显差异。在0.8mol.L的较高浓度 NaCl 胁迫下,发菜蛋白粗提液中 SOD 酶活性仍然很高,仅在胁迫处理36小时后活性下降了约15%左右:而在小于0.4 mol.L的较低浓度 NaCl 处理48小时后,地木耳和葛仙米的粗蛋白提取液中的 SOD 活性分别下降了50%和54%;在 NaCl 浓度大于0.4 mol.L胁迫48 小时后,二者 SOD 活性则分别下降了75%和77%。发菜在5.4-11.5范围的 pH 下培养48小时后,其粗蛋白提取液中 SOD 活性变化不大,且在较酸性环境下(pH5.4), 其 SOD 活性反而升高:地木耳和葛仙米在较酸性环境下其粗蛋白提取液中 SOD 活性下降幅度比在较碱性条件下(pH9.4和 pH10.4)的大,且在48小时后 SOD 几乎完全丧失活性。发菜在低温和高温胁迫下,其粗蛋白提取液中 SOD 活性下降幅度较小;而地木耳的粗大蛋白提取液中 SOD 活性明显下降,葛仙米的却出现上升趋势。本研究结果说明不同的念珠藻中,存在着不同的抗逆机理。发菜对胁迫环境有着很强的忍受性,其细胞内一定存在除 SOD 以外的稳定保护系统,在抵抗极端的胁迫环境中协助 SOD 共同发挥重要作用。
