Objective This study was undertaken to determine the role of glial glutamate transporter(GLT)-1a,one of splice variants of GLT-1,in the induction of brain ischemic tolerance by cerebral ischemic preconditioning(CIP) in a rat global cerebral ischemic *** The expression of GLT-1 a protein and mRNA in the CAl hippocampus was observed by using western blotting analysis,immunohistochemistry and *** appearance of neuron and the glutamate concentration in the CAl hippocampus was studied by using neuropathological evaluation and in vivo brain microdialysis combined with ***(1) CIP induced a significant up-regulation of the GLT- 1a expression in the CA1 hippocampus in a time course corresponding to that of neuroprotection of CIP against brain *** brain ischemia for 8 min induced delayed down-regulation of GLT-1a,obvious increase in glutamate concentration and delayed neuronal death of the pyramidal neurons in the CAl *** the animals were pretreated with CIP before the severe ischemia,the above changes induced normally by the severe ischemia were effectively prevented.(2) The preventive effect of CIP on the above changes was significantly inhibited by intracerebroventricular administration of GLT-1 a AS-ODNs which has been proved to specifically inhibit the expression of GLT-1 a protein and mRNA.(3) The concentration of aspartate and GABA was also elevated after severe brain ischemic *** the CIP had no effect on the elevated aspartate concentration and induced a further elevation in the GABA *** GLT-1a participates in the acquirement of brain ischemic tolerance induced by CIP in rats.

The olfactory bulb(OB),an important odor information processing center in olfactory system,not only has heavy intermutual neural projections with brain areas related to olfaction,including piriform cortex,amygdala, hippocampus and striatum,but also receives regulatory affections from other brain regions such as raphe nuclei and locus *** of these projections provide the possible modulation of the activity in the OB by brain *** awake and anesthetized states are two different brain conditions and are both used extensively in electrophysiological studies of ***,the activity patterns of OB local field potentials(LFP)in responding to odor stimulation at different brain states are *** clarify this question,the LFP oscillations at granule cell layer of the OB in awake and anesthetized animals were *** found that the LFP responses induced by odor in awake state were complex,with different frequency bands showed different patterns:13-30 Hz,increased significantly;30-60 Hz,almost no response;and 60-90 Hz,decreased *** addition,the increased response of 13-30 Hz was transient, whereas the decreased response of 60-90 Hz was ***,when the animals were anesthetized by chloral hydrate,all of the three frequency bands showed significantly increased and long-lasting ***, the activity patterns of OB LFP in responding to odor stimulation are significantly different when the animals are under awake and anesthetized states,reflecting different neural mechanisms involved in the odor information processing under the two brain states.

Orexin A and orexin B are newly discovered peptides synthesized exclusively within the hypothalamus. The orexinergic fibers widely project to various regions in the brain including the cerebellum,an important subcortical center for motor *** we reported previously,orexin A and orexin B excite cerebellar interpositus nucleus （IN）neurons via a direct postsynaptic *** this study,the underlying ionic mechanisms and their physiological function have been further *** results showed that both orexin 1 receptors（OXR）and orexin 2 receptors（OXR）are presented in the rat cerebellar *** cell patch-clamp recordings from cerebellar slices showed that orexin A and orexin B selectively excited projection neurons（43/47,91.5%）instead of interneurons（n=5） within the IN with a dose-dependent *** orexin-induced excitatory responses maintained in the presence of 0.3 uM tetrodotoxin,suggesting the actions of the peptides on IN neurons are ***-R7943 and Ba,selective blockers for Na-Caexchangers and inward rectifier K channels respectively,together totally blocked the orexin A-elicited inward current,suggesting that orexin A excites the IN projection neurons via both activation of Na-Ca exchangers and closure of inward rectifier K ***,bilaterally microinjection of orexin A into the IN significantly increased the rat exploratory behaviors in an open field,shortened the duration of passage through the balance beam and prolonged the time of the rats on an accelerated *** results demonstrate that the central orexinergic system arising from the hypothalamus may directly modulate the activities of IN projection neurons and participate in motor control through its modulation on final outputs of the spinocerebellum.

Objective Sacral neuromodulation(SNM) has been an effective method to recover and rebuild the bladder function after spinal cord injury(SCI).The mechanism of SNM is still not well *** is generally believed that neuromodulation of reflex pathways at the spinal cord level as well as in the supraspinal pathways is involved in this *** present,it is still a problem how to choose suitable stimulation parameters for effective *** want to improve the effects of SNM by using different stimulation *** present,we are presenting a complete SCI rat model for spastic bladder to study SNM and standardize the surgical procedures and post-operative care. Methods The adult female Sprague-Dawley rats are completely transected in the T9-T10 vertebra after laminectomy in general *** gelfoam is inserted between the cut ends to fill the transection cavity of spinal cord to ensure complete *** prevent infection,every rat is introduced penicillin(150 mg/kg) for one week following *** SCI,the bladder of rats was manually expressed once a day(Crede's maneuver).The weight of expressed urine in SCI rats is used to reflect the residual volume and assess the recovery of bladder activity. Results The weight of urine expressed daily increases and reaches at a peak in 1-2 weeks post *** are all kinds of complications in SCI rats,some leading to death,but the complications can be decreased with proper postoperative *** several pilot experiments,the surgical procedures and post-operative care are standardized. We consider that the rat model behaves a flaccid bladder in the first 2 weeks after *** 2 weeks,along the recovery of spinal cord the rat behaves a spastic *** urine retention decreased dramatically,then urges urinary incontinence happens and could be used to study SNM.

Circadian pacemaking requires the orderly synthesis,posttranslational modification,and degradation of clock *** mammals,mutations in casein kinase 1(CK1)εorδcan alter the circadian period,but theparticular functions of the WT isoforms within the pacemaker remain *** selectively targeted WT CK1εand CK1δusing pharmacological inhibitors(PF-4800567 and PF-670462,respectively)alongside genetic knockout and knockdown to reveal that CK1 activity is essential to molecular ***,CK1δis theprincipal regulator of the clock period:pharmacological inhibition ofCK1δ,but not CKle,significantly lengthened circadian rhythms in locomotor activity in vivo and molecular oscillations in the suprachiasmatic nucleus(SCN)and peripheral tissue slices in *** lengthening mediated by CK1δinhibition was accompanied by nuclear retention of PER2 protein both in vitro and in ***,phase mapping of the molecular clockwork in vitro showed that PF-670462 treatment lengthened the period in a phase-specific manner,selectively extending the duration of PER2-mediated transcriptional feedback. These findings suggested that CK1δinhibition might be effective in increasing the amplitude and synchronization of disrup-ted circadian *** was tested using arrhythmic SCN slices derived from Vipr2mice,in which PF- 670462 treatment transiently restored robust circadian rhythms of PER2::Luc ***,in mice rendered behaviorally arrhythmic by the Vipr2mutation or by constant light,daily treatment with PF-670462 elicited robust 24-h activity cycles that persisted throughout ***,selective pharmacological targeting of the endogenous circadian regulator CK1δoffers an avenue for therapeutic modulation of perturbed circadian behavior.

Objective To characterize green fluorescent protein(GFP) expression in the retina of the preproenkephalin -green fluorescent protein(PPE-GFP) transgenic *** Fluorescent in situ hybridization(FISH) and immunofluorescence(IF) were first employed to confirm the successful transgenic manipulation and its application in showing retinal ENKergic *** the GFP expression was characterized using IF with antibodies to Fluorogold (FG),glial fibrillary acidic protein(GFAP) and syntaxin 1(HPC-1).Double labeling with other retina cell markers was then performed to further clarify the neurochemical properties of GFP-expressing amacrine ***(1) The FISH and IF showed a good consistency of the expressions of PPE mRNA and *** mRNA signals were present preferentially in somata of the inner margin of the inner nuclear layer(INL),while GFP immunoreactivities were present in fibers in the inner plexiform layer(IPL) in addition to the positive somata in the INL.(2) Double immunofluorescence suggested that none of the GFP-positive cells were FG-positive ganglion cells or GFAP-positive astrocytes,while about 94%of the GFP-positive cells were HPC-1 -positive amacrine cells in the INL.(3) The colocalization of GFP with several retina cell markers,such as GlyT1,GAD67,ChAT,CB and vGluT3 indicated that the GFP-positive cells in the INL were different subtypes of the amacrine cell population.(4) No GFP and PV,CR, vGluT1 or vGluT2 double-labeled neurons were observed,although the distributions of some cells were similar to the distribution of *** The present results indicate the layer specific expression pattern of GFP-positive ENKergic neurons in the retina and also provided morphological evidence for further functional *** conclusion, the PPE-GFP mouse line can be highly useful for the study of specific retina amacrine cell population.

Objective Calcineurin is a serine/threonine protein phosphatase that regulates neurotransmission, neuronal structure and plasticity,and neuronal excitability in mood disorders,including *** evidence has suggested that calcineurin is involved in the regulation of depressive-like ***,little is known about the neurobiological mechanisms that underlie the mood-regulating effects of *** the current study,we aimed to investigate the potential mechanism by which calcineurin mediates the development of depressivelike behavior and the involvement of calcineurin in the action of antidepressant medication in the chronic mild stress (CMS) *** Rats were subjected to stress daily for 3 weeks and the hippocampal tissues were removed for the measurement of calcineurin activity by assessing CnAαand pSynapsin I ser62/67 levels using Western blot.
