生长在泰山的白花丹参是紫花丹参植物种的变型,系多年生草本植物,为山东特产药材之一,早年就被誉为妇科良药,因具有降血脂、降血压、延年益寿之功效。Myb转录调控因子广泛存在于植物中,其主要特征是含有1-3个由51或52个氨基酸残基组成的不完全重复的Myb-DNA结合结构域(R1、R2、R3)。每个结构域由3个α-螺旋组成,其中第二与第三个螺旋形成螺旋-转角-螺旋(HTH)结构,根据所含Myb-DNA结合结构域的数目可分为R1、R2R3、R1R2R3三个家族,其中R2R3家族对植物次生代谢具有十分重要的调控作用,R2R3-MYB转录因子作为调节蛋白广泛参与苯丙烷类代谢途径的调控,主要的证据来自对拟南芥、玉米和矮牵牛等植物中黄酮类分支途径的生化和遗传学研究。从我们建立的白花丹参cDNA文库中成功克隆到得R2R3-MYB转录因子基因,此基因的ORF为642bp,编码的氨基酸为214,命名为SmMYBP(GenBank ID KJ009321)。SmMYBP的氨基酸序列与调节胡萝卜PAL活性的Dcmyb1的相似性为77%,与MJ诱导的烟草NtMYB1、Ntmyb2的相似性为72%和71%,与拟南芥AtMYB15的相似性为70%,与雪莲myb-like P的相似性为78%。序列分析发现SmMYBP具有典型的MYB因子的R2R3区域。[D/E]Lx2[R/K]x3Lx6Lx3R为典型的与bHLHproteins互作的区域。A phylogenetic tree分析发现SmMYBP与天山雪莲的Myb-like proteinP、与调节胡萝卜PAL活性的Dcmyb1和MJ诱导的烟草NtMYB1、Ntmyb2聚为一类。推测SmMYBP可能参与调节茉莉酸诱导的白花丹参酚酸类物质的合成。从一个月龄的白花丹参叶片、茎和根中提取总RNA,采用real-time RT-PCR研究SmMYBP在不同组织的表达模式,结果显示,SmMYBP在白花丹参叶片茎和根中的都有表达,但在根里面的表达量最高,茎和叶片里的表达量基本一致。为了研究SmMYBP对于MeJA刺激的反应,我们用0.1 mM MeJA处理培养了15天的白花丹参毛状根,并分别提取了RNA。采用real-timeRT-PCR研究了SmMYBP对于0.1 mM MeJA处理后的表达模式。结果显示,0.1 mM MeJA处理毛状根36 h后,SmMYBP的表达量达到最大值,是对照的6.54倍,随后表达量逐渐下降。我们将SmMYBP连接到pMD18-T Vector得到pMD18-T-SmMYBP载体,测序验证。然后用带有BamHI接头的引物和KpnI接头的引物扩增pMD18-T-SmMYBP质粒,将PCR产物连入pMD19-T simple质粒,得到pMD19-T simpleSmMYBP质粒。用BamHI+KpnI同时酶切pMD19-T simple-SmMYBP和pCAMBIA2300质粒,分别回收pMD19-T simpleSmMYBP的MYBP基因片段和pCAMBIA1305的大片段,用T4 DNA连接酶连接,获得NPTII抗性的植物表达载体pCAMBIA2300-SmMYBP,为我们下一步的SmMYBP的功能研究奠定了基础。

MiRNAs are important gene regulators in the progress of tumorigenesis. Tumor proteinp53, a transcriptional factor, plays an important role in the progression of *** this study, we analyzed p53 can bind to the promoter of miR-10 a and miR-10 a as a tumorsuppressor in non-small cell lung cancer. We showed that MAP3K7 and WNK3 can bedirectly targeted by miR-10 a in SPCA-1 and 95-D cell line;the activity of luciferase tranfectedmiR-10 a mimics was lower by 60% and 45% than that of miR-10 a negative control(NC) did respectively. Moreover, the endogenous MAP3K7 was downregulated by 50% atleast when miR-10 a was transfected compared to miR-10 a NC or miR-183, which didn'tcontained binding sites with 3′-UTR of MAP3K7. And also the endogenous expression ofWNK3 was inhibited by 70% at least. In addition, we also detected the expression level ofmiR-10 a was adverse in different NSCLC cell lines, 95-D and SPCA-1. Furthermore, wefound that miR-10 a overexpression inhibited the proliferation and triggered apoptosis innon-small cell lung cancer cell line which is opposite with the oncogene WNK3 or MAP3K7 overexpression in NSCLC cell lines. The expression level of miR-10 a was found to be significantlycorrelated with tumor differentiation and pathological stage. These findings suggestedthat miR-10 a regulated survival of SPCA-1 and 95-D cell line partially through the down regulation of MAP3K7 and WNK3. These findings suggest that miR-10 a, p53 protein and relevant target genes are members of a regulatory network in NSCLC tumorigenesis.

Introduction. Achyranthes root(Niu xi) is one of the Chinese herbs often used to treat avariety of disorders such as low back pain, circulatory disorders and aging. Extracts ofachyranthes are found to be highly rich in Beta-Ecdysone(βEcd). In mammals including thehuman βEcd is known to stimulate muscular ***(GCs) are frequently used in clinical medicine to treat non-infectious inflammatorydiseases. However, GC use results in rapid trabecular bone loss and a high incidentfracture risk. We previously demonstrated that glucocorticoids excess(GC) alters osteocytecell viability through autophagy or apoptosis and that GC treatment dosedependentlyalter gene expression for autophagy, an intracellular degradative mechanismto remove the dysfunctional organelles to keep the cells alive, or apoptosis. Lower GC dose(0.7-2.1mg/kg/d) activating autophagy, while higher dose(5.6mg/kg/d) decreased autophagybut increased apoptosis. The purpose of this study was to evaluate the effects ofβEcd on osteogenesis, osteocyte viability(autophagy and apoptosis) and bone loss followingGC ***. Two-month-old male Swiss-Webster mice received either placebo(PL) or prednisolone(5.6mg/kg/d) and then treated immediately with βEcd(400μg sc 5x/wk) for threeweeks. Trabecular bone volume and surface based bone turnover were measured at thedistal femurs(DF), lumbar vertebral bodies(LVB) and mid-femoral shafts(FS) by Micro CTand bone histomorphometry. Osteocyte autophagy and apoptosis were measured at theLVB as well as on FS by western ***. Glucocorticoid(GC) treatment in growing mice inhibited age-dependent trabecularand cortical bone gain, which were accompanied by a 30-50% lower mineralizing surface(MS/BS) and bone formation rate(BFR/BS). Radiotherapy is one of the most effective and important measure for breast cancer ***, with the effect of furthest controlling tumor, radiotherapy also has a radiation-induced bystander effect on normal tissue around the tumor. double-strand breaks(DSBs) repair by normal cells around is the best way to reduce the low dose radiation-induced bystander effect. Thus, it is important to study the mechanism of DNA repair of low dose radiation(LDR). Caveolin-1(Cav-1) is a symbol protein of Caveolae and an important regulatory molecule of cell signal transduction. Our previous study showed that the expression of Cav-1 reducing induce the transformation and carcinogenesis of mammarycells. Recent study showed that Cav-1 is associated with the DNA damage repair induced by radiation, but the mechanism is unclear. In present study, radiation-induced bystander effect was simulated on human mammary epithelia cells MCF10, which is irradiated by LDR Gamma rays(50cGy and 100cGy), the sensitivity changes of MCF10 cells to LDR were observed by MTT. At the same time, Western blot was used to determine the expressions of main proteins related to the DNA repair pathways mediated by DNAP Kcs and ATM, the proliferation pathway by MAPK, the survival pathway by PI3K/AKT and the expression of Cav-1. The results showed that: After 50 cGy and 100 cGy radiation, cell proliferation could be inhibited but more serious by 100 cGy radiation, indicating that the sensitivity of human mammary epithelia cells to LDR was dose-dependent. The radiation of100 cGy could increase the expression of ATM, p53, DNA-PKcs, and other DNA repair-related proteins, indicating that DSBs repair induced by LDR could act through both homologous recombination(HR) and Non-homologous end-joining(NHEJ) pathways. The radiation of 100 cGy could enhance the activities of MAPK and AKT protein, increase the expression of transcription factor NF-κB and change the expression of Cav-1 protein, indicating that Cav-1 mediat

This study elucidated the role of CaN-NFAT signaling and neurotrophins on the transformation of myosin heavy chain isoforms in the rat soleus muscle fiber following aerobicexercise training. To do so, we examined the content and distribution of myosin heavychain(MyHC) isoforms in the rat soleus muscle fiber, the activity of CaN and expression ofNFATc1 in these fibers, and changes in the expression of nerve growth factor(NGF), brain derived neurotrophic factor(BDNF) and neutrophin-3(NT-3) in the soleus and striatum following high-and medium-intensity aerobic treadmill ***:Specific pathogen-free 2 month old male Sprague-Dawley(SD) rats were randomly divided into three groups: Control group(Con, n=8), moderate-intensity aerobic exercise group(M-Ex, n=8) and high-intensity aerobic exercise group(H-Ex, n=8). The aerobic exercise regime used in this study was proposed by Ding, with reference to the exercise load standards of Bedford. We used ATP staining to identify the muscle fiber type Iand II, SDS-PAGE to separate and analyze the isoforms MyHCI, MyHCIIA, MyHCIIB and My-HCIIx, and performed western blots to determine the expression of NFATc1, NGF, BDN Fand NT-3. CaN activity was measured using a colorimetric ***:(1) In the soleus muscle, 8 weeks of moderate-intensity exercise can induce transformation of MyHC IIA and MyHC IIB to MyHC IIX and MyHC I, while high-intensity treadmill exercise can transform MyHC IIx to MyHC IIB, MyHC IIA and MyHC I.(2) In comparison to the control group, CaN activity was significantly increased in both the M-Ex and H-Exgroups, with a more pronounced up regulation in the M-Ex group.(3) Eight weeks of moderate-and high-intensity aerobic exercise induced the expression of NGF, BDNF and NT-3in the soleus muscle and the striatum, with the most significant increase in the H-Ex ***: In the rat soleus muscle,(1) CaN–NFATcl signaling contributes to the conversion of MyHC I isoform in response to moderate-intensity

Cancer is one of the most important diseases threatening human's health. Current cancertherapies have their own limitations. For an instance, chemotherapy may lead to drugresistanceof cancer cells. Implementing chemotherapy can result in many changes in cancercell signaling pathways and markers, which may related to drug-resistance. These dynamicchanges are the key to find new strategies to overcome the limitations. Based oncurrent chemotherapy, finding such pathways and markers and screening novel compoundsor molecules to modify these dynamic changes to make combination therapiesmay become a new trend in the development of future cancer therapeutics. Monoclonalantibody has the advantages of higher specificity and fewer side-effects, which can be thefirst-choice combining with current chemotherapy. In this research, we reported a potentialanti-cancer combination therapy composed of Tamoxifen and monoclonal antibody18A4 against *** is a traditional anti-cancer drug. It is a selective estrogen receptor modulator(SERM), which can inhibit cancer cell growth. Some cancer cells may acquire Tamoxifen-resistance after a period of treatments. Anterior Gradient-2(AGR2) is a protein overexpressedin several kinds of cancer cells. Over-expression of AGR2 is closely related tocancer proliferation, metastasis. In our research, we find that AGR2 can be apparently inducedafter a 48-hour treatment with 5uM Tamoxifen. This dramatic dynamic changemakes combination therapy possible. To screen such a molecule to modulate this Tamoxifeninduced marker, we make an in-house patented monoclonal antibody against AGR2 named 18A4. 18A4 shows the apparent affinity of 10^7-10^9L/mol measured by noncompetetiveELISA. The combination therapy of 18A4 and Tamoxifen shows potential synergicanti-cancer *** vitro, the combination therapy can both inhibit cancer cell growth and arrest cell *** combination of 18A4 and Tamoxifen shows more growth inhibition of human breastcancer cell

Vitamin D insufficiency during pregnancy has been proved to be associated with increasedrisk of preeclampsia. We demonstrated that 25(OH)D in umbilical cord serum was also decreasedin preeclamptic patients. Meanwhile, maternal serum 25(OH)D concentrations correlatedsignificantly with umbilical cord serum 25(OH)D. VDR gene and protein expressionwere increased as well as CYP24A1 gene expression were decreased in placental tissue ofpreeclampsia. Placenta weight and newborn birth weight were negative correlative withVDR mRNA expression in placenta. We supposed that vitamin D may play a role in the processof placentation in the onset of preeclampsia. In order to assess the effect of vitamin Don preeclampsia, we conducted rat model of reduction in uteroplacental perfusion(RUPP).In the RUPP group, vascular stenosis surgery generated elevated blood pressure, increasedproteinuria, fetal death, decreased fetal weight and placental weight, decreased cardiacfunction and hyperglycemia. In RUPP plus 1,25(OH)2D injection group, compared withRUPP group, systolic blood pressure was reduced and offspring outcome was improved byincreasing fetus survival and placenta weight. 24 h urinary protein excretion and urine microalbuminconcentration decreased in RUPP+1,25(OH)2D group. Interestingly, hyperglycemiain RUPP group was relieved by 1,25(OH)2D addition. In conclusion, we proved that1, 25(OH)2D addition beginning at the early gestation had various effect on blood pressure,fetus and placenta, heart, kidney and metabolism. The apoptosis level of rat placentain three groups was assayed with TUNEL stain. Compared with RUPP group, the apoptosiscells decreased in RUPP+1,25(OH)2D group. Meanwhile, Immunohistochemistry stainingreleaved that caspase 12 and cleaved caspase 3 expression decreased inRUPP+1,25(OH)2D group compared with RUPP group. 1,25(OH)2D may inhibit apoptosisvia down regulated caspase 12 in placenta. α-glucosidase(EC 3.2.1.20) is a target enzyme to treat type 2 diabetes mellitus, hence, dueto the clinical relevance, it is important for studying this enzyme's inhibition. In this study,we investigated Co2+-induced inhibition as well as structural changes of α-glucosidase integratingwith computational simulations. α-glucosidase activity was inhibited by Co2+ ina dose-dependent manner. Co2+ inhibited α-glucosidase in a parabolic non-competitiveinhibition reaction(Ki = 0.78 ± 0.08 mM) and directly induced the regional unfolding ofthe enzyme resulting in a slight decrease of hydrophobic surface. The computational simulationsusing molecular dynamics showed that the simulation with Co2+ ions lost the secondarystructure by positioning Co2+ ions near the active site for glucose degradation, implyingthat the Co2+ ions can stimulate the unfolding of the enzyme. Our study revealedthe mechanism of Co2+ ligand binding mediated structural change as well as inhibition ofα-glucosidase, and suggested that Co2+ could act as a potent inhibitor of α-glucosidasefor the treatment of type 2 diabetes *** with computational simulations. α-glucosidase activity was inhibited by Co2+in a dose-dependent manner.
