近年来,越来越多的研究表明,骨条藻(Skeletonema Greville)是一类具有广泛地理分布的浮游硅藻,骨条藻的不同地理种群具有不同的遗传特征。本文比较研究了3种常用的遗传标记,即核糖体小亚基基因(SSU)、核糖体大亚基基因(LSU)和转录内间隔区(ITS)(包括ITS1、5.8S和ITS2)在区分不同地理株骨条藻中的作用。我们选用分离自威海沿岸6个不同地理水域的2种骨条藻—多恩骨条藻*** Sarno et Kooistra和拟中肋骨条藻*** Medlin *** et Kooistra,共25个单克隆藻株为研究对象,分别提取各株骨条藻的DNA,PCR扩增获得SSU、LSU和ITS序列,采用生物学软件计算各采样地点种内分子变异度,分别用3种序列进行系统发育分析,建立并比较进化分枝图。结果显示,同一采样地种内分子变异度较高,进一步确认骨条藻的遗传多样性;3种分子标记均能将不同地理种群的骨条藻加以明显区分,但3种分子标记的区分能力有一定差别,其中ITS的区分能力最强,LSU次之,SSU最差。
