Objective To investigate whether nifedipine,a blocker of L-type calcium channels can prevent iron accumulation and dopamine neuron degeneration in the substantia nigra(SN) of acute and chronic iron-overloaded *** The changes of tyrosine hydroxylase(TH) gene expression in the substantia nigra(SN) were detected by reverse transcription-polymerase chain reaction(RT-PCR),the TH-positive neurons were tested by TH immunohistochemistry,the dopamine content was measured by liquid chromatography-electrochemical detection and the iron contents of SN were determined by iron concentration *** Compared with the control group, the expressions of TH mRNA decreased,and the iron contents increased in the acute iron-overloaded SN of *** with nifedipine may partly restore these *** in the chronic iron-overloaded rats,the dopamine content in the striatum decreased after iron dextran treatment,which could be restored by *** dextran overload increased iron content in the SN and reduced the number of TH-positive *** with nifedipine prevented the increased iron content and restored the numbers of TH-positive neurons to control *** Nifedipine may inhibit iron-overload induced dopamine neuron damage in the iron-overloaded rats by suppressing iron toxicity.

Objective To identify the agonists for Neuromedin U 2 receptor by using NMU2R cell line which was established by cotransfecting HEK293 with the NMU2R plasmid（NMU2R/pCDNA3.1+）and reportergene plasmid （3xMRE/3xCRE/SRE-Luciferase/pGL3）.Methods Plate NMU2R cells in growth medium with 3%FBS in a 96- well format and incubate the cells at 37℃in a CO incubator for 12 *** the citrus flavonoids and Forskolin solutions to each well containing cells and *** the cells at 37°C in a CO incubator for 6-8 hours. Add an equal volume of 0.5xBright-Glo? solution,then test the value of a luciferase *** the activation effect of the various citrus flavonoids to Neuromedin U 2 receptor by the value of a luciferase ***（1） Naringin,Naringenin and Tangeretin can not activate Neuromedin U 2 receptor.（2）Hesperidin and Nobiletin can activate Neuromedin U 2 receptor.（3）The EC50 values of Hesperidin and Nobiletin for the receptor were 319μM and 6μ*** Hesperidin and Nobiletin can be the agonists of Neuromedin U 2 receptor.

By sustained activation of opioid receptors,chronic opioids cause analgesic tolerance,physical dependence,and opioid addiction,for which an effective treatment is still ***,as a part of limbic system,plays an important role in opioid *** this study,we using patch clamp recording method combined with pharmacological approaches investigated chronic morphine-induced adaptations in glutamatergic synaptic transmission in neurons of the basolateral amygdala（BLA）.Chronic morphine significantly increased glutamate synaptic transmission in pyramidal neurons in BLA containingμ-opioid *** naive rats,DAMGO,μ-opioid receptor's activator,could inhibit the release of presynaptic glutamate,while DAMGO would lost this inhibitory effect in chronic morphine *** found that in saline rats,DAMGO's effect was blocked by AACOCF,an inhibitor of phospholipase A and 4-AP,which blocks the presynaptic voltage-dependent potassium channels regulated by metabolites of the PLA *** PKA didn't play any roles in naive ***,we confirmed that in chronic morphine group,PLA2-AA-Kv pathway was downregulated and PKA was upregulated,which were involved in the effect of DAMGO on presynaptic glutamate *** job provided the new mechanism of opioid receptors in morphine treatment for further clinical perspective.

Objective Changes of neurotrophin-3(NT-3)expression in rat red nucleus following complete spinal cord transection have been studied in order to provide the experimental evidences for the mechanisms of regeneration and repair after spinal cord injury and for neurotrophins to treat spinal cord *** Male Sprague-Dawley rats weighing 240-260 g were divided into three groups at random:(1)Spinal cord transection group which was randomly subdivided into four subgroups according to the survival time at the day 1,3,7 and 14 d after operation(n=6 for each time point),in which the spinal cord of rat was transected completely at thoracic level 10-11.(2)Sham operation group(laminectomy only,n=6).(3)Normal control group(n=6).The expression of NT-3 in red nucleus was examined by immunohistochemistry and image *** NT-3 immunoreactive cells were found in red nucleus in both control and experimental *** operation,the expression of NT-3 increased gradually and reached the peak at the day 7 in red *** data reveal the profile of NT-3 expression in red nucleus after complete spinal cord transection at thoracic *** The results suggest that the demand of NT-3 increases in red nucleus after spinal cord injury and the increase of endogenous NT-3 may be beneficial for the survival and regeneration of the injured rubrospinal neurons.

Objective Literatures have shown that strong associations between HLA-B*1502 and Stevens Johnson syndrome(SJS)and toxic epidermal necrolysis(TEN)induced by carbamazepine in Hong Kong and Guangzhou *** are some conflicts between primers in the *** aim of this project is to find correct methods to determine HLA-B**** We enrolled 138 cases including the SJS/TEN patients,milder non-SJS/TEN cutaneous adverse drug reactions(mainly maculopapular)patients,tolerant controls and normal ***-SSP was performed using sets of HLA-specific primers and control primers to genotype HLA-B*1502 and high-resolution genotyping was used to sequence HAL-B**** After testing primers of published papers,we found that 6 cases should be HLA-B*1502 *** among them,high-resolution genotyping proved that only one was HLA-B*1502 *** analyzed HLA-B*1502 sequence and designed new primers for genotyping HLA-B*1502 and confirmed that testing results were exact same with sequence *** New primers used to genotype HLA-B*1502 have been *** strategies can help to guide personalized medicine and predict severe cutaneous adverse reaction in the clinic.

Objective Growing evidences suggest that electroacupuncture(EA)can improve traumatic stress-induced immuno-depression,but the mechanism needs further *** the present study,the contribution of EA on the neuroimmune modulation,as well as the involvement of calreticulin(CRT)-c-Src pathway in the traumatic stress was *** Two age groups of rats(3 month and 18 month)were undergone traumatic *** was performed on bilateral acupoints 'zusanli'(ST36)and 'sanyinjiao'(SP6).Immune consequences,cortical CRT and c-Src expression were detected by Western blot,immunofluorescence and CCK8 respectively.A long-term neuron culture was also used herein,time-lapse effect of lipopolysaccharide(LPS)on CRT-c-Src pathway was ***(1) Traumatic stress resulted in a suppression in lymphocyte proliferation and NK cell activity with age dependent manner, EA stimulation could significantly improve this immuno-suppression.(2)The age-dependent immuno-suppression was due to different triggering in CRT-c-Src pathway in cortical neuron.(3)Central modulation of CRT infienced c-Src expression,in the meantime improved traumatic stress induced *** EA could agedependently improved traumatic stress induced immuno-suppression,the effect was proposed to correlated with CRTc -Src pathway in cortical neuron.

MicroRNAs(miRNAs) are involved in the fine control of cell proliferation and differentiation during the development of the nervous ***-124,a neural specific miRNA,is expressed from the beginning of eye development in Xenopus and has been shown to repress cell proliferation in the optic ***,its role at earlier developmental stages is *** we show that miR-124 exerts a differential role in cell proliferation at the optic vesicle stage,the stage which precedes the optic cup *** show that miR-124 is both necessary and sufficient to promote cell proliferation and repress neurogenesis at the optic vesicle stage,playing an anti-neural role. Loss of miR-124 upregulates expression of neural markers NCAM,N-tubulin while gain of miR-124 downregulates these ***,miR-124 interacts with a conserved miR-124 binding site in the 3'UTR of NeuroD1 and negatively regulates expression of the proneural marker NeuroD1,a bHLH transcription factor for neuronal *** miR-124-induced effect on cell proliferation can be antagonized by *** results reveal a novel regulatory role of miR-124 in neural development and uncover a previously unknown interaction between NeuroD1 and miR-124.

Mutations in the parkin gene are currently thought to be the most common cause of familial Parkinsonism. Overexpression of parkin has been reported to prevent neuronal degeneration under various *** generated a transgenic mouse model in which expression of wild type parkin was driven by neuron specific enolase promoter. Parkin transgenic mice exhibited less reduction of striatal TH protein and number of TH positive neurons in the substantia nigra induced by l-Methyl-4-Phenyl-l,2,3,6-Tetrahydropyridine(MPTP).Such a phenomenon was especially more obvious in old transgenic ***-induced mitochondrial impairment in the substantia nigra pars compacta was improved in parkin transgenic mice accompanied by elevated transcriptional expression of bcl-2 and *** striatal a-synuclein accumulation was shown in old parkin transgenic mice,however,striatal Hsp70 expression revealed no significant changes within groups in both young and old *** results provide the first mammalian genetic evidence that overexpression of parkin may attenuate dopaminergic neurodegeneration induced by MPTP through protection of mitochondria and reduction of a-synuclein in the nigrostriatal pathway.

Objective Bipolar disorder is a kind of mood disorder,cycling between the episodes of depression and *** the symptoms of bipolar disorder overlap with some other psychosis,bipolar disorder may often be misdiagnosed at the first *** some patients with bipolar disorder showed hallucinations,and they would be misidentified as schizophrenia and treated *** genetic studies also found the schizophrenia and bipolar disorder shared common polygenic *** some research found both bipolar disorder and schizophrenia showed higher prevalences of neurological soft signs comparing to healthy controls,very few studies have been conducted to specifically investigate the differential prevalences of neurological soft signs between the these two clinical *** this study we attempted to examine the discrimination pattern of neurological soft signs between patients with schizophrenia and bipolar *** Ninety-six patients(48 patients in each group of schizophrenia and bipolar disorder) meeting the diagnostic criteria of DSM-Ⅳwere recruited for the current *** the patients were *** neurological soft signs were examined by the Cambridge Neurological Inventory. The age,gender,education,IQ was matched between the *** analysis was conducted to examine the differential patterns of neurological soft sigs in these two *** 66.7%of schizophrenia and 79.2% of bipolar disorder were correctly *** 72.9%of the all patients were correctly *** Schizophrenia and bipolar disorder showed a differential prevalence of neurological soft signs that may aid the clinical diagnosis of these two disorders in the near future.

Objective The study investigated whether the directionality of neural information flow(NIF) could be used to characterize the synaptic plasticity on thalamocortical(TC) pathway,and examined which oscillatory frequency band reveals the most in the cognitive deficiency caused by *** EMA(Evolution map approach) and IM algorithms were used to determine the direction of NIF between the thalamus and prefrontal cortex in five frequency bands,which are delta(0.5-4 Hz),theta(4-8 Hz),alpha(8-13 Hz),beta(13-30 Hz) and gamma(30- 40 Hz).Meanwhile,the experiment was performed by inducing long-term potentiation of TC pathway after recording local field *** The data presented that the energies were increased in delta and theta bands,and the synchronization between these two regions was decreased in theta band in Str group compared to that in control *** coupling direction index d and the unidirectional influence TC c were significantly decreased in stressed state at theta rhythm in the rats of CUS ***,the field LTP data presented that chronic stress decreased medial prefrontal cortex(mPFC) synaptic plasticity,which was certainly in line with the LFP ***, the decreased SampEn only in mPFC implied that there was a significant role of mPFC(the recipient region) in the information transmission though TC *** The results suggest that the directionality index of NIF in physiological oscillatory bands could be as a measure of synaptic plasticity.
