Probiotics are increasingly applied in popularity in both humans and *** of research has revealed their beneficial effects,including the immune modulation in intestinal pathogens ***—a cellular process that involves the delivery of cytoplasmic proteins and organelles to the lysosome for degradation and recirculation— is essential to protect cells against bacterial ***,the mechanism of probiotics-mediated autophagy and its role in the elimination of pathogens are still ***,we evaluated Bacillus amyloliquefaciens SC06(Ba)-induced autophagy and its antibacterial activity against Escherichia coli in RAW264.7 *** blotting and confocal laser scanning analysis showed that Ba stimulated autophagy in a dose-and time-dependent ***-induced autophagy was found to play a role in the elimination of intracellular bacteria when RAW264.7 cells were challenged with Escherichia coli(***).Ba induced autophagy by increasing the expression of Beclin1 and Atg5-Atg12-Atg16 complex,but not the AKT/mTOR signaling ***,Ba pretreatment attenuated the activation of JNK in RAW264.7 cells during *** infection,further indicating a protective role for probiotics via modulating macrophage *** above findings highlight a novel mechanism underlying the antibacterial activity of *** study enriches the current knowledge on probiotic-mediated autophagy,and provides a new perspective on the prevention of bacterial infection in intestine,which further the application of probiotics in food products.

Numerous studies have focused on the beneficial effects of probiotics in *** so,additional information should be obtained about the mechanisms by which a useful probiotic strain successfully exerts such beneficial *** this study,we evaluated the effect of the dietary supplementation of both live and disrupted Lactobacillus johnsonii(LJ) strain BS15 in broilers at different ***,growth performance,lipid metabolism,gut microbiota,intestinal development,and digestive ability of the broilers were assessed.A total of 180 one-day-old Cobb 500 chicks were randomly distributed into three *** chicks were fed diets supplemented with1×10～6 colony-forming units(cfu) LJ per gram of feed(LJ group);1×10～6 cfu disrupted LJ per gram of feed(D-LJ group);and de Man,Rogosa,and Sharpe liquid medium(control group),respectively,throughout a 42-day experimental *** results demonstrated that LJ supplementation of feed had a positive effect on the average daily gain and starter feed conversion *** addition,LJ supplementation of feed decreased serum triglyceride and low-density lipid cholesterol levels,as well as abdominal fat *** also reduced the mRNA levels of lipoprotein lipase in adipose tissue and stearoyl-CoA desaturase-1 in the *** diminished the mRNA quantities of the sterol regulatory element binding protein-1 c and fatty acid synthase,as well as increased the level of serum high-density lipid *** increased the mRNA quantities of peroxisome proliferator-activated receptor a,acyl CoA oxidase in the liver,and carnitine *** also improved the intestinal development and digestive ability mainly by increasing the villus height/crypt depth ratio in the *** probiotic increased the levels of epidermal growth factor and insulin-like growth factor-1,as well as the activities of trypsin and lipase in the jejunum and *** exerted beneficial effects on the intestinal ***,LJ ma

Captive breeding has been used as an effective approach to protect endangered animals but its effect on the gut microbiome and the conservation status of these species is largely *** giant panda is a symbol of *** integrated efforts including captive breeding,this species has been recently upgraded from "endangered" to "vulnerable".Since a large proportion(21.8%) of the population is captive,it is critical to understand how captivity changes the gut microbiome of these pandas and how such alterations to the microbiome might affect their future fitness and potential impact on the ecosystem after release into the *** we use 16 S rRNA marker gene sequencing and shotgun metagenomics sequencing to demonstrate that the fecal microbiomes differ substantially in wild and captive giant *** microbiome diversity was significantly lower in captive pandas,as was the diversity of functional ***,captive pandas had reduced functional potential for cellulose degradation but enriched metabolic pathways for starch metabolism,indicating a reduced fitness when releasing into the wild since their major diet in the wild will be ***,antibiotic resistance genes and virulence factors,as well as heavy metal tolerance genes were enriched in the microbiomes of captive pandas,which raises a huge concern of spreading these genes to other wild animals and the ecosystem after releasing them to the wild *** results clearly show that captivity has altered the giant panda microbiome,which could have unintended negative consequences on their fitness and the ecosystem during the re-introduction of giant pandas into the wild.

Along with banning of antibiotics,necrotic enteritis(NE),especially subclinical NE(SNE) whereby no clinical signs are present in chicks,has become one of the most threatening problems in poultry ***,increasing attention has been focused on research and application of effective probiotic strains,as an alternative to antibiotics,to prevent SNE in *** the present study,we evaluated the effects of Lactobacillus johnsonii BS15 on prevention of SNE in ***,assessment determined growth performance and indexes related to intestinal mucosal immunity in the ileum and cecal tonsil of broilers.A total of 300 one-day-old Cobb 500 chicks were randomly distributed into the following five groups:control group(fed with basal diet + de Man,Rogosa,and Sharpe liquid medium[normal diet]),SNE group(normal diet),BS15 group(basal diet + 1 × 10～6 colony-forming units BS15/g as fed[BS15 diet]),treatment group(normal diet[days 1-28]+ BS15 diet[days 29-42]),and prevention group(BS15 diet[days 1-28]+ normal diet[days 29-42]) throughout a 42-day experimental *** infection was treated for all chicks in the SNE,BS15,treatment,and prevention *** present results demonstrated that BS15 supplementation of feeds in BS15 and prevention groups exerted a positive effect on preventing negative influences on growth performance;these negative influences included low body weight gain and increased feed conversion ratio caused by *** no changes were detected in all determined indexes in cecal tonsils,BS15-treated broilers were free from SNE-caused damage in villi in the ***15 inhibited SNE-caused decrease in immunoglobulins in the *** the lamina propria of ileum,T-cell subsets of lymphocytes influenced by SNE were also controlled by ***15 affected antioxidant abilities of the ileum and controlled SNE-induced mitochondrion-mediated apoptosis by positively changing contents and/or mRNA expression levels of apoptosis-related ***

Background:Subclinical necrotic enteritis(SNE) widely outbreaks in chickens which inflicted growth-slowing,causing enormous social and economic *** better understand the molecular underpinnings of SNE on lipid metabolism and explore novel preventative strategies against SNE,we studied the regulatory mechanism of a potential probiotic,Lactobacillus johnsonii BS15 on the lipid metabolism pathways involved in chickens with ***:180 one-day-old chickens were randomly divided into three groups and arranged with basal diet(control and SNE group).Added with BS15(1×10～6 cfu/g) or Man Rogosa Sharpe(MRS) liquid medium for 28 *** hepatic gene expression of each group was then measured using high-throughput analysis methods(RNA-Seq).Quantitative real-time PCR(qRT-PCR) was used to detect the expression changes of the related ***:The results showed that there are eleven lipid metabolic pathways were found during the prevention of BS15 treatment in SNE chickens by RNA-Seq,including the peroxisome proliferator-activated receptor(PPAR) signaling pathway and arachidonic acid ***15 notably facilitated the expressions of fatty acid binding protein 2(FABP2),acyl-CoA synthetase bubblegum family member 1(ACSBG1),perilipin 1(PLIN1) and perilipin 2(PLIN2),which were involved in PPAR signaling pathway of SNE ***,suppression of phospholipase A2 group IVA(PLA2 G4 A) in arachidonic acid metabolism was observed in SNE chickens after BS15 *** expression patterns of FABP2,ACSBG1,PLIN1,PLIN2 and PLA24 G in qRT-PCR validation were consistent with RNA-Seq ***:These findings indicate that SNE may affect the hepatic lipid metabolism of ***,BS15 pretreatment may provide a prospective natural prophylaxis strategy against SNE through improving the PPAR signaling pathway and arachidonic acid metabolism.

植物微生态制剂的研发依据植物微生态学理论,以获得国家科技进步二等奖"芽孢杆菌生物杀菌剂的研制与应用"和中华农业科技一等奖"植物微生态制剂的研制与应用"的核心技术为指导,从生态学的角度,整合了微生物组学和生态学理念,综合考虑植物及其体内体表的微生物的相互关系,选用作物共生内生的促生防病芽胞杆菌、木霉菌等高效菌株,运用现代微生物发酵技术加工制备而成。通过菌株在作物根表、根际和体内定植、繁殖和转移,充分发挥菌株微生态调控功能,不仅抑制病原微生物,还有效提高了植物微生态种群的多样性和丰富度,增强了植物的抗逆能力和品质。目前植物微生态制剂已经在大田作物、蔬菜、果树、中药材等农作物上广泛应用。植物微生态制剂产品在大田作物上的应用主要集中在内蒙古、河北、甘肃、湖北等地。在湖北省宜昌市长阳县玉米试验基地施用植物微生态制剂,可以有效促进苗期玉米植株的生长,玉米开花早;在水稻上施用植物微生态制剂可以有效防控水稻叶瘟病;在内蒙古武川县、固阳县、多伦县、商都县及乌兰察布前旗地区,河北省张家口,甘肃山丹县等地在马铃薯上进行试验示范,植物微生态制剂对马铃薯疮痂病、粉痂病、黑痣病等土传病害防效达60%以上,马铃薯商品率增加了10%以上,产量增加20%,增产防病效果显著。植物微生态制剂在白菜、甘蓝、菜花、萝卜、洋葱、芹菜、茼蒿、白菜、生菜、大蒜、西红柿、茄子等多种蔬菜作物上均有应用,应用地区包括北京、内蒙、福建、河南、山东等蔬菜种植大省。西红柿施用植物微生态制剂后,死棵烂秧现象减少,株高增加,长势良好,果实光滑、整齐、商品率高,发病率低于20%,增产幅度介于15-36%;茄子施用植物微生态制剂后株高增加,叶片增大,促生效果明显,产量提高,土传病害减轻;辣椒施用植物微生态制剂后,株高增加,叶色深绿,长势旺盛,死棵烂秧减少,促生效果十分明显,枯黄萎等重茬病害发病率低于15%,增产20-28%;甘蓝施用植物微生态制剂10天后,平均株高增加3.7厘米,叶面积增大,植株开展度增加5.0厘米,叶色浓绿,叶质脆嫩,提早成熟,促生效果显著,亩增产30%以上;内蒙古自治区巴彦淖尔市杭锦后旗、新疆昌吉、山东聊城莘县等地西葫芦示范地块使用植物微生态制剂后死棵烂秧、枯黄萎病等明显减轻,发病率低于20%,增产达到15-36%。植物微生态制剂在葡萄、草莓、柑橘、猕猴桃等多种果树上均有应用,应用地区包括北京、河北、湖北、广西、四川等果树种植大省。在北京市延庆区和昌平区的草莓试验基地施用植物微生态制剂,草莓平均株高增加5厘米以上,叶片数增加4个,死苗率减少20%,单果重增加7.3克,发病率降低70%以上;在广西柳州、湖北宜昌市夷陵区和四川省成都市蒲江县柑橘试验地块施用植物微生态制剂,柑橘根系白根多,没有线虫,土壤疏松,叶面肥厚油绿,柑橘黄化病防治效果显著;河北昌黎县酿酒葡萄遭冰雹和连阴雨导致严重烂果、掉果现象,施用植物微生态制剂后抗逆效果明显,处理区比对照区产量增加15%,糖度增加21%;在猕猴桃上施用植物微生态制剂,处理区果实采摘后叶片仍然深绿色、厚,猕猴桃水份保留充足,商品率显著提高。植物微生态制剂产品在中药材上的应用主要集中在甘肃、吉林、内蒙古等地,应用作物包括罂粟、人参、三七、当归、当参等。山东威海文登地区西洋参由于重茬连作土传病害突出,施用植物微生态制剂后发病率控制在20%以下,增产15-36%;在吉林长白县人参施用植物微生态制剂,人参水根发达,单参重量增加,形状好;甘肃岷县应用植物微生态制剂预防当归麻口病效果显著,党参施用植物微生态制剂五个月后,处理区域长势较好,死棵现象大大减少。使用植物微生态制剂达到预防土传病害、解决连作障碍及改良土壤的作用效果。且环境兼容性好、对人类安全,有利于农业的可持续发展,是实现植物病害绿色防治和作物科学种植的最佳选择。

引言近年来,已有许多研究注意到肠道菌群在机体免疫、营养代谢及疾病的发生发展中的变化及作用,而关于寄生虫感染过程中肠道菌群变化相关研究相对滞后。本研究初步阐明旋毛虫感染小鼠肠道菌群变化与免疫应答关系,旨在为肠道寄生线虫病的防治及肠道炎性疾病生物学疗法提供新的思路。材料与方法本研究将24只雌性SPF级BALB/c小鼠平均分为两组（旋毛虫感染组、对照组）,每组12只。旋毛虫感染组小鼠灌服旋毛虫肌幼虫300条/只,对照组小鼠灌服等体积PBS。分别于感染后7天、14天、21天和28天无菌条件下采集小鼠新鲜粪便。提取粪便细菌基因组总DNA,然后对总DNA进行16S rRNA基因序列V1-V3高变区PCR扩增产物进行定量、混匀,Illumina高通量测序。流式细胞术检测小鼠感染旋毛虫感染不同时期的肠系膜淋巴结（MLN）Foxp3Treg数量的变化情况、Th2型细胞因子IL-10及Th17型细胞因子IL-17表达量的变化情况;并采用间接ELISA法检测小鼠感染旋毛虫不同时期血清IFN-γ、IL-4、IL-22、IL-17表达量变化情况。结果与讨论测序分析结果表明旋毛虫感染7、14、21、28天时小鼠肠道菌群OTU均高于对照组,所有序列属于以下四个菌门:拟杆菌门（Bacteroidetes）、硬壁菌门（Firmicutes）、变形菌门（Proteobacteria）和脱铁杆菌门（Deferribacteres）,所占比例达99%以上。感染7天、14天、21天、28天时实验组小鼠肠道拟杆菌门细菌数量均高于对照组,且7天时两组差异极显著（P<0.01）;感染7、14、21天时实验组小鼠肠道硬壁菌门细菌数量均低于对照组,且7天时两组差异极显著（P<0.01）;感染28天时硬壁菌门细菌数量高于对照组,差异不显著（P>0.05）。感染7天、14天、21天、28天时小鼠肠道变形杆菌门细菌数量均低于对照组,差异显著（P<0.05）。感染7天时小鼠肠道脱铁杆菌门细菌数量高于对照组,差异显著（P<0.05）;感染14天、21天、28天时实验组小鼠肠道脱铁杆菌门细菌数量均低于对照组,其中感染28天时两组差异显著（P<0.05）。流式结果显示在旋毛虫感染过程中Th2型细胞因子分泌量始终高于对照组,呈上升趋势;而Th1型细胞因子分泌量始终低于对照组,呈下降趋势;Treg数量始终高于对照组,也呈上升趋势;Th17型细胞因子在旋毛虫感染前期分泌量高于对照组,感染后期基本与对照组持平。Thl型、Th2型、Th17型细胞因子水平随着旋毛虫感染时间的延长,变化趋势逐渐趋于平缓。因此可以推断在旋毛虫感染过程中肠道菌群的变化对小鼠免疫应答起到了促进作用。肠道菌群与各种细胞因子协同调节肠道,使其尽快恢复到相对稳定状态,以达到肠道菌群和旋毛虫的共生。

Bamboo-eating giant panda(Ailuropoda melanoleuca) is an enigmatic species,which possesses a carnivore-like short and simple gastrointestinal tract(GIT).Despite the remarkable studies on giant panda,its diet adaptability status continues to be a matter of *** resolve this puzzle,we investigated the functional potential of the giant panda gut microbiome using shotgun metagenomic sequencing of fecal *** also compared our data with similar data from other animal species representing herbivores,carnivores,and omnivores from current and earlier *** found that the giant panda hosts a bear-like gut microbiota distinct from those of herbivores indicated by the metabolic potential of the microbiome in the gut of giant pandas and other ***,the relative abundance of genes involved in cellulose-and hemicellulose-digestion,and enrichment of enzymes associated with pathways of amino acid degradation and biosynthetic reactions in giant pandas echoed a carnivore-like *** significantly,the enzyme assay of the giant panda's feces indicated the lowest cellulase and xylanase activity among major herbivores,shown by an in-vitro experimental assay of enzyme activity for cellulose and *** of our results consistently indicate that the giant panda is not specialized to digest cellulose and hemicellulose from its bamboo diet,making the giant panda a good mammalian model to' study the unusual link between the gut microbiome and *** increased food intake of the giant pandas might be a strategy to compensate for the gut microbiome functions,highlighting a strong need of conservation of the native bamboo forest both in high-and low-altitude ranges to meet the great demand of bamboo diet of giant pandas.

In this work,we searched for an effective probiotic that can help control intestinal infection,particularly enterotoxigenic Escherichia coli K88（ETEC） invasion,in giant panda（Ailumpoda melanoleuca）.As a potential probiotic strain,Lactobacillus plantarum BSGP201683（*** G83）was isolated from the feces of giant panda and proven beneficial in *** study was aimed to evaluate the protective effect of *** G83 in mice challenged with *** mice were orally administered with 0.2 mL of PBS containing *** G83 at 0 colony-forming units（cfu） mL（control;negative control,ETEC group）,5.0 xl08 cfu mL（LDLP）,5.0×10～9 cfu mL（MDLP）,and5.0×10 cfu mL（HDLP） for 14 consecutive *** day 15,the mice（LDLP,MDLP,HDLP,and ETEC groups） were challenged with ETEC and assessed at 0,24,and 144 *** health status;chemical and biological intestinal barriers;and body weight were *** showed that *** G83 supplementation protected the mouse gut mainly by attenuating inflammation and improving the gut *** indices significantly changed at 24 h after challenge compared to those at 0 and 144 *** treatment groups showed inhibited plasma diamine oxidase activity and D-lactate ***-junction protein expression was down-regulated,and interleukin（IL）-1β,IL-6,IL-8,TLR4,and MyD88 levels were up-regulated in the jejunum in the LDLP and MDLP *** number of the Enterobacteriaceae family and the heat-labile enterotoxin（LT） gene decreased（P <0.05） in the colons in the LDLP and MDLP *** data indicated that *** G83 could attenuate acute intestinal inflammation caused by ETEC infection,and the low and intermediate doses were superior to the high *** findings suggested that *** G83 may serve as a protective probiotic for intestinal disease and merits further investigation.
