目的:卵泡的发育和闭锁是卵泡发生过程中的两个重要生理现象,对维持卵巢正常的生理功能起到至关重要的作用。卵泡的生长发育包括卵泡中颗粒细胞的增殖、分化和周期控制。而卵泡闭锁则起始于颗粒细胞的凋亡,颗粒细胞的凋亡最终导致卵泡的闭锁。同时,颗粒细胞产生的类固醇激素等,对卵泡的生长发育也起着重要作用。而现阶段越来越多的研究结果表明内质网应激在卵泡生长发育闭锁过程中有一定的作用,但具体机制还不很清楚。X盒结合蛋白(XBP1)是非折叠蛋白应答途径(unfolded protein response,UPR)信号途径的重要调节因子,广泛参与细胞分化、增殖、凋亡等多种信号途径。研究XBP1对小鼠颗粒细胞凋亡、周期及类固醇激素分泌作用具有重要意义。材料与方法:本研究对小鼠卵巢颗粒细胞在体外进行了分离培养,通过免疫组化、RNA干扰、western blot、荧光定量PCR、ELISA、流式细胞术和免疫荧光等技术对XBP1在小鼠颗粒细胞凋亡、周期及类固醇激素分泌的作用进行了研究。结果:本部分实验成功构建了shRNA慢病毒干扰载体,并且筛选出最佳干扰质粒,为了鉴定其干扰效果,小鼠颗粒细胞经慢病毒转导后,测定其干扰效率在80%左右。结果显示XBP1干扰后颗粒细胞雌激素的分泌显著下降。通过检测相关基因的表达发现,芳香化酶基因Cyp19a1表达显著降低。同时,XBP1干扰后通过抑制cyclinA1、cyclin B1的表达阻滞细胞周期。但是,在正常体外培养条件下,细胞凋亡率没有显著变化。结论:本研究的结果表明XBP1通过对颗粒细胞的生长及相关基因的调控而影响颗粒细胞雌激素的分泌,阻滞细胞周期,对小鼠颗粒细胞起到重要作用,为进一步研究卵泡发育及排卵提供了理论依据。
