Objective To investigate the main factors of semen alkaline single cell gel electrophoresis(SCGE) and optimize the conditions, which can conductive to the developmentof recommended standard operating. Methods DNA fragmentation of sperm treated withdifferent concentrations of H2O2 was determined by alkaline SCGE. Gel layers, pH duringunwinding and electrophoresis, time of unwinding or electrophoresis and sperm countswere optimized. The reproducibility of optimized method was analyzed. Results Three layerscould reduce backgroud. PH10 was the optimal pH during unwinding and electrophoresis.40 minutes and 30 minutes were best time for unwinding and *** sperm was adequate. The intra-assay reproducibility of SCGE were 2.26%(1.29%), and the inter-assay reproducibility were 7.63%(5.84%). Conclusion Optimized semen SCGE was an repeatable and easily standardized method for clinical application.
