Objective Microglia are the main immune cells in *** monitor the microenvironment in healthy brain and respond rapidly to changes under pathological *** are activated and accumulate around lesion sites in various neurodegenerative *** it is widely accepted that microglia are embryonically derived from mesoderm,the source of microglia under pathological conditions in adult animals remains controversial. Here we combined a parabiosis animal model with a photothrombosis stroke model and investigated the origin of microglia under ischemic *** C57BL/6 mice and transgenic mice expressing GFP in microglial precursors were used to set up a parabiotic and blood chimeric *** the blood chimerism of the parabiotic animals was established,we induced ischemic stroke by injecting rose bengal intravenously and exposing the cortex to green *** brains from coupled-mice were fixed with 4%paraformaldehyde after photothrombosis and sectioned into 30 urn slices on a vibrating *** slices were examined under an epifluorescence microscope or a confocal *** We have successfully induced photothrombosis in parabiotic *** observed GFP-positive microglia in ischemic area of the brain in wild type C57BL/6 *** Microglia can be recruited into the brain from blood-derived progenitors after ischemic stroke.

Objective In the rabbit retina,orientation-selective ganglion cells（OSGCs）were discovered in *** orientation selectivity results from an incomplete antagonistic surround along the preferred orientation. Pharmacological studies confirmed that the inhibition,originated from amacrine cells,shaped the orientation selectivity of retinal ganglion *** in the mouse retina,there is no information about the OSGCs,that is why we try to identify them and characterized their physiological,morphological and pharmacological ***（1） On-cell and whole-cell patch clamp recording were performed to indentify OSGCs and to examine synaptic inputs.（2） Pharmacological experiments were carried out to check which kinds of neurotransmitters are essential for orientation ***（1）In the mouse retina,ganglion cells classed into B3 and Dl subtypes have orientation selectivity.（2）These cells preferred either vertical or horizontal orientation.（3）The orientation selectivity can be reduced apparently by picrotoxin,which is an antagonist of GABA and GABA *** B3 and D1 ganglion cells,who respond to onset and termination of flashing spot,have orientation *** of them are sensitive to GABA blocker.

Methods In the present study,we characterized the 3-D midbrain vasculature pattern formation during development using in vivo time-lapse confocal imaging of transgenic zebrafish larvae,of which blood endothelial cells express *** We found that midbrain angiogenic sprouts emanated from the bilateral choroidal vascular plexuses, elongated dorsally,and targeted to the mesencephalic central artery at the midline during 2-5 days post-fertilization (dpf).During later stages,some vessels sprouted from the pial vessels,grew ventrally and anatomosed with the mesencephalic central ***,stereotyped expression patterns of semaphrin 3E and wnt3 were required for this pattern formation of brain *** experiments are being performed to examine its detailed molecular mechanisms.

Therapeutic application of Asarum,a herbal medicine that has been used for centuries,reportedly causes acute respiratory *** responsible constituents,the sites of action,and the mechanisms involved in this side effect are *** investigated the effects ofβ-asarone,a volatile constituent of Asarum,on neurotransmission in the medullary respiratory neuronal network using extracellular recording of the rhythmic hypoglossal activity and voltage clamp recordings of the postsynaptic activity of the airway preganglionic parasympathetic motoneurons (APPMs)in vitro,β-asarone caused progressive decreases in the duration and area of the hypoglossal bursts in a concentration-dependent *** frequency and amplitude of the bursts were initially unaltered or temporarily increased,but were then inhibited progressively after prolonged *** with the inhibition of the hypoglossal bursts,the tonic and the phasic excitatory and inhibitory postsynaptic currents in the APPMs were attenuated. These data suggest that the Asarum-caused acute respiratory disturbance involvesβ-asarone-induced inhibition of neurotransmission in the medullary respiratory neuronal network.

Nkx2.2 homeodomain transcriptional factor is initially expressed in differentiating oligodendrocyte precursor cells(OPCs) and plays a critical role in regulating the differentiation *** studies showed that Nkx2.2 expression in oligodendrocytes is rapidly down-regulated as OPCs undergo terminal differentiation, but regained in mature myelinating oligodendrocytes in mature *** biphasic pattern of Nkx2.2 expression in oligodendrocyte lineage suggests that Nkx2.2 may have distinct functions during oligodendrocyte lineage progression. To define the role of Nkx2.2 in oligodendrocyte differentiation and maturation,we have used tetracycline-inducible expression system to specifically induce Nkx2.2 expression in oligodendrocyte cells at different developmental stages. Our results demonstrated that Nkx2.2 promoted OPC differentiation at embryonic stages,but repressed myelin protein production at neonatal stages,indicating that regulation of oligodendrocyte differentiation and myelin gene expression by Nkx2.2 is stage-dependent.

Objective The present study was undertaken to observe the effect of ceftriaxone(Cef) on the global brain ischemia and the changes of GLT-1 expression after preventive administration of Cef in rat with global brain ischemia. Methods Thionin staining was used to observe the morphological changes of pyramidal neurons in the hippocampal CAl *** and western blot analysis were used to observe the changes of GLT-1 expression. Results It was found that 8-min global brain ischemia resulted in severe delayed neuronal death(DND) on day 7 after the reperfiision,while preventive administration of Cef once daily for 5 consecutive days significantly prevented the DND in the CAl subfield induced by the global brain ***,the GLT-1 expression was significantly decreased on day 1 after global brain ischemia compared with sham group,while the preventive Cef administration could significantly increased the GLT-1 expression on day 1 after the global brain *** The preventive administration of Cef could up-regulate the expression of GLT-1 and protect neurons against global brain ischemia in hippocampal CAl subfield.
