目的研究急性冷刺激对大鼠心脏、肝脏、脾脏、肺脏、肾脏、肌肉及十二指肠中α-烯醇化酶(α-enolase,ENO1)基因表达量的影响。方法取健康大鼠10只,随机分为2组:冷应激组和常温对照组,每组5只。正常对照组饲养环境为24℃,冷应激组大鼠环境温度为4℃,应激时间为12 h。应激处理后分别提取两组内大鼠的心脏、肝脏、脾脏、肺脏、肾脏、肌肉及十二指肠组织样品,应用实时荧光定量PCR方法检测冷应激前后大鼠五种内脏器官及肌肉和十二指肠的EN01 m RNA表达量。结果冷应激组大鼠的肝脏、脾脏组织中ENO1基因m RNA的表达水平极显著高于正常对照组(P<0.01),肺脏与十二指肠组织中ENO1 m RNA表达水平显著高于正常对照组(P<0.05);而冷应激组大鼠心脏、肾脏与肌肉组织中ENO1 m RNA表达水平与常温对照组相比却显著下降(P<0.05)。结论冷应激前后大鼠的心脏、肝脏、脾脏、肺脏、肾脏、肌肉及十二指肠中ENO1基因mRNA的表达水平均发生显著改变,这表明冷刺激可通过影响动物机体ENO1基因m RNA的表达量调控各内脏器官的代谢燃料选择及其抗应激状态。
