With the objective to optimize the culture systems of dairy goat SSCs,10000 goat SSCs were seeded per well in a different extracellular matrix coated 96-well *** matrix include Laminin(5μg/ml), Fibronectin(1 ug/ml),Matrigel and Galetain(0.2%) were compared for the enriching efficiency and enhancing growth *** including basic Fibroblast growth Factor(bFGF,4ng/ml) or BIO(1μM) in basal SSCs medium and two other commercial medium(NF,mTesR,commercial feeder- and serum-free medium for mouse and human ESCs) were compared for the ability of promoting proliferation and mataining *** markers by immunocytochemical analysis and proliferative index were carried out to measure extracellular matrix and cytokines' function on SSCs'*** results showed that Fibronectin has the most robust role in enriching goat SSCs after the cells were seeded 2 hours(AP:1%;Oct4:70%;CD49f:57%;c-kit:6.5%and Gfra1:50%).Laminin enhance the SSCs proliferate mostly (nearly 10-fold after 7 days' cultlure),BIO nearly is equal to bFGF's fuction in sustaining the cells' pluripotency(AP:75%to 73%;Oct4:100%respectively).mTesR promote the colonies formed easiest(nearly 20 clones formed per 10000 cells).These data may provide important information for the further goat SSCs' study.

Objective:To study the effects of high altitude hypoxia environment on rat lung ultrastructure and the expression of HIF-1αin the rat ***:50 SD rats were randomly divided into 5 groups,namely group 1 d Golmud(altitude 2 700 m),2 d Tanggula group(altitude 5 000 m),3 d Nagqu group(altitude 4 500 m) and 30 d Nagqu group(altitude 4 500 m),the control group(in Xi'an,elevation 5 m).4 time-consuming experimental animals 1 d from Xi'an to the Golmud,Qinghai(altitude 2 700 m),2 d to the Tibetan Tanggula(altitude 5 000 m),3 d to the Naqu(altitude 4500 m),30 d in Tibet *** and electron microscopy of the lung tissue samples,Western Blot method to detect the lung tissue of hypoxia inducible factor-1αexpression,RT-PCR method for detection of high altitude hypoxic group,lung tissue expression of hypoxia inducible factor-1αmRNA ***:2d Tanggula acute hypoxia in lung tissue microstructure and ultrastructure of apparent high altitude pulmonary edema,and after hypoxic group after 30d Nagqu significantly reduce high altitude pulmonary edema,lung tissue was no Canon of Western blotting,HIF-1αProtein expression,RT-PCR detection of HIF-1αmRNA 30d Nagqu group in the expression of a certain amount,the remaining three experimental groups the expression of HTF-1αmRNA not significant(P> 0.05),30d expression of HIF-lamRNA Nagqu group were significantly higher(P<0.01). Conclusion:The hypoxia acclimatization for improved HIF-1a mRNA expression is beneficial to the hypoxic altitude pulmonary edema.

The ubiquitin-proteasome degradation pathway plays an essential role in multiple cellular processes,including cell cycle progression,proliferation,apoptosis and *** vitro,in vivo and clinical studies suggest the potential use of proteasome inhibitors as novel anticancer *** the current study,we are providing experimental evidence for that celastrol isolated from traditional Chinese medicine potently and preferentially inhibits the chymotrypsin-like activity of a purified 20S proteasome and human prostate cancer cellular 26S *** of the proteasome activity by celastrol in PC-3 (androgen receptor- or AR-negative) or LNCaP(AR-positive) cells results in the accumulation of ubiquitinated proteins and three natural proteasome substrates(I B-,Bax,and p27),accompanied by suppression of AR protein expression(in LNCaP cells) and induction of *** with celastrol resulted in tumor growth inhibition in prostate cancer xenografts,associated with suppression of the proteasomal activity and induction of apoptosis in *** results show that celastrol is a natural proteasome inhibitor that has a great potential for cancer *** computational modeling,it was predicted that the ketone carbons of celastrol might interact with the catalytic site of chymotryptic subunit of the ***,the reduced form of celastrol had significantly decreased proteasome inhibitory and apoptosis-inducing activities,suggesting that the ketone structure was an active site of *** further investigated the role of AR on cell death modulation in response to proteasome inhibition and found that proteasome inhibitor induced significantly higher caspase-3 activation in parental PC-3 cells compared to PC3-AR cells that stably overexpressed wild type ***-treatment of proteasome inhibitor and the AR antagonist Casodex caused significant decrease in AR expression associated with an increase in caspase-3 activity in both LNCaP and PC3-AR cel

ABCC4 is a member of the MRP/ATP-binding cassette transporter super-family involved in the active transport of endogenous anions and *** the overexpression of ABCC4 protein is found in many kinds of human *** evaluated the expression of ABCC4 in a panel of KYSE cell lines and found that ABCC4 was high expressed in KYSE 140 and KYSE *** we knocked down the expression of ABCC4 in these two cell lines and found that the expression of EP1,EP2,COX-2 and c-Myc were *** also found that ABCC4 siRNA successfully attenuate the phosphorylation of ERK. These results suggested that ABCC4 may contribute to the process of ESCC by altering the PGE/ERK/c-Myc signal ***,we treated cells with MK571 to inhibit the function of ABCC4 then tested the concentration of PGE2 in cell culture supernates and the growth rate of *** found that the concentration of PGE in cell culture supernates was decreased and the growth of cell was inhibited after the inhibition of ABCC4 by *** our opinion,some environment factors and biological factors may induce the expression of COX-2 and the production of PGE in the early stage of ***4 can transport the PGE out of the cell to combine with the receptors to active the downstream signal,and to promote the progression of *** high expression of ABCC4 can promote the tumorigenesis of cancer.

As is well known,the liver has a tremendous regenerative *** a classic two-thirds partial hepatectomy(PHx) in rat,the residual hepatocytes have an ability to reconstitute the lost liver mass within 7 *** tritiated thymidine or BrdU incorporation,there have been numerous studies tried to explore the cellular kinetics of liver ***,it should be emphasized that previous studies only demonstrated the general trend of liver cell *** can neither detail the dynamic proliferation of individual cells, nor reveal the heterogeneity among cells with different proliferation *** questions still remain unanswered,*** fate of early proliferation cells(EPC).Would EPC take further divisions? When and how many cycles? Methods:In this study,a combination of pulse and continuous labeling using three thymidine analogues,Bromodeoxyuridine(BrdU),Chlorodeoxyuridine(CldU) and Iododeoxyuridine(IdU) were employed to determine the temporal-spatial manner of the proliferation management during liver regeneration. This strategy allowed us to follow an individual cell for more than one cell cycle and to define how many cells and which cells undergo multiple ***:The results revealed that the residual hepatocytes clustered into three subpopulations to initiate the proliferation sequentially,and the corresponding percentage of each was 32%,17%,and 36%.Meanwhile,the remaining 15%of hepatocytes never *** addition,the periportal hepatocytes were identified as EPC,which firstly responded to PHx stimulation and re-proliferated synchronously at 54 ***,at least 11%of residual hepatocytes were identified to divide thrice or more. Conclusion:Based on the present analysis,we concluded a sequential model of the initial proliferation in residual hepatocytes,and for the first time,quantitatively elucidated the proliferation manner of three subpopulations during hver regeneration.

Although much effort has been devoted to the delineation of factors involved in the migration of neural stem/ progenitor cells(NSCs),the relationship between the chemotactic response and the differentiation status of these cells remains *** the present study,we found that NSCs in varying differentiation states possess different chemotactic responses to vascular endothelial growth factor(VEGF): first,the number of chemotaxing NSCs and the optimal concentrations of VEGF that induced the peak migration vary greatly;second,time-lapse video analysis shows that NSCs at certain differentiation states migrate more efficiently toward VEGF,although the migration speed remains unchanged irrespective of cell states;third,the phosphorylation status of Akt,ERK1/2,SAPK/JNK,and p38MAPK is closely related to the differentiation levels of NSCs subjected to VEGF;and,finally,although inhibition of ERK1/2 signaling significantly attenuates VEGF-stimulated transfilter migration of both undifferentiated and differentiating NSCs,NSCs show normal chemotactic response after treatment with inhibitors of SAPK/JNK or ***,interference with PI3K/Akt signaling prevents only NSCs of 12 hr differentiation,but not NSCs of 1 day or 3 days differentiation,from migrating in response to VEGF. Moreover,blocking of PI3K/Akt or MAPK signaling impairs the migration efficiency and/or speed,the extent of which depends on the cell differentiation ***,these results demonstrate that differentiation of NSCs influences their chemotactic responses to VEGF:NSCs in varying differentiation states have different migratory capacities,thereby shedding light on optimization of the therapeutic potential of NSCs to be employed for neural regeneration after injury.

Spermatogenesis is an intricately regulated process of cellular differentiation transforming primordial germ cells to *** gene expression during male germ cell development can lead to abnormal sperm production and *** transcription factors are known to play key roles in the differentiation of spermatogonia and meiotic ***250a,a defining subunit of the BAF chromatin remodeling complex that hydrolyze ATP to noncovalently restructure,mobilize,or eject nucleosomes to modulate transcription factor access to chromosomal DNA,is a trithorax group(TrxG) *** studies mapping the targets of TrxG action in mouse and human ES cells suggest these proteins also play a role in sustaining a heritable epigenetic state uniquely associated with ***,disruption of BAF250a results in loss of ES cell self-renewal and developmental arrest at the blastocyst ***,to date there is very little known about BAF250a in spermatogonial stem cells(SSCs) self-renewal and/or *** this study,we sought to test the relevance of BAF250a subunit in *** first conducted western-blot assays to confirm that BAF250a expressed in testes tissue,and then performed immunohistochemistry and immunofluorescence to identify BAF250a protein localization in the developing murine ***250a was detected in both germ and somatic cells in the neonatal testis but was specific to germ cells inside the seminiferous epithelium and Sertoli cells from juvenile development *** addition,BAF250a-deficient testis tubules regress from relatively normal to only Sertoli cell only over *** results support a role for BAF250a in maintenance of SSC self-renewal and extend our current knowledge on how SWI/SNF chromatin remodeling contributes to regulation of gene expression in mammalian cells.
