AMP激活的蛋白激酶(AMP-activated protein Kinase,AMPK)是一类重要的代谢相关激酶,在调节机体代谢和能量平衡中起着重要的作用,因而成为治疗肥胖症、II型糖尿病和某些癌症的药物靶点。哺乳动物AMPK蛋白是一个异源三聚体,由alpha/beta/gamma三个亚基组成,在人类基因组中,alpha亚基有两种,beta亚基有两种,gamma亚基有三种,这些亚基的组合可以形成多达12种的AMPK亚型蛋白,而不同亚基组合的蛋白在人体内的分布和功能各有差异。我们主要针对AMPK全长蛋白的结构进行了研究,希望获得全长蛋白的晶体结构,以揭示其行使生物学功能的结构基础。对此我们进行了多种尝试,制备了多种不同亚基组合、不同修饰状态或者不同截短片段的蛋白样品。在12种全长蛋白的组合中,除了gamma3亚基组合的蛋白之外,其余的8种均能获得高纯度的可溶蛋白样品。我们对拿到的可溶性蛋白进行了初步的结晶筛选,包括将AMPK全酶蛋白与不同核酸分子(AMP/ATP/ADP)进行共结晶,在某些条件下获得了细小的针状晶体,正在进行进一步的筛选和优化。同时,我们也测定了不同组合的AMPK全酶蛋白的激酶活力以及AMP对其活力的影响,对AMP激活不同AMPK亚型蛋白的作用有了一定的了解。
