试验于2013年四月中旬进行,试验选用饲养在黑龙江省大庆市大同区北方种鹅场8~10月龄的20只纯种籽鹅(Zie Goose)。采用背腹式按摩采精法进行个体采精,采出的精液一部分进行常规的精液质量分析,另外一部分精液进行4℃保存,1h内送到黑龙江八一农垦大学动物遗传育种实验室,利用南宁松景天伦生物科技有限公司生产的彩色精子质量检测系统进行计算机辅助精子质量分析(CASA)。常规精液质量分析测定的指标有:精液量、精子密度、精子活力和正常形态活精子数。利用集精杯来测定个体采精量;采用血球计数板测量精子密度;采用Hanging Drop方法测量精子活力(每个样本重复检测三次);精子形态采用Bakst and Ceci方法,将精子在伊红-苯胺黑中的抹片在1000倍显微镜下观测,
