Objective Increasing evidence indicates that in vitro neurogenesis is enhanced by hypoxia,and recent studies have shown that Notch1 signaling pathway,a classic pathway regulating neurogenesis,can be activated by *** aim of this study was to investigate whether neurogenesis in dentate gyrus(DG) of hippocampus can be promoted by intermittent hypoxia(IH) and the role of Notchl in the hippocampal neurogenesis enhanced by IH. Methods The proliferation of neural stem cells(NSCs) was identified by Brdu *** differentiation and migration of NSCs were identified by Brdu and NeuN double *** morphology of astrocyte was identified by GFAP immunohistochemistry *** 1-transgenic mice were used to identified the structure of *** deficiency mice were used to identified the role of ***(1) Intermittent hypoxia could promote the neurogenesis in DG of hippocampus,including proliferation of NSCs,migration of neuron,development of spine and astrocyte.(2) The promoted effects of intermittent hypoxia on neurogenesis were inhibited in Notchl deficiency *** Intermittent hypoxia could promote the neurognesis in DG,which was mediated through Notch 1 signaling pathway.

Neuronal morphogenesis requires extensive membrane remodeling and cytoskeleton *** we show that GRK5,a serine/threonine kinase of G protein-coupled receptors,is critically involved in neurite outgrowth, dendrite branching,and spine morphogenesis,through promoting filopodia ***,GRK5 is not acting as a kinase,but rather provides a key link between the plasma membrane and actin ***5 promotes F-actin bundling at membranes of dynamic neuronal structures through interacting with both F-actin and PI(4,5)***,coupling regulation of actin cytoskeleton dynamics and membrane remodeling is mediated by separate domains of GRK5 and is required for its effects on neuronal ***,GRK5 knockout mice exhibit immature spine morphology and deficient learning and *** findings reveal GRK5 as a critical mediator of dendritic development and suggest that coordinated actin cytoskeleton and membrane remodeling mediated by bifunctional actin-bundling and membrane-targeting molecules such as GRK5 is crucial for proper neuronal morphogenesis and the establishment of functional neuronal circuitry.

KCNQ potassium channels play a critical role in controlling membrane *** a typical voltagegated ion channels(Kv),KCNQ channels undergo closed to open transitions by sensing transmembrane potential changes and mediate inhibitory potassium *** of KCNQ channel activity as results of genetic mutation is causal to a variety of human diseases including epilepsy,arrhythmia and ***,discovery of small compounds to activate voltage-gated ion channels is an important strategy for clinical intervention in human diseases caused by membrane hyperexcitability,including anxiety,epilepsy,arrhythmia and neuropathic *** the binding of these molecules induces a conformational change leading to the opening of these channels,there is also a considerable conceptual interest in understanding the molecular basis of these "gain of a function" *** molecule activators of cation channels are rare;however,several novel compounds have been identified for KCNQ potassium ***,we reported two new molecules which potentiate either cardical or neuronal KCNQ channels and identified molecular determinants for their activity,which provide exciting insights into activation of voltage-gated channels by small molecuars.

Many GPCR agonists can activate receptor-tyrosine-kinases (RTKs) through transactivation, the mechanisms of which are poorly ***, we have investigated IGF-1 receptor (IGF-1R) transactivation by GABAB receptor using a chemical biology approach in rat primary *** show that GABAB receptor activation induces a dynamic assembly and disassembly of a protein complex, including both receptors and their downstream ***, a non-receptor tyrosine kinase, plays a key role in coordinating this dynamic ***, such a dynamic process is critical for transactivation and transactivation-dependent neuronal *** results reveal an important mechanism underlying GPCR transactivation of RTKs.

目的糖皮质激素（GC）除经典基因组作用机制外,还存在非基因组机制。GC非基因组作用的临床意义已比较明确,但这一与生俱来的重要调节机制不可能仅为临床用药而准备,一定有其尚未揭示的重要内在生理意义。本研究以应激时GC快速大量分泌的积极意义为切入点,探索糖皮质激素非基因组机制在应激调节中的生理意义。方法将去肾上腺小鼠随机分成GC（给予应激剂量GC）、RU486、RU486+GC、溶剂对照4组,负荷10%体重强迫游泳至力竭,记录各实验组力竭时间,并在各组强迫负荷游泳5分钟时,采用HE染色、电镜、反相高效液相色谱法观察检测心肌和骨骼肌的组织病理学和超微结构、ATP合成改变;采用血糖分析仪、氧化酶比色、Griess Reagent法检测血浆葡萄糖、乳酸、一氧化氮（NO）水平。结果 GC和RU486+GC组力竭时间显著长于RU486和溶剂对照组,各组GC总作用时间均少于30分钟,RU486不能阻断GC这一作用。溶剂对照组心肌和骨骼肌HE染色示组织充血肿胀,电镜下线粒体肿胀、骨骼肌M线消失。GC组较溶剂对照组血糖升高,血乳酸和NO降低,骨骼肌ATP增多。结论结果表明GC在应激调节中的存在快速非基因组防护机制,其中,以葡萄糖和ATP合成为重要节点的能量代谢和以NO为重要节点的自由基损伤可能为GC非基因组机制主要靶点。这一研究提示GC非基因组机制的生理意义可能在应激调节中有重要意义,因此进一步深入研究十分必要,可以预见其可能为抗应激损伤维护机体内环境稳态的新产品开发提供重要新策略。

Spreading depression(SD) is a temporary disruption of local ionic homeostasis that propagates slowly across the *** depolarisation associated with SD occurs in different types of stroke and may contribute to the initiation of migraine *** receptor(NMDAR) antagonists are known to be the most effective drμgs to suppress *** far,NMDAR antagonist are still perceived as unlikely candidate as anti-SD drμg for migraine aura treatment because of their unacceptable side effects,but the NMDAR antagonist,memantine is now used chronically in elderly for Alzheimer's and epilepsy patients and some subtype selective receptor antagonists may well be effective with reduced side *** an efficient in vitro chick retinal SD model with imaging for NMDAR pharmacology, we have determined whether NMDAR antagonists,with different type NR2 subunit selectivity,can suppress spreading depolarization associated with SD in the chicken retina *** results sμggested NR2A plays a key role in mediating SD,indicating potential therapeutic benefit for NR2A-preferring antagonist to treat SD-related neurological diseases,*** with alleviated side effect.

There are two major subtypes of neurons in the brain:projection neurons and *** Drosophila nervous system development,a subpopulation of single neuroblasts(neural stem cells) can give rise to both projection neurons and ***,in mammalian brains,whether single neuroepithelial cells or radial glias(primary neural stem cells) can produce both projection neurons and interneurons remains largely unknown. Using fluorescence-activated cell sorting(FACS),we cultured single Emxl-expressing neural stem cells derived from E10.5-11.5 mouse *** weeks after culturing in defined serum-free medium,multiple-immunostaining revealed that a subpopulation of single Emxl-expressing neural stem cells can produce both projection neurons and *** projection neurons in a single cell-derived clone express typical projection neuronal markers,such as Tbr1,Cux1,Ctip2,Foxp1 and Satb2,while interneurons in the same clone co-express Sp8 and ***, our preliminary results suggest that,during marine forebrain development,at least some projection neurons and interneurons share single multipotent progenitors.

Objective Oligodendrocyte precursor cells(OPCs)differentiate into oligodendrocytes(OLs),which are responsible for ***,the molecular mechanisms of maturation and myelination by oligodendrocytes remain *** present study is aim to explore the roles of SHP-2 in oligodendrocytes *** In this study,we first searched the potential tyrosine phosphatases involved in oligodendrocytes maturation using OPCs as the ***,we characterized SHP-2 expression during the development of oligodendrocytes by RTPCR, immunostaining and Western *** roles of SHP-2 in olidendrocytes maturation were detected by gene manipulations ***,the mechanism was examined by detecting activities of concerned kinases. Results(1)We found that SHP-2 was persistently expressed during developmental process of oligodendrocytes.(2) SHP-2 knock down attenuated oligodendrocytes maturation and this effect was triiodo-L-thyronine(T3)dependent. Over-expression of SHP-2 promoted maturation in primary cultured oligodendrocytes.(3)It has been identified that SHP-2 may regulate oligodendrocytes maturation through Akt and Erkl/2 *** SHP-2 promotes oligodendrocytes maturation via Akt and ERK1/2 signaling in vitro.

Objective Astrocyte becomes active in response to CNS ***,the mechanism of astrocytic activation is largely *** this study,we explored the roles of TRPC on astrocyte activation induced by lipopolysaccharide（LPS ） in *** we treated cultured rat neonatal cortical astrocyte with LPS to establish a model of astrocytic ***,the expression of TRPC mRNA were detected by real-time PCR and the concentration of intracellular calcium([Ca]) by laser scanning confocal *** characterizations of TRPC on astrocytic activition were observed with two kinds of TRPC *** LPS,a well-known inflammatory stimulus,induced astrocytic activation and increases in TRPC3 and TRPC6 mRNA levels and[Ca].Furthermore,TRPC antagonists,2-APB and SKF96365,inhibited LPS-induced cell proliferation,the increase in glial fibrillary acidic protein expression and the release of pro-inflammatory cytokines,IL-6 and IL-1β. Meanwhile,we found that 2-APB and SKF96365 inhibited an increase of[Ca].Conclusion TRPC,especially TRPC3 and TRPC6 are involved in astrocytic activation.

Brain malformations and retinal dysplasia in a subset of congenital muscular dystrophies(CMDs), dystroglycanopathies,are caused by disruptions of the basement *** extracellular matrix binding by the hypoglycosylated a-dystroglycan because of mutations in several glycosyltransferases is the underlying molecular *** is not known how the basement membrane becomes *** hypothesized that assembly of the basement membrane is reduced in rate thus causing a physically compromised basement *** test this hypothesis,we analyzed assembly of extracellular matrix on cultured neural stem cells and physical properties of the mutant basement membrane by atomic force *** assembly on the protein O-mannose pi,2-Nacetylglucosaminyltransferase 1(POMGnTl) knockout neural spheres was reduced when compared to the wildtype. When incubated with Matrigel,extracellular matrix(ECM) molecules including all 4 major components of the basement membrane,laminin,collagen IV,perlecan,and nidogen *** of ECM aggregation was reduced on POMGnTl knockout neural sphere as revealed by slower growth in aggregate size when compared to the *** staining and proteomic comparison revealed that the mutant basement membrane exhibited compositional changes from the *** force microscopic analysis revealed that the mutant basement membrane had reduced elastic modulus with surface topography showing bigger valleys than the controls. Thus,disruptions of the basement membrane in dystroglycanopathies may be caused by its weakened strength resulting from biochemical changes in composition and physical changes in structure and reduced assembly rate.
