Hepatocellular carcinoma(HCC) is linked to inflammation and ***2 is an innate immune pattern recognition receptor(PRR),which is specifically expressed in liver tissue and implicated in the regulation of ***,the role of PGLYRP2 in HCC remains *** this study,we aimed to investigate the epigenetic mechanism of PGLYRP2 silencing in HCC tissues and whether PGLYRP2 is able to influence HCC progression by regulating tumorassociated *** we demonstrated that PGLYRP2 significantly decreased in tumor tissues of HCC patients by hypermethylation of promoter region,and tumor PGLYRP2 level was positively associated with the *** array data indicated that PGLYRP2 level was also associated with antitumor immune responses in tumor microenvironment,including activation of complement system(C2,C3,C5),recruitment of tumor-infiltrating CD8+ T cells,macrophages,mature B cells and activated mast *** an orthotopic mouse model of HCC,immune-competent mice transplanted with low-PGLYRP2 expressing tumor cells exhibited aggressive tumor growth,whereas PGLYRP2 overexpression greatly inhibited tumor growth,but the inhibitory effect of PGLYRP2 in nude mice ***,our study suggested that PGLYRP2 inhibited HCC through enhancing antitumor immune responses in tumor microenvironment and offered therapeutic implications for immunosuppression-associated cancers,such as HCC.

神经管畸形(NTDs)是由胚胎发育期间神经管闭合缺陷引起的严重先天畸形。大量研究表明,microRNAs(miRNAs)通过不同的机制参与神经管发育的调节。迄今为止,只有少数研究评估了miRNAs在NTDs中的表达,且主要采用miRNA芯片分析。为了进一步研究miRNAs在NTDs中的分子机制,我们首次对全反式维甲酸(ATRA)诱导的小鼠NTDs进行了miRNA深度测序研究,并在三个不同的胚胎时间(E8.5、9.5和10.5)建立了小鼠神经管的总miRNA谱。在所有文库中,我们共检测到1309个miRNAs,其中308个为潜在的新miRNAs。在文库Bra8.5-vs-Brc8.5,Bra9.5-vs-Brc9.5和Bra10.5-vs-Brc10.5中,我们分别鉴定了241、271和213个差异表达的miRNAs,包含41个共同差异表达的miRNAs。对差异表达miRNAs的靶基因分析结合功能富集分析,我们筛选了对神经管的发育至关重要的细胞过程和分子相互作用网络,其中与CNS发展相关的共19个miRNA/mRNA负调节对,包括miR-124-3p/Hoxb1,miR-181b-1-3p/Egr2,miR-107-3p/Egr2和miR-6240/Arsa等。此外,E9.5和E10.5分别有19和5个负调控对参与wnt信号通路和hedgehog信号通路,如miR-124-3p/wnt2,miR-135a-5p/wnt2和miR-470-5p/Shh,表明miRNAs可能通过这些信号通路在NTDs中发挥重要作用。通过对差异表达的miRNA/mRNA的功能相互作用网络分析,我们又筛选了8个miRNA/mRNA负调节对,包括let-7e-5p/Hoxd1,let-7e-5p/Hoxb1,miR-181b-5p/Evx1,miR-125a-5p/Hoxb3,miR-9-5p/Evx1,miR-9-5p/Hoxb1,miR-125b-5p/Hoxb3和miR-222-3p/Ddit4。综上,我们的研究建立了第一个小鼠NTDs的总miRNA表达谱,可为解决NTDs分子机制或生物标志物发现等重要问题提供独特资源。

泡沫病毒(foamy virus,FV)是一类特殊的反转录病毒,能感染体外培养的细胞,引起细胞病变死亡,但其在体内并不引发任何病症,仅保持持续感染并伴随宿主终身,机体的免疫系统在此过程中发挥了重要作用。宿主限制性因子是指在病毒复制过程的某个特定步骤与病毒相互作用,从而阻断病毒复制的宿主蛋白。Schalfen(Slfn)家族蛋白可以抑制细胞增殖、调控造血细胞及T细胞分化等免疫应答活动,近年来发现其具有抗病毒作用,是一类新的宿主限制性因子。目前研究表明Slfn家族成员SLFN11能够结合tRNA,抵消因HIV存在所引起的tRNA的A/U偏好,从而抑制HIV病毒蛋白的合成,但目前机制并不清楚。为了探究SLFN11的抗病毒范围并解析机制,我们展开了SLFN11对原型泡沫病毒(prototype foamy virus,PFV)复制影响的研究。构建SLFN11的真核表达质粒并将其与PFV的感染性克隆共转293T细胞,结果显示SLFN11削弱了PFV的复制能力;HT1080细胞中感染实验结果亦相同。当敲低HT1080细胞中的SLFN11时,PFV的复制能力得以回复。表明SLFN11能够抑制PFV的复制。为了探究SLFN11拮抗PFV感染的机制,我们首先构建了PFV病毒蛋白Bet的真核表达质粒Bet (Wt)。随后根据宿主蛋白表达的密码子偏好,优化Bet (Wt)的密码子并将新的克隆命名为Bet (Opt)。将Bet (Wt)和Bet (Opt)分别与SLFN11共转293T细胞,结果发现SLFN11仅对Bet (Wt)的表达有抑制,而对Bet (Opt)的表达水平没有显著影响。由此推定SLFN11很可能通过改变密码子偏好性的方式限制PFV的感染。进一步确定SLFN11发挥抗病毒功能的关键结构域,我们构建了SLFN11的截短体SLFN11-N和SLFN11-C,利用其进行PFV的感染实验发现,SLFN11的N端和C端结构域对SLFN11抑制PFV都是不可或缺的。综上,本研究发现SLFN11能够通过改变密码子偏好性的方式拮抗PFV的感染,且这种限制作用需要SLFN11蛋白保持结构的完整性。这一研究有利于了解SLFN家族成员在天然免疫中扮演的重要角色,为进一步探索PFV的潜伏感染奠定了基础,可能丰富了我们对病毒宿主相互作用的认识。

In 2018,merely breast cancer occupies 30% of all cancers expected to occur in ***,tumor metastasis is one of the main reasons for poor tumor efficacy,especially for triple-negative breast cancer cell lines(TNBC).Thus,how to find effective biomarkers to track the metastatic development of breast cancer has been a hot *** critical enzymatic activator of glycolysis,the lactate dehydrogenase-A(LDH-A),lactate dehydrogenase-B(LDH-B) could be used as the breakthrough point to solve *** firstly investigated the expression level of LDH-A and LDH-B in several breast cancer cell lines and chose BT-549 cell(TNBC) as the research subject in which both LDH-A and LDH-B were lowly *** was confirmed that overexpression of LDH-A,LDH-B or both could significantly promote the growth,migration,invasion and clone formation ability of cells through combining Western blotting analysis,MTT Assay,wound-healing Assay,matrigel invasion assays,gelatin zymography and colony formation ***,overexpression of LDH-A had stronger promoting effect on procession of breast cancer cells than overexpression of LDH-B or both,while simultaneous overexpression of LDH-A and LDH-B did not induce superimpose *** together,this study investigated the overexpression of both LDH-A and LDH-B in BT-549 cell for the first time and presented that overexpression of LDH-A,LDH-B or both could enhance the metastatic biological behavior of breast *** results not only improve our understanding of lactate dehydrogenase but also provide potential biomarkers to track the metastatic development of breast cancer.

Analysis of blood-based glycans represents a new strategy for cancer *** G(IgG),a highly abundant glycoprotein in serum,is known to mediate a variety of blood immune *** has been an increasing interest in the analysis of the N-glycan profile of human IgG in healthy and disease states,such as auto-immune diseases and *** altered glycosylation of total serum IgG has been profiled individually in ovarian,prostate,gastric cancers,no studies have systematically investigated the alteration of IgG glycosylation in multiple cancers *** addition,the sample sizes in previous studies were *** high throughput analytical method based on mass spectrometry for relative quantitative analysis of IgG glycans we previously developed,we designed for the first time a large-scale(n=5600),multi-institute study to assess the quantitative changes of IgG glycosylation in 12 types of cancers,inflammatory diseases,benign conditions as well as healthy individuals *** demonstrated that the distribution patterns of serum IgG galactosylation have similar alterations in multiple types of *** thus we provided evidences for the first time that serum IgG galactosylation has great potential to be used as a non-invasive pan-cancer biomarker for early-stage cancer detection and cancer *** studies are underway to investigate the role and mechanism of IgG galactosylation in readout of immunological status.

10-Hydroxycamptothecin(HCPT),a natural camptothecin(CPT) analog that is more potent anti-cancer activity,was widely used in recent clinical *** is becoming recognized as an important biological mechanism in human ***,the involvement of autophagy in the antiproliferative effects of HCPT on lung cancer remains *** we found that HCPT was able to reduce the cell viability on non-small cell lung cancer(NSCLC) H1299 cells after 24-h treatment as determined by MTT *** cytometry was used to distinguish between cells in different phases of the cell *** results showed that HCPT induced S phase and G2/M phase arrest,suggested that HCPT induced H1299 cell death by altering the cell cycle ***,HCPT remarkably induced the formation of autophagosomes with fluorescence microscopy after monodansylcadaverine(MDC) ***,HCPT obviously enhanced the expression of microtubule-associated protein 1 A/1 B-light chain 3-II(LC3-II) and down-regulated the expression of P62 in timedependent manners in H1299 ***,autophagy inhibitor 3-methyladenine(3-MA) and chloroquine(CQ) could inhibit HCPT mediated *** results demonstrated that HCPT was able to suppress proliferation and induce autophagy in H1299 cells,providing new mechanistic insights into the anticancer functions of HCPT.

To observe the changes of miRNA expression profiles in the brain of aging rats,and their relationship with the expression of learning and memory related genes,and the intervention effect of Reinforcing Kidney and Nourishing Qi herbs.24 months old rats were used as the aging model and were randomly divided into old group,Zuogui pill group,Yiqi Congming decoction group,mixture group and antagonist group,and 5 months old rats as youth *** microarray technology was used to observe the changes of miRNA expression profiles in brain tissue of each group of *** software was used to predict the target genes of the differential miRNAs and analyze the effect of miRNAs on the learning and memory *** results showed that a total of 761 miRNAs were *** with the young group,there were 126 abnormally expressed miRNAs in the old *** these abnormally expressed miRNAs,there were 8,21,7,18 miRNAs that had been improved by Zuogui Pills,Yiqi Congming Decoction,their Mixture,and antagonist(RU486) *** software analysis showed that the abnormally expressed miRNAs in the old group were significantly enriched in multiple signaling pathways which related to brain function degeneration,and many of these miRNAs have a targeted relationship with learning and memory related ***,Yiqi Congming decoction,their mixture and RU486 could correct the change of miRNA expression profile in brain tissue of aged rats in different degree.

Marine microalgae have been widely applied in cosmaceuticals,nutraceuticals,and functional *** the present study,we first investigated the hepatoprotective effects of peptide purified from microalgae,Isochrysis zhanjiangensis on HepG2 cells alcoholic *** was hydrolyzed utilizing chymotrypsin,trypsin,pepsin,and by in vitro gastrointestinal *** hydrolysates,the gastrointestinal hydrolysate showed relatively high free radical scavenging ability preliminarily and was purified with following sequential chromatography *** amino acid sequence and molecular mass of the purified peptide from ***(PIZ) was identified as Asn-Asp-Ala-Glu-Tyr-Gly-Ile-Cys-Gly-Phe(NDAEYGICGF;MW,1088.16 Da) via Q-TOF ESI/***,PIZ attenuated ethanol-induced cytotoxicity and inhibited the production of intracellular reactive oxygen species(ROS) by DFCH-DA *** blot results showed that superoxide dismutase(SOD) and glutathione(GSH) levels up-regulated with PIZ treatment before alcohol exposure while gamma-glutamyltransferase(GGT) protein expression *** results provide an opportunity to discover new highly active peptide(PIZ) against alcohol toxicity in HepG2 cells.
