Stimulus specific adaptation (SSA) is widely demonstrated in many brain regions of human and intact animals from multiple spatial scales including extracellular units recording and *** models are proposed to explain that repetitive stimulus induces adaption of physiological responses to favor deviance stimulus detection in the network *** the present study, we would examine the SSA at the synapse level with whole cell patch clamp recording from the cultured cortical *** cultured pyramidal neurons shared the similar SSA properties in response to the odd-ball stimulation paradigm as those reported with in vivo *** different stimuli were from electrical stimulations that were spatially *** cultural neurons showed SSA to the electrical *** SSA was not attributable to a reduction of the intrinsic membrane properties in that cultured neurons faithfully followed the intracellular current injectionup to 200 *** AMPA receptors mediated excitatory postsynaptic currents (EPSC) also displayed stronger SSA compared with the GABA receptors mediated inhibitory postsynaptic currents (IPSC).The different dynamic of short term synaptic depression characterized by EPSC and IPSC might account for the generation of SSA at the supra-threshold spike *** findings support that the synapse soecific fatigue would be the candidate neural mechanism for the stimulus specific adaptation.

In the rabbit and mouse retina, some ON-OFF direction selective ganglion cells (DSGCs) showed tracer coupling when injected with neurobiotin, and a majority of morphological ON-OFF DSGCs showed coupling in the early postnatal rabbit *** is not clear whether the coupled ON-OFF DSGCs belong to the same subtype, if they do, what preferred directions they exhibit? And whether the coupling in early development is heterologous and is gradually becoming homologous? In this study, we showed that after postnatal day 15 (P15), all coupled ON-OFF DSGCs exhibited preferred directions to *** P13, the ON-OFF DSGCs with preferred directions to posterior also showed ***, every ON-OFF DSGC in all four subtypes exhibited coupling at *** results illustrated a rapid decoupling process around eye *** a transgenic mouse line with GFP exclusively expressed in the posterior subtype of ON-OFF DSGCs, we demonstrated that ON-OFF DSGCs with the preferred directions to anteriorand posterior are coupled together at P12, forming a "metapopulation"separated from the other two *** results suggest that early in development there exist subtype-specific dendro-dendrite/soma interactions among the four classes of mouse ON-OFF *** mechanism of how different subtypes of juvenile ON-OFF DSGCs recognize each other still needs investigation.

By sustained activation of opioid receptors, chronic opioids cause analgesic tolerance, physical dependence, and opioid addiction, for which an effective treatment is still ***, as a part of limbic system, plays an important role in opioid *** this study, we using patch clamp recording method combined with pharmacological approaches investigated chronic morphine-induced adaptations in glutamatergic synaptic transmission in neurons of the basolateral amygdala (BLA).Chronic morphine significantly increased glutamate synaptic transmission in pyramidal neurons in BLA containingμ-opioid *** naive rats, DAMGO,μ-opioid receptor's activator, could inhibit the release ofpresynaptic glutamate, while DAMGO would lost this inhibitory effect in chronic morphine *** found that in saline rats, DAMGO's effect was blocked by AACOCF3, an inhibitor of phospholipase A2 and 4-AP, which blocks the presynaptic voltage-dependent potassium channels regulated by metabolites of the PLA2 *** PKA didn't play any roles in na(i)ye ***, we confirmed that in chronic morphine group, PLA2-AA-Kv pathway was downregulated and PKA was upregulated, which were involved in the effect of DAMGO on presynaptic glutamate *** job provided the new mechanism of opioid receptors in morphine treatment for further clinical perspective.

Objective Irritable bowel syndrome (IBS) is a common gastrointestinal disorder characterized by abdominal pain and bloating in association with altered bowel ***, its pathogenesis and the underlying molecular mechanisms of visceral hyperalgesia remain *** therefore investigated whether adrenergic signaling pathway is involved in visceral hyperalgesia at adult induced by neonatal colonic inflammation (NCI).Methods Visceral hypersensitivity was induced by colonic injection of 0.5% acetic acid (AA, 0.2 mL) in 10-day-old *** response to colorectal distension (CRD) was measured by visual observation of abdominal withdrawal reflex (AWR) by a blinded *** NCI significantly increased the visceromoter response to CRD, which started 6 weeks and returned to normal level 12 weeks after neonatal colonic infusion of *** administration of nonselective β-adrenergic receptor antagonist (propranolol) attenuated NCI-induced visceral hypersensitivity in a dose-dependent manner while α-adrenergic receptor antagonist (phentolamine) did not produce any effect in AA-treated *** of propranolol did not produce any effect on AWR scores in healthy rats, indicating that this agent did not act as a non-specific analgesic and that adrenergic system did not normally participate in the responses to *** These data suggest that NCI-induced visceral hypersensitivity is mediated, at least in a large part, by β-adrenergic receptors, thus identifying a specific neurobiological target for the treatment of chronic visceral pain.

Brain malformations and retinal dysplasia in a subset of congenital muscular dystrophies (CMDs), dystroglycanopathies, are caused by disruptions of the basement *** extracellular matrix binding by the hypoglycosylated α-dystroglycan because of mutations in several glycosyltransferases is the underlying molecular *** is not known how the basement membrane becomes *** hypothesized that assembly of the basement membrane is reduced in rate thus causing a physically compromised basement *** test this hypothesis, we analyzed assembly of extracellular matrix on cultured neural stem cells and physical properties of the mutant basement membrane by atomic force *** assembly on the protein O-mannose β1,2-N-acetylglucosaminyltransferase 1 (POMGnT1) knockout neural spheres was reduced when compared to the *** incubated with Matrigel, extracellular matrix (ECM) molecules including all 4 major components of the basement membrane, laminin, collagen Ⅳ, perlecan, and nidogen *** of ECM aggregation was reduced on POMGnT1 knockout neural sphere as revealed by slower growth in aggregate size when compared to the *** staining and proteomic comparison revealed that the mutant basement membrane exhibited compositional changes from the *** force microscopic analysis revealed that the mutant basement membrane had reduced elastic modulus with surface topography showing bigger valleys than the ***, disruptions of the basement membrane in dystroglycanopathies may be caused by its weakened strength resulting from biochemical changes in composition and physical changes in structure and reduced assembly rate.

Objective Changes ofneurotrophin-3 (NT-3) expression in rat red nucleus following complete spinal cord transection have been studied in order to provide the experimental evidences for the mechanisms of regeneration and repair after spinal cord injury and for neurotrophins to treat spinal cord *** Male Sprague-Dawley rats weighing 240-260 g were divided into three groups at random: (1) Spinal cord transection group which was randomly subdivided into four subgroups according to the survival time at the day 1, 3, 7 and 14 d after operation (n=6 for each time point), in which the spinal cord of rat was transected completely at thoracic level 10-11.(2) Sham operation group (laminectomy only, n=6).(3) Normal control group (n=6).The expression of NT-3 in red nucleus was examined by immunohistochemistry and image *** NT-3 immunoreactive cells were found in red nucleus in both control and experimental *** operation, the expression of NT-3 increased gradually and reached the peak at the day 7 in red *** data reveal the profile of NT-3 expression in red nucleus after complete spinal cord transection at thoracic *** The results suggest that the demand of NT-3 increases in red nucleus after spinal cord injury and the increase of endogenous NT-3 may be beneficial for the survival and regeneration of the injured rubrospinal neurons.
