Tyrosinase(EC 1.14.18.1) is the key enzyme of browning in certain fruits and vegetables atthe time of postharvest handling, and is considered to be deleterious to the color qualityof their derived foods and beverages. Canarium Album also is called Chinese olive and ithas a very high economic and practical value. But the fresh Canarium Album is easy tobrowning and hard to store. Hence, it is important to search a potent and safe CanariumAlbum Fresh Keeper. p-Ethlybenzaldehyde cysteine schiff base(ECS) was firstly synthesizedand established by 1H NMR and 13 C NMR. The ECS's purity is 98% and the synthesisproductive rate is 75.3%. The ECS can inhibit the diphenolase acitivty of mushroom *** IC50 value was determined to be 0.735 μM, and the inhibitory type was found tobe mix-typed with KI of 0.213 μM and KIS of 0.472 μM. Furthermore, the fresh Canariumfruit which was treat with 20μM ECS can store longer as 120 days than control. Throughthe transmission electron microscope(TEM) and bacteriostatic test, the result shown thatthe ECS can effectively protect Canarium fruit from browning and bacteria invasion. Theacute toxicity result suggested that the ECS is completely non-toxic. Cysteine schiff basecan lead new generation of Canarium Album fresh keeper: Effective and Non-toxic.

Raf1 kinase inhibitor protein(RKIP)directly interacts with Raf1 kinase,thereby regulatingthe Raf1/MEK/ERK pathway which is vital for cell growth and ***,little is known about the molecular mechanism of RKIP binding with *** N-terminaldomain of human Raf1(hRaf11-147aa)and human RKIP(hRKIP)were recombinant preparedhere and GST pull down experiment suggests an interplay involving hRaf11-147aaand hRKIP.15N-1H HSQC NMR experiments show hRaf11-147aa binds with hRKIP at itsconserved ligand-binding pocket,loop"127-149",and the C-terminal *** dissociationconstant is 6.49±0.39μM determined by exogenous fluorescent experiments.D70,D72,E83,Y120,and Y181 were further verified as key residues for hRKIP binding withhRaf11-147aa by site-directed *** G143-R146 was critical for the interactionof RKIP and Raf1,whose deletion causes binding affinity decreasing around *** results can provide important structural clue for designing lead compound by disruptingthe RKIP-Raf1 interaction.

Spontaneous regression of neuroblastoma(NB)resembles the developmentally regulatedprogrammed cell death(PCD)of sympathetic *** tumor cells express highlevels of the nerve growth factor(NGF)receptors TRKA and p75NTR and are dependent onNGF for survival;however,the underlying molecular mechanism remains ***,weshow that UNC5D,a dependence receptor that is directly targeted by p53 family members,is highly expressed in favorable *** withdrawal strongly upregulated UNC5D,E2F1,and p53 in human primary favorable *** induced UNC5D was cleaved by caspases2/3,and the released intracellular fragment translocated into the nucleus and interactedwith E2F1 to selectively transactivate the proapoptotic target *** cleavage ofUNC5D and its induction of apoptosis were strongly inhibited by addition of ***5d(-/-)mice consistently exhibited a significant increase in dorsal root ganglia neuronsand resistance to NGF depletion-induced apoptosis in sympathetic neurons compared withwild-type *** data suggest that UNC5D forms a positive feedback loop with p53 andE2F1 to promote NGF dependence-mediated PCD during NB regression.

Distant metastasis is the major cause of nasopharyngeal carcinoma(NPC) treatment failure,while epithelial-mesenchymal transition(EMT) is a critical process for NPC invasionand metastasis. Pigment epithelium-derived factor(PEDF) is a multifunctional secreted glycoproteinthat displays broad anti-tumor activity. In this study we report that PEDF is apparentlydecreased in 218 human NPC tissues compared with chronic inflammation of thenasopharynx by immunostaining. Furthermore, PEDF expression is high in nasopharyngealepithelial cells, whereas it is lost during malignant progression of human NPC. More importantly,Overexpression of PEDF suppressed spontaneous liver metastasis in nude ***, fored overexpression of PEDF in highly metastatic NPC cells greatly decreasestheir invasive/migratory ability, increasesepithelial markers(E-cadherin and α-catenin),down-regulates mesenchymal markers(N-cadherin and vimentin). Finally, we demonstratePEDF directly targeted Wnt coreceptor LRP6 and decreased the Wnt/β-catenin signalingrelated protein in NPC cells. PEDF-reversed EMT could be abolished by either the applicationof the glycogen synthesis kinase3β(Gsk3β) inhibitor CHIR99021 or the specific neutralizingantibody for the binding domain of PEDF with LRP6. Our findings indicate thatPEDF could suppress the progression of NPC via EMT inhibition, and therefore serve as apromising candidate for NPC therapy.

Increasing evidence has shown that microRNAs(miRNAs) are involved in regulating tumordevelopment and progression. In this study, miR-409-3p was identified as a tumor suppressorin human colorectal cancer metastasis. MiR-409-3p expression was significantlydecreased in primary colorectal cancer tissues than in the paired normal tissues and it wasfound to be associated with tumor lymph node metastasis. Overexpression of miR-409-3pin CRC cells dramatically suppressed their migration and invasion in vitro and distal pulmonarymetastases in vivo. Further studies revealed that miR-409-3p suppressed the expressionof Grb2-associated binding protein(GAB1) by directly binding to its 3'-untranslated region. Silence of GAB1 could recapitulate the effects of miR-409-3p overexpression,while restoration of GAB1 in miR-409-3p overexpression CRC cells partially reversedthe miR-409-3p-mediated suppressive effects. Taken together, these results demonstrated that miR-409-3p suppressed CRC cells migration and invasion though directly targeting the GAB1 and provided a promising insight in developing novel diagnostic and therapeutic strategy for colorectal cancer treatment.

The polyamines are small basic molecules essential for cellular proliferation and *** levels of polyamines are associated with several human diseases and diverse cancers,and the enzyme activity is related to the beginning and successive development of neoplasticdiseases. An autoregulatory circuit that responds to the intracellular level ofpolyamines regulates their production. In the center of this circuit is a family of small proteinstermed antizymes. Antizymes themselves are regulated at the translational level bythe level of polyamines. Antizymes bind ornithine decarboxylase(ODC) subunits and targetthem to ubiquitin-independent degradation by the 26 S proteasome. In addition, antizymesinhibit polyamine transport across the plasma membrane via an as yet unresolved *** may also interact with and target degradation of other growthregulatingproteins. An inactive ODC-homologous protein termed antizyme inhibitor regulatespolyamine metabolism by inhibiting antizymes' function. The ability of antizymes todegrade ODC, inhibit polyamine uptake and consequently suppress cellular proliferationsuggests that they act as tumor suppressors, while the ability of antizyme inhibitors tocounteract antizyme function indicates their growth-promoting and oncogenic *** this study, we designed a series of antizyme inhibitor-1(AZI-1) mutants that can not inhibitAZ-1, but do not affect the other functions of AZI-1. To observe the effects of AZI-1mutants' over-expression on cellular proliferation without acting on AZ-1, we hope tofind out a new role of AZI-1 in regulating cancer proliferation found in our previous study.

In recent years, the global temperature warming has become difficult to reverse climatetrends. Same time, with the accelerated process of industrialization in China, the hightemperatureoperation growing groups of workers. This is a direct result of the growingnumber of people have to face the threat of heat stress. Heat stress lead to cardiac stressincreased dramatically, thereby giving rise to cardiac dysfunction. Mitochondria play a keyrole in cardiomyocytes apoptosis induced by heat stress. However, up to now, the regulatorymolecular mechanism by which heat stress induces mitochondrial apoptosis pathwayopening in cardiomyocyte remains unknown. Recent investigation indicates that heatstress leads to increased levels of mitochondrial GRP75. In our previous study, we foundthat GRP75 protected cells against the disorder of mitochondrial energy metabolism inducedby over expression of NGFI-B under stress. The present study shows that heat stressinduced mitochondria disfunction and cardiomyocyte apoptosis. We also found the protectiveeffect of GRP75 in cardiac disfunction induced by heat stress. The GRP75 on Ca2 +metabolism and mitochondrial energy metabolism in myocytes of rats have been revealedin this study. GRP75 can inhibit the rised of calcium levels induced by heat stress, andmaintain normal energy metabolism of mitochondria. Those results will provide some scientificevidence for revealing the cellular and molecular mechanism of GRP75 preventcardic disfunction induced by heat stress.

Natural BH3-memitic(-)-gossypol((-)-Gos) shows promising antitumor efficacy in severalkinds of cancers, but it induces protective autophagy in hepatocellular carcinoma(HCC)cells, which leads to drug resistance. In the present study, we for the first time demonstratedthat Hsp90 inhibitor 17-AAG enhanced(-)-Gos-induced apoptosis via suppressing(-)-Gostriggered protective autophagy and Mcl-1 accumulation. The suppression effect of17-AAG on autophagy was mediated by ERK inactivation, which was not associated withthe instability of Beclin1 or LC3 protein. Morover, 17-AAG downregulated(-)-GostriggeredMcl-1 accumulation by inhibiting Mcl-1Thr163 phosphorylation and promotingprotein degradation. Collectively, Our study indicates that Hsp90 plays an important role intumor maintenance, and repression of Hsp90 may be a novel strategy for Bcl-2-targetedchemotherapies, especially for overcoming the drug resistance(This work was supportedby cstc2013jjB10015 and NSFC81273226).
