The technology of genome shuffiing has been presented as a novel whole genome engineering approach for the rapid improvement of cellular phenotypes without knowing its genetic *** this study,we used genome shuffiing in an attempt to improve propionate production and acid-tolerance of the wild-type strain *** *** initial mutant population of M. elsdenii H6 were treated with UV irradiation and NTG,resulting in an improved population producing more propionate than the original strain at pH *** mutants were used as the starting strains for 3 rounds of genome *** each round,the pH of the medium plates used for selection was decreased by *** improved strains were selected based on their acid tolerance and propionate *** libraries were established after each round and these improved mutant served as the starting pool for the next round of *** acid tolerance of the mutants on the gradient plate increased progressively with each round of shuffiing up to 3 *** mutants after three rounds of genome shuffiing exhibited an improved acid tolerance and propionate production,which could grow at pH 5.0 and the yield of propionate was increased 41.5±1.4%compared with the original strain after fermented for *** mutant F32 of *** H6 with the characterization of the high propionate production and acid tolerance could be potential candidates for preventing rumen acidosis and production disease during the transition period of dairy cows.

Medial hypertrophy of pulmonary arterioles during pulmonary arterial hypertension(PAH) in humans is associated with enhanced proliferation of smooth muscle cells(SMCs).Elevated matrix metalloproteinase (MMP)-2 has been found in pulmonary artery SMCs(PA-SMCs) in patients with idiopathic PAH,leading to the hypothesis that MMP-2 contributes to the proliferation and migration of SMCs in the progression of *** models of PAH are thus needed to further investigate the role of MMP-2 in human *** growing meat-type chickens(broilers) provide a model of spontaneous PAH caused by excessive pulmonary blood flow. The present study was conducted to determine whether the MMP-2 is involved in PA-SMCs proliferation and the medial hypertrophy in this model. Cultured PA-SMCs from normal birds were used to evaluate the effect of MMPs on cell *** zymography showed that the endothelin(ET)-l-induced proliferation of PA-SMCs was concomitant with increased pro and active MMP-2 *** transcription PCR demonstrated an up-regulation of MMP-2 mRNA. However,PA-SMC proliferation was inhibited by MMP inhibitors doxycycline and *** vivo experiments revealed a significant increase of MMP-2 expression(P < 0.01) in the hypertrophied pulmonary arterioles of PAH broilers,which was positively correlated to the wall thickness and medial *** context,this study suggests that the MMP-2 might contribute to the medial hypertrophy in pulmonary arterioles during PAH by enhancing the proliferation of vascular smooth muscle cells. This model of PAH may be valuable for researches focused on targeting the MMPs to prevent and reverse human PAH.

BACKGROUND:Human health may be improved if milk with a favorable fatty acid composition and Se concentration is ingested. The present study is to determine howa basal diet supplemented with daily 5 mg Se as Se-enriched yeast(SY) or sodium seienite (SS) affects the fatty acid composition and Se concentration of bovine *** effects of Se form on blood Se concentration, erythrocyte glutathione peroxidase 1(GPxl) activity,serum GPx3 activity and milk yield and component were also studied. RESULTS:Both Se forms,when compared to control group,increased Se concentrations of blood(P < 0.01) and milk(P < 0.01), erythrocyte GPx1 activity(P < 0.05) and milk percentages of polyunsaturated fatty acids(PUFA)(P < 0.05) and c/'s-9,cis-12 linoleicacid(P < 0.05).Cows supplemented with SY had higher Se levels in blood(P < 0.01) and milk(P < 0.01) and percentage of PUFA in milk(P < 0.05) when compared with those supplemented with *** yield,milk component and serum GPx3 activity were not significantly affected by Se form. CONCLUSION:Supplementation of diet with SY appears to be of more benefit than SS in producing favorable milk with high PUFA and Se concentrations.

Zearalenone(ZEN) is an estrogenic mycotoxin produced by several Fusarium species,which can contaminate food and *** compounds elicit a wide spectrum of toxic effects,including the capacity to alter normal immune *** this study,the in vitro effects of the treatment of ConA-stimulated splenic lymphocytes with ZEN(0-25μg/mL) were *** modulates the expression of IL-2,IL-6,and IFN-γ. The IL-2 levels were up to four fold higher(P<0.05) compared with the levels in the control levels at toxin concentrations of 25μg/mL after 48 h of *** IL-6 levels were critically suppressed at this concentration;these changes were very statistically significant(P<0.05).At lower ZEN concentrations(0.1.0.4 and 1.6μg/mL), the IFN-γlevels changed slightly;however at 6.25 and 25μg/mL,the IFN-γresults reached statistical significance compared with the control levels(P<0.05).These data suggest that ZEN has potent effects on the expression of chicken splenic lymphocytes cytokines at the mRNA level.

The immunotoxicity of deoxynivalenol(DON) as well as the induction of cytokines and related genes in the splenic lymphocytes of chicken were *** the present study,the in vitro effects of DON(0-50μg/mL) treatment on ConA-stimulated splenic lymphocytes were examined after 48 *** modulates the expression of IL-2,IL-6,and IFN-γ.The IL-2 mRNA levels at 0.2.3.2,12.5,and 50μg/mL DON were significantly higher(P < 0.05) than the control group(ConA-only stimulated cells group).The highest IL-2 mRNA level was reached at 12.5μg/mL *** IL-6 levels were critically suppressed at all concentrations,and the differences from the control group(P < 0.05) were statistically ***-6 expression was highest at 12.5μg/mL *** IFN-γlevels were significantly higher(P < 0.05) than the control group,and IFN-γexpression was lowest at 0.8μg/mL *** data indicate the potent effects of DON on cytokine mRNA expression in chicken splenic lymphocytes.

Major histocompatibility complex takes the capital contribution to maternal immune responses to the semiallogenic fetus. As the menber of *** machenism of class I chain-related genes 2(MIC2) on pregnant setup and delivery,till remains to be *** evaluate the possible effect of MIC2 on pregnant establishment and delivery,neonatal ear tissues,fetal placenta, peripheral blood Polymorphonuclear Neutrophils(PMN) of dairy cow in different pregnant stages were collected to analyze MIC2 expression by quantitative ***2 on PMN in dairy cows was downregulated expressed in first and second trimesters and recoveried back to normal level in ***,MIC2 was novel upregulated expressed in neonatal ear tissues and downreguJated expressed in fetal *** results suggested that MIC2 lowly expressed in the first and second trimester of pregnant cows could suppress NKG2D binding MIC2 on PMN and therefore,let embryo escape from maternal immune response. Anabiotic MIC2 expression in peripartum suggested that maternal immune to full grown foetus recover back to normal level which be in favour of *** foetus,novel upregulated expression of MIC2 in tissues was favourible for foetus to escape its ***,downregulated expression in fetal placenta let placenta decoherenced from maternal placentome which was beneficial to delivery.

In this study,lactic acid bacteria in canine feces were isolated and identified,and their oxalate-degrading capacities were *** oxalate-degrading capacities were determined for 24 of 47 (51.06%) lactic acid bacteria *** these,8 isolates[Leuconostoc mesenteroides(RL75),Lactococcus garvieae(CD2),Lactococcus ***(CS21),Enterococcus faecium(CL71 and CL72),and Enterococcus faecalis(CD14,CS62,and CD12)]degraded more than 5%of the oxalate present,while the others degraded less than 5%of the oxalate in *** that degraded more than 5%of the oxalate present were selected for further *** oxalate-degrading capacities of individual isolates,a mixture of Enterococcus,a mixture of Lactococcus,and a mixture of the eight isolates were evaluated in media containing different concentrations of glucose(sufficient,insufficient,or no glucose).In comparison with the control medium,all of the individual isolates and mixtures of isolates could degrade oxalate in all three groups(P < 0.05).In most cases,the isolates growing in medium with 20 g/L of glucose had higher oxalate-degrading capacities than those growing in medium with 2.5 g/L of glucose or no *** mixture of all isolates showed higher oxalate-degrading capacity than the individual isolates and other *** oxalate-degrading capacities of the isolates were isolate dependent.

A 35-d experiment was conducted to evaluate the effect of selenium-enriched probiotics(SP) on laying performance,egg quality,egg selenium(Se) content,and egg glutathione peroxidase(GPX) *** hundred 58-wk-old Rohman laying hens were randomly allotted to 5 dietary treatments of 100 *** treatment had 5 replicates and each replicate had 5 cages with 4 hens per *** SP was supplemented to a corn-soybean-meal basal diet at 3 different levels that supplied total Se at 0.2,0.5,and 1.0 mg/*** basal diet served as a blank control, while the basal diet with supplemental probiotics served as a probiotics *** results showed that dietary SP supplementation not only increased(p < 0.05) the rate of egg laying,day egg weight,mean egg weight,egg Se content,and egg GPX activity,but also decreased(p < 0.05) the feed:egg ratio and egg cholesterol *** egg Se content was gradually increased(p < 0.05) along with the increasing level of dietary *** SP supplementation also slowed down(p < 0.05) the drop of Haugh units(HU) of eggs stored at room *** egg GPX activity had a positive correlation(p < 0.01) with egg Se content,and a negative correlation(p < 0.01) with egg HU drop. These results suggested that Se contents,GPX activity,and HU of eggs were affected by the dietary Se level,whereas the egg laying performance and egg cholesterol content were affected by the dietary *** was concluded that this SP is an effective feed additive that combines the organic Se benefit for hen and human health with the probiotics benefit for laying hen production *** was also suggested that the eggs from hens fed this SP can serve as a nutraceutical food with high Se and low cholesterol contents for both healthy people and patients with hyperlipidemia,fatty liver,or cardiovascular disease.

Dairy cows are highly susceptible to ketosis after parturitioa In the present study,we evaluated the expression of fatty acidβ-oxidation-related enzymes in the liver of ketotic cows(n=6) and non-ketotic cows(n=6). Serum levels of nonesterified fatty acids(NEFA),β-hydroxybutyrate(BHBA),and glucose were determined by using standard biochemical *** mRNA abundance and protein content of acyl-CoA synthetase long-chain(ACSL),carnitine palmitoyltransferaseⅠ(CPTⅠ),carnitine palmitoyltransferaseⅡ(CPTⅡ),acyl-CoA dehydrogenase long chain(ACADL),3-hydroxy-3-methylglutaryl-CoA synthase(HMGCS),and acetyl-CoA carboxylase(ACC) were evaluated by real-time polymerase chain reaction and enzyme-linked immunosorbent *** found that serum glucose levels were lower in ketotic cows than those in non-ketotic cows,but serum BHBA and NEFA concentrations were *** mRNA and protein levels were significantly higher in livers of ketotic cows than those in non-ketotic *** contrast,the mRNA levels of CPT I and both mRNA and protein levels of CPT II,ACADL,HMGCS,and ACC were decreased in the liver of ketotic *** NEFA concentration positively correlated with ACSL protein levels and negatively correlated with protein levels of CPT II,HMGCS,ACADL,and *** addition,serum BHBA concentration negatively correlated with protein levels of CPT II,HMGCS,and ***,fatty acidβ-oxidation capability was altered in the liver of ketotic compared to non-ketonic ***,high serum NEFA or/and BHBA concentrations play key roles in affecting pathways of fatty acid metabolism in the liver.
