High levels of low-density-lipoprotein cholesterol(LDL-C) is a major risk factor for cardiovascular *** LDL-C levels vary among humans and are heritable,the genetic factors affecting LDL-C are not fully ***,we identified a rare frameshift variant in the LIMA1(EPLIN or SREBP3) gene from a Chinese Kazakh ethnic family with inherited low LDL-C and reduced cholesterol ***1 was mainly expressed in the small intestine and localized on the brush border ***1 bridged NPC1 L1,an essential protein for cholesterol absorption,to a transportation complex containing myosin Vb and facilitated cholesterol ***1-deficient mice displayed reduced cholesterol absorption and were resistant to diet-induced hypercholesterolemia similar to the human ***,this study identifies LIMA1 as a key protein regulating intestinal cholesterol absorption.

Thioredoxin interacting protein(TXNIP) plays an important role in glucose-lipid *** acid synthase(FASN) catalyzes the last step in fatty acid biosynthesis,and thus,it is believed to be a major determinant of the maximal hepatic capacity to generate free fatty acid by de novo *** studies found that TXNIP protein level decreased(P<0.05) and FASN level increased(P<0.05) in the LO2 nonalcoholic fatty liver cell *** prompt TXNIP involved in lipid *** we knockdown TXNIP by siRNA(P<0.01) in LO2 *** then we detected the expression of TXNIP and FASN by western *** found knockdown TXNIP(P<0.05) caused the FASN protein increased(P<0.05),and induced the lipid accumulation by oil red staining in LO2 ***,TXNIP could play an important role via in hepatocellular lipid accumulation through *** TXNIP would cause lipid accumulation in hepatic cells.

This study aimed to elucidate the effects of supplementation with different doses of vitamin C(VC) and vitamin E(VE) on prothrombin time(PT) in mice;Forty-eight mice were randomly divided into six treatment groups that were fed different dose VC and VE except for control check(CK) for eight *** eight weeks,blood samples were obtained from the mouse canthus for the analysis of PT and platelet *** PT and platelet parameters results show that VC group,different doses of VE and VC&VE group was effective in PT,Platelet count(PLT) and Platelet hematocrit(PCT).F-test There is no significant difference between the platelet distribution width(PDW) and mean platelet volume(MPV) of different groups revealed an F-value of 0.547(p>0.05).The combined use of VC and VE decreased coagulation function in mice.

While many studies have shown that histamine and its signaling regulate energy homeostasis through the central nervous system (CNS), their roles in adipose tissues remain poorly understood. The gene of chondroitinase 1816(ChSase1816) was overexpressed and purified from Escherichia coli BL21(DE3) with a molecular mass of 86 *** the chondroitin sulfate B(CS-B) as the substrate,the Km and Vmax values of ChSase1816 towards CS-B was 7.85 μg/mL and 1.21 U/*** optimum reaction temperature and pH for ChSase1816 were 30℃ and 8.0,*** ChSase1816 activity was stable at 30°C,but not stable at 50°C and 60°***,ChSase1816 retained more than 60% of its maximum activity at a broad pH range of pH 4.0 and *** the metal ions detected had inhibitory effects on the *** result of Circular dichroism(CD) indicated the enzyme contained many antiparallel structure and random *** antioxidant activity assay showed that hydroxyl radical scavenging activity of chondroitin enzymatic hydrolysates was higher than chondroitin sulfate.

Tau蛋白和锌离子都主要富集在人中枢神经系统神经细胞的轴突中;Tau蛋白的异常磷酸化与Tau蛋白毒性直接相关,也是阿尔茨海默病另一个重要的病理特征。我们利用可诱导的细胞模型研究了病理浓度锌离子对全长Tau蛋白病理突变体Tau-K280积聚的影响,发现锌离子通过与全长Tau蛋白的Cys-291和Cys-322及其它两个氨基酸(His-330/362)形成四配位结构来促进Tau蛋白的积聚及细胞毒性;该聚集体能进一步诱导内源Tau蛋白积聚及异常磷酸化,从而使得Tau蛋白毒性放大;半胱氨酸突变后,锌离子与Tau蛋白相互作用模式改变,四配位结构被打乱从而丧失了这种对Tau蛋白积聚和毒性的促进作用,表明Cys-291和Cys-322是锌离子调控Tau蛋白体外和细胞水平异常积聚与毒性的关键位点。我们也发现锌离子显著促进全长Tau蛋白的磷酸化修饰和乙酰化修饰;锌离子作用下,Tau-K280稳转细胞中活性氧的含量升高,并诱导细胞产生自噬反应。我们建立了病理浓度锌离子调控Tau蛋白磷酸化修饰和积聚的细胞模型,锌离子不仅通过与Tau蛋白结合直接促进其积聚,更能对Tau蛋白异常的翻译后修饰起调控作用而进一步促进Tau积聚及细胞毒性。这些发现为研究锌离子对Tau蛋白积聚和毒性的调控机制及其在神经退行性疾病中的作用提供了有用的线索。

三陰性乳癌造成之遠端器官轉移經常導致病患預後不良且存活率較差,而三陰性乳癌經常轉移的器官包含肺、腦、肝及骨骼。長鏈非編碼核糖核酸被定義為一類新穎的核糖核酸,其轉錄物長度大於200個核苷酸且缺乏蛋白質轉譯之能力,目前長鏈非編碼核糖核酸已被報導為乳癌轉移重要的調控者。然而,長鏈非編碼核糖核酸調控三陰性乳癌轉移的相關機制目前仍然尚未完全了解。在我們研究中,我們發現長鏈非編碼核糖核酸ZNF469-3高度表現於肺轉移之三陰性乳癌細胞株LM2-4175,而長鏈非編碼核糖核酸ZNF469-3過表現也會促進細胞侵入能力及細胞之癌幹性。此外,於異種移植小鼠模式動物中,穩定表達長鏈非編碼核糖核酸ZNF469-3亦能促進三陰性乳癌之肺轉移。長鏈非編碼核糖核酸ZNF469-3也藉由結合其對應之微小核糖核酸574-5p,進一步阻止微小核糖核酸574-5p影響ZEB1之蛋白表現量降低。從臨床結果上,我們發現高表現長鏈非編碼核糖核酸ZNF469-3及ZEB1和三陰性乳癌肺轉移腫瘤復發有顯著相關性。綜合以上結果,我們的研究指出藉由調控微小核糖核酸574-5p及ZEB1訊息傳遞路徑,長鏈非編碼核糖核酸ZNF469-3能夠促進三陰性乳癌之肺轉移,且能夠作為三陰性乳癌肺轉移病患復發病患診斷及預後標準。
