Thaumatin-like proteins(TLPs) are the products of a large, highly complex gene family involvedin host defence and belong to the pathogenesis-related family(PR-5) of plant defenseproteins. Most TLPs exhibit potential antifugal activities and theiraccumulation inthe plant is related to many physiological *** this paper a novel antifungal proteinwas isolated from naked oat(Avena nuda) seeds by a procedure involving acetonefractionation,(NH4)2SO4 precipitation, Q-Sepharose ion exchange chromatography, chitinaffinity chromatography and Sephacryl S-200,identified by matrix-assisted laser desorptionionization–time of flight mass spectrometry analysis. The antifungal activity of thisprotein was detected by methed of Sterile Oxford cups and observed by Delta Vision *** results showed that the protein isolated from naked oat seeds exhibited a molecularmass of 23,760 Da in gel filtration and SDS-PAGE, which is homology with thaumatin-likeand pathogenesis-related proteins of the PRP-5 family. The protein also exhibited antifungalactivity against Trametes sp. SQ01(IC50=5.68 μM), Trichoderma spp.(IC50=0.83μM) by inducing mycelium deformation, rupture, and protoplasm lysis.

pp-GalNAc-T6(GALNT6) one of the large enzyme family of UDP-N-acetylgalactosamine:polypeptide N-acetylgalactosam inyltransferases(ppGalNAc-Ts), which catalyzes the initial step in the biosynthesis of mucin-type O-glycan by transferring GalNAc from UDP-GalNAc to the Ser and Thr residue of polypeptide acceptors. It has been reported to be involved in breast tumor, but the role of GALNT6 in the biological characters of breast cancer is not well understood. In this study, we found the lower expression of GALNT6 in MCF-7 cells decreased cell proliferation and migration, whereas over expression GALNT6 increased them. And the expression of Mucin protein 1(MUC1), matrix metall oproteinases-2(MMP-2) and matrix metalloproteinases-9(MMP-9), is positively correlated with the expression ofGALNT6. The expression of E-cadherin was increased in the lower expression of GALNT6 inMCF-7 cells. These results suggest that the effects of GALNT6 on proliferation, invasion and migration of MCF-7 may be through affecting the MMPs and E-cadherin. Taken together,our findings indicate that GALNT6 plays a role in cell proliferation, migration and invasion in breast cancer, and provide molecular evidence of a new adjuvant strategy for therapy of breast cancer.

Heparin-derived oligosaccharide(HDO) is isolated and purified from Low MolecularWeight Heparin(LMWH), the product obtained from standard heparin via chemical andenzymatic depolymerization. As the study of structure-activity relationship of HDO furthers,it reveals that HDO with different polymerization degrees(dp) can interact with variousproteins, which results in such biological activities as anti-inflammation, antiproliferationof vascular smooth muscle cells(VSMCs). In our previous study, we found thatheparin-derived oligosaccharide(HDO) with average molecular weight of 3000 could inhibitVSMC proliferation. In this study, we use HDO to construct VEGF-induced and PDGFinducedVSMCs proliferation model firstly. Then Use the MTT assay and cell cycle determinationto explore heparin oligosaccharides on VSMCs proliferation activity. Results of MTTassay showed that 0.01, 0.1 and 1 μmol?L-1 HDO could inhibit VEGF-induced and PDGFinducedVSMCs proliferation. Results of flow cytometric showed that HDO inhibited VEGF-induced and PDGF-induced VSMC proliferation by blocking G1/S transition. HDO couldaffect the expression of the level of VEGF、PDGF. In this study, the level of VEGF、PDGFreceptors were determined by RT-PCR 、Western blotting and IP. The results showed thatHDO could affect the expression of VEGF、PDGF receptors. So we thought that HDO mightinhibit vascular smooth muscle cells proliferation through down-regulating receptors andthen.

Poria extracts used in traditional Chinese medicine have widely physiological effects includingcontributing to improving the immunity of the organism. The polysaccharide is animportant active ingredient in Poria extracts. We have previously shown that MAP4K3 is acomponent on the amino acid-sensitive signaling pathway to mTOR. Moreover, MAP4K3 kinase links signal transduction from SLP-76 to PKC-θ in the TCR signaling cascade andcould be used as new diagnostic biomarkers and therapeutic targets for autoimmunity ***, we report that Pior polysaccharide extracts have gotten by using water extractionmethod and alkaline extraction method. Further we also purify the *** results show that the acid polysaccharide extracts can inhibit the growth of CandidaAlbicans and its MIC is 12.5mg/mL, and neutral polysaccharide extracts can significantly attenuatewild type and its mutant MAP4K3 kinase activity in vitro. This study provides somebasis data and useful method for the subsequent in vivo experiments.

EZH2(enhancer of Zeste homolog2),the core subunit of polycomb protein complex(PcG), is a histone methyltransferase which involves in cell density maintenance, stem cellpluripotent, cell cycle regulation and other important physiological roles. Recent studieshave found that EZH2 is over-expressed in many tumor tissues and can be used as a targetfor anti-tumor therapy.3-Deazaneplanocin A(DZnep), an S-adenosyl-L-homocysteine hydrolase inhibitor, can inhibitsthe expression of EZH2 and reduces the trimethylation level of lysine27 on H3, whichfinally leads to the death of tumor cells. The aim of this study is to determine the effect ofDZnep on human HL-60 acute myeloid leukemia cells in cell growth and *** experiment confirmed that DZnep could inhibit the growth of HL60 human leukemiacells in a dose- and time-dependent manner. Notably, even in very low concentration(0.05mmol/L), DZnep could induce apoptosis of HL-60 cells measured by DNA fragmentation,Cytochrome C release from mitochondria and decreased mitochondrial transmembranepotential. These data suggest that DZnep has the potential value in the treatment of the acute promylocytic leukemia.

In our previous work, we found water-soluble polysaccharide extracted from Gynostemmapenta phyllum Makino directly stimulate the proliferation of mouse splenocytes and *** order to evaluated Systematically the immunostimulatory activity of G. Pentaphyllum Makino polysaccharide(GPMPP), GPMPP were investigated to determine their invitro and in vivo immunomodulatory activities. Immunomodulatory activity assays in vitro showed that GPMPP could directly stimulate the proliferation of mouse splenocyts aloneor combining with ConA or LPS, stimulate the proliferation of mouse macrophages, increases the production of NO in a dose-dependent manner, which was significantly induced at 400 μg/mL. in vivo, immunosuppressed mouse models were generated by administration of cyclophosphamide(Cy) and then treated with GPMPP. The results show that GPMPP significantly increased the spleen and thymus indices, activated the proliferation of leukocytes from peripheral blood and bone marrow karyote, and enhanced the spleen lymphocyte activity and macrophage function. Furthermore, GPMPP increased the activity of NK cell in Cy-treated mice, as well as IgM and IgG levels in the sera of Cy treat edmice. In addition, GPMPP recovered the decreasing CD4+ lymphocyte numbers and theCD4+/CD8+ ratio. In conclusion, our study demonstrates that GPMPP can enhance the cellularimmunity, humoral immunity and non-specific immunity of immunosuppressed *** results suggested that GPMPP might be a potential natural immunomodulator.

Most β-thalassemia are caused by β-globin gene mutations, while it comprehend clinicallyvery heterogeneous group of thalassemia-like disorders, ranging in severity from theasymptomatic state to the severe transfusion-dependent type. Recently, an increasingnumber of researches suggested that epigenetic changes anticipated the occurrence anddevelopment of β-thalassemia. In order to investigate if lncRNAs expressed differentially inβ-thalassemia, we collected 30 β-thalassemia major and 30 β-thalassemia carrier statesamples, matched with 30 normal controls. With abundant and varied probes accountingfor 33,045 lncRNAs in Arraystar Human lncRNA Microarray assay, 28,443 lncRNAs thatwere expressed at a specific level were detected. Compared with normal control, 1424 lncRNAs were identified up-regulated and 2045 down-regulated in β-thalassemia major,623 lncRNAs were identified up-regulated and 349 down-regulated in β-thalassemia carrierstate. Compared with β-thalassemia carrier state, 1367 lncRNAs were identified upregulatedand 2356 lncRNAs downregulated in all β-thalassemia major samples. Rankingthe expression level of lncRNAs based on the microarray results, we selected 5 lncRNAs toanalyze the relationship between lncRNAs and β-thalassemia. DQ583499, Xist, LincRNA-TPM1, MRFS16 P and LincRNA-RUNX2-2 were evaluated using qPCR in all samplesand the results were consistent with those of the microarray assay. This study implicated that the abnormal expression of lncRNAs in β-thalassemia may be correlated with variant clinical phenotypes.

MiR-290 cluster miRNAs are the most dominant stem cell specific miRNAs in mouse andexpressed specically in early embryos and embryonic germ cells. These miRNAs play criticalroles in the maintenance of pluripotency and self-renewal in mouse stem cells. However,only several genes associated with roles in cell cycle, differentiation, apoptosis and NFkBsignaling were identified as target genes of these miRNAs. Here we showed miR-290 cluster miRNAs could prompt cell proliferation and G1-to-S phase transition of cell cycle inmouse stem cell CGR8. Cyclin D1(Ccnd1) was validated as a target gene of these *** were negative relationships between the expressions of Ccnd1 protein and miR-290 cluster miRNAs in undifferentiated CGR8 cells and differentiated cells. Our results provideda new insight into the function of the miR-290 cluster miRNAs in G1-to-S phase transitionin mouse stem cell cell cycle.

Tyrosinase(EC 1.14.18.1) is the key enzyme of browning in certain fruits and vegetables atthe time of postharvest handling, and is considered to be deleterious to the color qualityof their derived foods and beverages. Canarium Album also is called Chinese olive and ithas a very high economic and practical value. But the fresh Canarium Album is easy tobrowning and hard to store. Hence, it is important to search a potent and safe CanariumAlbum Fresh Keeper. p-Ethlybenzaldehyde cysteine schiff base(ECS) was firstly synthesizedand established by 1H NMR and 13 C NMR. The ECS's purity is 98% and the synthesisproductive rate is 75.3%. The ECS can inhibit the diphenolase acitivty of mushroom *** IC50 value was determined to be 0.735 μM, and the inhibitory type was found tobe mix-typed with KI of 0.213 μM and KIS of 0.472 μM. Furthermore, the fresh Canariumfruit which was treat with 20μM ECS can store longer as 120 days than control. Throughthe transmission electron microscope(TEM) and bacteriostatic test, the result shown thatthe ECS can effectively protect Canarium fruit from browning and bacteria invasion. Theacute toxicity result suggested that the ECS is completely non-toxic. Cysteine schiff basecan lead new generation of Canarium Album fresh keeper: Effective and Non-toxic.
