The processing of amyloid precursor protein(APP) is a key event in the pathogenesis of Alzheimer disease. Certain cleavage pathways tend to occur in particular subcellular compartments when APP travels *** a result,the processing of APP is greatly influenced by factors which regulate its *** we report that SCG10, a membrane-associated protein,directly interacts with the KFFEQ motif of APP intracellular domain(AICD) and promotes the non-amyloidogenic processing of *** of SCG10 leads to a reduction ofα-cleaved products, sAPPαandαCTF,and an increase of two Aβspecies,Aβ1-40 and Aβ***,elevated level of SCG10 results in the accumulation of APP in the post-Golgi vesicles and cell surface by facilitating anterograde trafficking of APP out of the Golgi *** addition,the ability of SCG10 to regulate APP processing is significantly compromised when the palmitoylation- mediated membrane-anchoring of SCG10 is *** together,these results suggest that SCG10,as a binding partner of APP,paly a critical role in the regulation of APP processing and may play a potential role in the prevention of Alzheimer disease.

Recent studies have demonstrated that more than 80 genes are linked to monogenetic *** of these genes are related to ion ***1A remains to be the most relevant epilepsy gene so *** of the important features of the phenotypes of SCN1A mutations is the quantity-dependence,shown both experimentally in KO mouse and clinically in mosaic *** is thus suspected that the expression level of SCN1A may play a role the manifestation of *** promoter region and some non-coding exons of SCN1A have been identified *** is therefore possible to search genetic abnormalities in such regions in patients of epilepsy with febrile seizure plus（EFS+） but without mutations in coding exons.A variety of phenotypes and types of functional defect （funotypes） have been found in Na1.1 *** studies failed to correlate the funotypes with the phenotypes. Studies on the genotype-funotype-phenotype relationship will help us to understand the biological basis,and further in management,of *** will also be helpful in studying the normal physical functioning of the sodium channels.

Cyproheptadine is a histamine- and serotonin- receptor antagonist,and its effects are observed in the modulation of multiple intracellular signals in previous *** demonstrate here that intracellularly delivered cyproheptadine increased the delayed rectifier outward potassium current（I） by reducing the intracellular levels of phosphorylated PKA in mouse cortical ***-receptor binding assay indicated that cyproheptadine bound to sigma-1 receptor and mediate the cAMP/PKA *** to the sigma-1 receptor mimicked effects of CPH and knock down this receptor abolished the effects of CPH on *** of G eliminated the cyproheptadine-induced effect on I and PKA activity.K2.1α-subunit blocker reduced cyproheptadine-induced effect on I in mouse cortical ***,CPH also enhance the K current when expressed the K2.1α-subunit on HEK293 *** reveal for the first time that CPH enhances I by a sigma-1 receptor-dependent mechanism and selective targeting of the K2.1α-subunit. This result may help to explain some of the uncharacterized effects of CPH on neuron excitability and contribute to the understanding of CPH as more than just a competitive inhibitor of the histamine H and serotonin receptors.

The neuronal activity in SI cortex was collected when monkeys performed a haptic unimodal working memory *** found that in trials with correct task performance a substantial number of cells showed significant differential neural activity only when the monkeys had to make a choice between two different haptic *** of incorrect trials showed that such a difference in neural activity was significantly ***,the choice differential neural activity was barely observed in monkeys who performed a control task that was identical to the haptic unimodal working memory task but did not require the animal to either actively memorize the sample,or make a choice between the two objects at the end of the *** data also suggested the top-down influence on decisionmaking neural activity in SI cortex when the animal performed a haptic crossmodal working memory *** all the above results together,we believe that the differential activity may be a neural correlate of decision making, and thus conclude that some SI cells function particularly in the process of decision making when the animal has to make a haptic *** findings indicate for the first time that primary sensory cortex may play an important role in decision-making.

MicroRNAs(miRNAs) play important roles during development of the central nervous system(CNS). Several reports indicate that tissue development and cellular differentiation in the developing forebrains are disrupted in the absence of ***,miRNA functions during cerebellar development have not been systematically *** we conditionally knockout Dicer gene under the activity of human glial fibrillary acidic protein (hGFAP) promoter to examine the effect of miRNAs in the developing *** found that ablation of miRNAs resulted in a smaller cerebellum,mis-positioned Purkinje neurons,decreased proliferation and increased apoptosis in granule neuron precursors,decreased number of basket and stellate intemeurons and aberrant morphology of Bergmann *** addition,Notchl signaling appears to be blocked in miRNA-ablated Bergmann *** further identified miR-9 as differentially expressed in Bergmann glial cells,and its expression was promoted by Notchl *** also demonstrated that cortical radial glia exhibited normal morphology in the absence of miRNAs and yet reduced radial glial marker expression at embryonic stage,and that the postnatal radial glial transition to astrocytes was promoted in miRNA-ablated *** results further stressed the critical involvement of miRNAs in the developing CNS including cerebellum and provided first evidence of differential miRNA function in regulating radial glial phenotype in different regions of the developing CNS.

Increasing evidence suggests that the ubiquitin-binding histone deacetylase-6（HDAC6） plays an important role in the clearance of misfolded proteins by *** this study,we treated PC-12 cells overexpressing human mutant（A53T）α-synuclein（α-syn） and SH-SY5Y cells with *** was found that HDAC6 expression significantly increased and mainly colocalized withα-syn in the perinuclear region to form aggresome-like ***6 deficiency blocked the formation of aggresome-like bodies and interfered with the autophagy in response to MPP-induced ***,misfoldedα-syn accumulated into the nuclei,resulting in its reduced clearance,and finally, the number of apoptotic cells significantly *** together,HDAC6 participated in the degradation of MPP-induced misfoldedα-syn aggregates by regulating the aggresome-autophagy *** the mechanism may disclose potential therapeutic targets for synucleinopathies such as Parkinson disease.

Cannabinoid signaling system plays an important role in regulation of many neuronal functions in the CNS. Endocannabinoids function as a retrograde signaling to regulate the synaptic plasticity via activating presynaptic CB1 receptors in many brain *** date,the effects of this signaling on synaptic transmission in retina are poorly *** the present study,we aimed to explore the modulation of cannabinoid signaling on glycinergic and GABAergic inhibitory inputs,and glutamatergic excitatory inputs to retinal ganglion cells(RGCs)by patch-clamp techniques in rat retinal *** results show that activation of presynaptic CB1 receptors in both GABAergic and glycinergic amacrine cells(ACs)suppressed the frequency of miniature inhibitory postsynaptic currents(mIPSCs)in the RGCs through inhibiting the presynaptic voltage-gated calcium *** the effect of cannabinoid on glutamatergic excitatory inputs from bipolar cells to the RGCs is basically similar,the tonic inhibitory inputs from the ACs to the bipolar cells are *** these inhibitory inputs eliminated completely the suppression effect of cannabinoid on the excitatory inputs to the *** underlying mechanism is that deinhibition-induced depolarization of bipolar cells inactivates the presynaptic T-type calcium channels that is a target of the activated presynaptic CB1 receptors in the bipolar *** data revealed a new mechanism of cannabinoid modulating synaptic transmission in the inner retina.

Objective Carbonic anhydrase I(CA I) protein levels were found to be significantly increased in the spinal cords of both mutant SOD1-linked and sporadic ALS *** I is the cytoplamic isoform of the CA family which comprises of a total of 16 *** is not known how change of CA I might be related to ALS *** aims of this study are:(1) determine whether CA I is expressed in motor neurons in the spinal cord;(2) determine how is this expression is affected by ALS at the cellular level;(3) explore the mechanism by which CA I might play in ALS *** Immunohistochemistry and fluorescent immunostainning techniques were used to determine the pattern and changes of CA I expression in human spinal *** biochemistry and Western Blot analysis were used to examine the potential mechanism of subcellular location of CA ***(1) CA I is expressed in the motor neurons in human spinal cord;(2) a subpopulation of CA I is associated with ER membranes;(3) CA I change in ALS is more enhanced with membrane-associated CA *** CA I might play a role in ALS pathology via ER stress-mediated pathway.

Objective This study represents the most complete morphological classification of mouse retinal displaced ganglion cells(DGCs)in the inner nuclear layer available to date and provides a foundation for further understanding of the correlation of physiology and morphology and ganglion cell function with genetically manipulated ***(1)Flurogold was injected in two adjacent sites or one left site at the SC and the soma of DGCs in the mouse retina was stained by retrograding with Flurogold.(2)The morphology of DGCs was revealed by microinjection with Lucifer yellow.(3)The diameter of soma and dendritic field and distance from soma to the optic nerve were ***(1)The diameter of soma(or dendritic field)and distance from soma to the optic nerve were irrelevant.(2)The DGCs were classified into five groups based on their soma size,dendritic field size,and pattern and level of ***(1)It was proved that all morphological types of GCs in the GCL also existed in the ENL.(2)For monostratified DGCs,there is similar distribution of four morphological types in contralateral projection group and significant preference of B and C types in ipsilateral projection group.
