Schistosomes infect around 280 million people *** worms survive in the veins of the final host, where thioredoxin glutathione reductase (TGR) activity helps the parasites to survive in the aerobic *** the present study,we synthesized two small interfering RNAs (siRNA S 1 and S2) targeting the Schistosomajaponicum (Sj) TGR gene, and used these to silence the TGR *** effects of the siRNAs were evaluated in vitro and in mice infected with *** effects of the siRNAson the thioredoxin reductase (TrxR) activity of SjTGR were also *** effects oftransfection with the siRNAs via the soaking method in vitro were confirmed by flow cytometry.S2 siRNA at a final concentration of 200 nM partially inhibited the exPression of SjTGR at both the transcript and protein levels in *** vivo,S2 siRNA reduced the incidence of worms in schistosomE-infected mice and down:regulated SjTGR transcript ***:activity was lower in worms in the S2 siRNA:treated group compared with the control *** analysis revealed that purified recombinant SjTGR could remove oxygen free radicals but not H2O2 directly, which may help to explain the effects of RNA *** results of this preliminary study indicate that SjTGR may play an important role in the clearance of oxygen free radicals and protection of S.j aponicum parasites against oxidative damage.

The distribution of genetic diversity ofToxoplasma gondii in wildlife is of interest to understand the transmission of this parasite in the *** information on *** genotypes has been reported in wildlife in *** PCR and multilocous polymerase chain reaction:restriction fragment length polymo rphism (PCR:RFLP) technology,we conducted a survey to examine genetic diversity of this parasite in wildlife on the Qinghai:Tibet Plateau,*** total,183 brain tissues of different wild animals, including 48 Qinghai vole (Microtus fuscus),101 Plateau Pika (Ochotona curzoniae) and 34 Tibetan Ground:tit (Pseudopodoces hum ilis),were tested for *** infection.11 of which were found to be positive for the *** B 1 gene by PCR *** positive DNA samples were typed at 10 genetic markers, including 9 nuclear loci (SAG 1,5':and 3':SAG2,alternative SAG2,BTUB,GRA6,L358,PK1,c22:8,c29:2),and an apicoplast locus *** were successfully genotyped at eight or more genetic loci, and were grouped to three distinct *** ur samples belonged to ToxoDB Genotype #10 and the other two samples were identified as two new genotypes (http://***/toxo/).The results show that there is a potential risk for the transmission of this parasite thro ugh the wildlife in this *** our knowledge, this is the ?rst report of genetic typing of *** isolates in wildlife on the Qinghai:Tibet Plateau,China.
