研究、预防和治理赤潮,首先要对引发赤潮的生物种类进行准确鉴定与识别。骨条藻是我国近海的典型赤潮藻类。最近,骨条藻Skeletonema Greville的分类学发生了重大改变,骨条藻成为微藻多样性研究的热点,因此有必要按照新的分类学标准(包括形态学和系统发育学标准)对我国的骨条藻多样性进行研究。本文以威海市区沿岸水域为考查对象,小范围较高密度的进行骨条藻水样的采集(共设8个采样站点,每个采样站点进行多位点采样),采用微吸管法共获得30个单克隆藻株,对各藻株进行光镜和电镜观察,并通过PCR扩增获得各藻株的SSU、LSU和ITS序列进行系统发育分析,最后结合形态观察和系统发育分析结果对骨条藻种进行鉴定。结果只鉴定出3种骨条藻,即中肋骨条藻***(Greville)Cleve *** and Sarno、多恩骨条藻*** Sarno et Kooistra和拟中肋骨条藻*** Medlin *** et Kooistra,其中后者是在我国海域新发现的记录种,而且结果还表明在每一采样站位都有2种骨条藻种的存在。因此,威海沿岸的骨条藻组成为中肋骨条藻、多恩骨条藻和拟中肋骨条藻,新记录种的发现进一步表明我国沿海骨条藻的多样性,同一水域存在2种骨条藻也表明了骨条藻地理分布的复杂性。
