Objective To investigate the role of histamine in spontaneous neuropathic pain following peripheral nerve *** To prepare the model of spontaneous neuropathic pain(neuroma model),histidine decarboxylase gene knock out(HDC-KO)mice and their wild-type littermates(n=14-16/group)underwent complete transection of the sciatic and saphenous nerves *** of mice was scored daily until postoperative day ***1.3, Nav1.7 and Nav1.8 protein levels in the sciatic nerve and L4/L5 DRGs from both wild-type and HDC-KO mice were measured by immunohistochemistry and west *** HDC-KO mice showed higher levels of autotomy than ***1.3,Nav1.7 and Nav1.8 protein levels significantly increase in the sciatic nerve after nerve transection both in HDC-KO mice and their wild-type littermates,and presence of endogenous histamine had no effect on this ***.3 expression was increased,while Navl.8 was decreased by axotomy in DRGs of both ***,a role of endogenous histamine was only detected in regulating Navl.8 expression in DRGs after *** These results indicate that endogenous histamine probably suppresses the development of spontaneous neuropathic pain by down-regulating Navl.8 expression in DRGs.

Objective Generation and deposition of amyloid beta peptides（AB） and neurofibrillary tangle formation are key mechanisms involved in Alzheimer's disease（AD） *** is evidence supporting that tumor necrosis factor-α（TNF-α） receptors（TNFRs）-mediated signal pathway might contribute to production of AB and the subsequent cytotoxicity in the ***,the levels of soluble form of TNFRs（sTNFRs） and TNF-αconverting enzyme（TACE/ADAM-17） was not determined in AD *** In the present study,we analyzed sTNFRs levels and TACE activity in plasma at baseline in 76 patients of AD and 40 age- and sex-matched *** The AD patients had higher levels of both sTNFRs and TACE activity in plasma at baseline when compared with age-and sex-matched *** the subjects of AD,the sTNFRl levels correlated significantly to TACE activity（r = 0.385, P<0.01） and there were also positive and strong correlations between the sTNFRl and sTNFR2 in AD（r = 0.526,P<0.001）.Conclusion These results suggest that sTNFRs and TACE might be involved in the neuropathogenesis of AD.

＜正＞Objective To investigate the effects of acute and chronic stress on the spatial learning-memory function in mice and its *** Using animal model of single-factor acute stress（foot shock） and multivariate chronic unpredictable stress（crowding,cold / hot stimulation,flat noise,hathpace,foot shock,28 days）.The ability of spatial learning and memory in mice were examined by Morris water maze task,and the expression of c-fos in the hippocampus（HP） and prefrontal cortex（PFC） were measured by immunohistochemical *** The results showed that compared with control group mice,the spatial learning and memory of acute stress group mice were significantly increased in Morris water maze,and the expression of c-fos in the CA1,CA3 region and dentate gyrus（DG） of hippocampus and PFC were significantly increased in acute stress group *** chronic stress,compared with the control group mice,the spatial learning-memory function of chronic stress group mice were markedly decreased, and the expression of c-fos in the CA1,CA3,DG and PFC were remarkably reduced in chronic stress group mice. Conclusion The results suggested that acute and chronic stress causes the changes of spatial learning and memory function in mice which may be nearly related to the up- or down- regulation of c-fos expression in the HP and PFC.

Neuronal restrictive silencing factor (NRSF) is an epigenetic repressor with important role in regulating neurogenesis and ***, the physiological function of NRSF in postnatal brain remains an interesting ***, we reported that activity-dependent synaptic plasticity was suppressed by *** cre/ loxP system, nrsfwas conditionally knockout (cko) in forebrain excitatory *** recording of hippocampus CA1 radiatum area revealed a significantly upregulated long-term potentiation (LTP).We also found that activity stimulation led to significant enhancement of mature BDNF in cko mice, and this enhancement was mediated by the increased level of tissue plasminogen activator (tPA).Moreover, we found tPA was transcriptionally repressed by NRSF through direct binding to RE1 sequence in tPA *** of tPA activity abolished the upregulation of LTP in nrsf-cko *** findings provide an NRSF mediated epigenetic mechanism that moderates LTP in the hippocampus CA3-CA1 pathway.

Objective Pattern completion, the ability to retrieve complete memories initiated by partial cues, is a critical feature of the *** study investigates the role of dopamine in memory *** We have analyzed dopamine transporter heterozygous knockout mice (DAT+/2).Results (1) While these mice possess normal learning, consolidation, and memory recall under full cue conditions, they exhibit specific deficits in pattern completion under partial cue condition; (2) This form of memory recall deficit in the dopamine transporter heterozygous knockout mice can be reversed by a low dose of the dopamine antagonist haloperidol, further confirming that the inability to retrieve memory patterns is a result of dopamine *** Our results reveal that a delicate control of the brain's dopamine level is critical for pattern completion during associative memory recall.

Objective The medial geniculate body(MGB)and inferior colliculus(IC)are two important stages in the auditory ascending *** order to study their functional roles in the relaying pathway,we recorded simultaneously the neuronal responses of the two nuclei in behavioral rats under different physiological conditions. Methods(1)Teflon-coated tungsten electrodes were chronically implanted into the MGB and IC to record the spontaneous activity and the responses to sound stimuli.(2)The electroencephalogram and electromyogram were recorded to define the different stages of sleep,awake and anaesthetic states.(3)Histology of brain slides was used to confirm the location of the ***(1)Both MGB and IC neurons showed low spontaneous(tonic) activity in slow wave sleep(SWS)and under anaesthesia and high spontaneous activity in the awake *** rapid eye movement(REM)sleep stage,the spontaneous activities of these two nuclei were also lower than that in awake state,but higher than that in SWS stage.(2)In the MGB,the phasic responses to white noise stimulation increased as the sleep going to *** the IC,the phasic activity conversely decreased as the sleep changed to the REM stage. Conclusion The differences in the auditory responses of the MGB and IC in different physiological conditions would imply the differential modulations of the corticofugal projections to the two nuclei.

The mammalian brain is formed through a series of intricately orchestrated events whereby neurons born in germinal zones migrate for long distances to reach their final positions and form specific connections. The postmitotic neurons use the radial glial fibers as scaffold to migrate to their final destination during the brain *** has been established that the Reelin signaling pathway is crucial for neuronal migration and positioning during ***,the mechanism of Reelin action on migrating neurons has remained *** plasmid containing green fluorescence protein(GFP) sequence was injected into the lateral ventricles and transfected into the cells in ventricular zone of cerebral cortex in mouse embryos(E14.5-E17.5) by using in utero *** transfected brains were fixed with 4%PFA two or five days after the electroporation and used for immunocytochemisty against Reelin and different markers for radial glail *** results showed that the transfected cells expressed strong *** GFP-positive cells showed the typical morphology of radial glial cells possessing a long and thin process that spans the whole developing cerebral cortex and gave rise to intensive branching in marginal zone where Reelin is *** GFP-labeled cells located between ventricular zone and marginal zone showed the typical morphology of migrating neurons having a long and thick unbranched leading process and a short and thin trailing *** soon as the tip of leading process of migrating neurons reach Reelin-abundant marginal zone, they generate intensive branches and anchor to the marginal zone of the cortex,which facilitates somal translocation to finish the migration *** the present study,we first used in vivo research method-in utero electroporation to elaborate that Reelin induces branching of the leading process of migrating neurons,as well as the apical process of radial glial cells.

＜正＞Alcohol dependence is a complex psychiatric disorder demanding development of novel pharmacotherapy. Since the cyclic AMP（cAMP） signaling cascade has been implicated in mediating the behavioral response to alcohol, it would be of interest to determine whether phosphodiesterase-4（PDE4）,an enzyme that selectively hydrolyzes intracellular cAMP,is involved in the regulation of alcohol ***,the selective PDE4 inhibitor rolipram was used to measure its anti-drinking properties in alcohol-preferring Fawn-Hooded（FH/Wjd） *** showed that acute administration of rolipram（0.05,0.1,and 0.2 mg/kg） selectively and dose-dependently reduced 5%ethanol, but not 10%sucrose,operant self-administration;it also decreased voluntary 5%ethanol intake,but not water consumption,for the initial 30 min in rats showing high stable preference for *** course assessment revealed the significant anti-drinking effect of rolipram（0.1,0.2 mg/kg） lasted for 5 h in both continuous- and intermittentaccess to 5%or 10%***,chronic rolipram treatment decreased 5%ethanol consumption and preference during the drug treatment *** effect lasted even after withdrawal of drug *** locomotor activity was inhibited by rolipram at 0.1 and 0.2 mg/kg,the effect was present only in the initial about 20 min,which did not seem to alter long-term ethanol *** data suggest that PDE4 may play a promising role in ethanol drinking and dependent *** may be a potential treatment target for further development of anti-drinking drugs.
