目的:大电导钙激活钾通道(BKCa)是血管平滑肌细胞上的钾离子外流通道,负责调节血管的张力,本研究将探讨活血化瘀药物水蛭素是否可以通过激活BKCa通道来发挥血管舒张作用。方法:首先利用血管环实验观察水蛭素对血管的收缩作用,再通过脂质体转染法建立稳定表达BKCa通道的HEK293单克隆细胞系,研究其基本的生理学特性,同时建立BKCa通道动力学研究模型,并利用单通道膜片钳技术观察水蛭素对BKCa通道电流的影响。结果:血管环实验表明血管张力随着水蛭素浓度的增加而降低,在稳定转染BKCa通道的HEK293细胞上通过膜片钳记录到BKCa通道的电流,通过inside-out膜片钳记录,在140m M[K+]0/140m M[K+]i记录条件下观察I-V曲线,曲线通过0点反转电位为0 m V,HEK293细胞上转染BK通道的电导值为236.733.27 p S(n=5),同时观察到水蛭素可以激活BKCa通道。BKCa通道的开放概率分别从0.00683±0.0049增加到0.0159±0.0014(n=5,P<0.05)、0.0228±0.0016(n=5,P<0.05)、0.0469±0.0073(n=5,P<0.05)、0.0856±0.0103(n=5,P<0.01),在此基础上加入20 m M EGTA(钙离子螯合剂),该通道概率降低到0.0599±0.0070(n=5,P<0.01),HIR激活BKCa通道电流的作用仍存在,证实水蛭素对稳定转染的BKCa通道有浓度依赖性激活作用,且不依赖于钙离子。结论:本研究通过水蛭素对大鼠肠系膜血管收缩力影响的观察,说明水蛭素具有舒血管作用,这一作用很可能是通过对BKCa通道的激活来实现的。
