Fibroblast growth factors (FGFs) play an important role in the formation of mesodermaland ectodermal tissues during early development of vertebrate embryos. We have identified anovel member of FGF family in zebrafish, Fgf17b, an ortholog of human FGF17. Theexpression of fgf17b occurs during late blastulation and early gastrulation in the blastodermalmargin where cells commit to the mesoderm fate. During segmentation, fgf17b is expressed inthe forebrain and developing somites. Overexpression of fgf17b can dorsalize embryos, resultingin increased expression of dorsal markers and decreased expression of ventral markers.

It seems certain that COX-2 is related ito tumor and some data suggested that COX-2 might have relation with tumor malignancd and angiogenesis. In order to elucidate the relationship between COX-2 and tumor invasive and angiogenic ability, we transfected human transitional cell carcinoma(TCC) cell line, EJ, permanently with a COX-2 expression vector or the mock vector. The EJ-COX2 cells, which overexpressed COX-2, acquired increased invasiveness and angiogenic ability by activation of VEGF, uPA and MMP-2. Increased invasiveness and angiogenic ability were reversed by treatment with either selective COX-2 inhibitor, NS-398 , or dual COX inhibitor, indomethacin. These results demonstrate that overexpression of COX-2 can lead to phenotypic changes that alter the metastatic and angiogenic potential of TCC cander cells.

Members of TGFbeta superfamily play a central role in regulating a broad range of cellularresponse in a variety of organisms. The lipid-secreting exocrine Harderian gland contains a large amount ofporphyrins （mainly protoporphyrin IX, PPIX）in the glandular cells, the physiologicalsignificance of which is rather poorly understood. In the present study, the possibilityof using Fura-2 to measure intracellular calcium ([Ca]) changes in these cells wasassessed. It was found tha when Fura-2 loaded oolls were excited by ligh at 340/380nm, [Ca] increased spontaneously, indicating a photodynamic action powered byligh at 340/380 nm. In contrast, with the visible. spectrum calcium probe Fluo-3 (λ475 nm), carbachol at 10 μM induced [Ca]. increase;[Ca] did not change withoutcarbachol stimulation. 将本实验室筛选的耐甲硫氨酸草木樨状黄芪（Astragalus melilotoides Pall）变异系试管苗分别在含甲硫氨酸20mM和0mM的MS培养基中培养60天,期间更换一次培养基。分别取叶片提取总RNA并分离出其中的mRNA。通过锚锭引物01igo dTGC反转录和9个10核苷酸随机引物进行PCR扩增。电泳后发现有3个差异DNA片段只在甲硫氨酸胁迫条件下的草木樨状黄芪变异系基因组中表达,而在未胁迫的变异系中没有表达。这三个差异cDNA片段分别命名为tm01（280bp）,tm02（350bp）,tm03（360bp）。3个差异cDNA片段的RNA杂交结果显示,只有tm03片段存在明显差异,在胁迫条件下有杂交斑点而在未胁迫条件下没有杂交斑点;其余2个cDNA片段无论在胁迫或未胁迫条件下都没有杂交斑点。表明tm03是草木樨状黄芪变异系中甲硫氨酸胁迫应答基因cDNA片段。将tm03片段克隆并进行DNA序列分析,全序列在GeneBank中查询后未发现相关同源片段。进一步分析表明,该cDNA克隆可编码完整的蛋白。在该基因片段两端分别合成含HindⅢ和BamHI酶切位点的引物,用PCR扩增后亚克隆到pRSET A表达载体,转化宿主菌JM109（DE3）,经IPTG诱导后表达了N’端含6个组氨酸的蛋白质,分子量约为14kD,这为我们进一步研究该蛋白的结构和生物学功能奠定基础。

羊栖菜多糖（Sargassum fusiforme polysaccharides,SFPS）是从羊栖菜的全藻中分离、取得到的多糖。近年来关于SFPS及马尾藻科植物有效成分的抗肿瘤作用及其机理的研究已有报道,并证实具有一定的抑瘤作用,而其对于人体肿瘤细胞凋亡的诱导作用,迄今未见报道。本实验选择早幼粒白血病细胞系HL-60细胞作为SFPS抗肿瘤的研究对象,探讨羊栖菜多糖（SFPS）诱导人白血病HL-60细胞凋亡及其机理。实验采用MTT法观察SFPS的体外抗HL-60细胞增殖作用;扫描电镜、透射电镜观察SFPS作用前后HL-60细胞形态学变化;DNA琼脂糖凝胶电泳和流式细胞仪检测HL-60细胞凋亡。MTT结果发现,SFPS浓度越高,抑制作用越显著,浓度为300mg/L-1000mg/L时对HL-60细胞的增殖具有明显的抑制作用（P<0.01）;24,36,48和72h的IC分别为390,362,402和421mg/L。琼脂糖凝胶电泳显示,5mg/L和50mg/L SFPS处理后,无DNA梯形条带（DNAladder）出现;300mg/L SFPS作用24h后,产生典型的DNA ladder,呈180～200bp整数倍递增;SFPS浓度为500mg/L时,DNA ladder模糊,开始出现“涂片状”（smear）;随着药物浓度增高达1000mg/L时,DNAladder完全消失,呈“涂片状”。细胞形态学观察发现,细胞微绒毛减少、染色质固缩、边集,形成“帽状”或“新月状”结构,凋亡小体形成。流式细胞术结果显示,DNA直方图出现亚G峰;在一定浓度范围内,SFPS诱导细胞凋亡的作用呈现浓度和时间依赖性,同时G/M期的细胞比例有不同程度的增高,相应的G/G期细胞比例降低。结果表明,SFPS抗肿瘤作用与诱导细胞凋亡和G/M期细胞阻滞有关。深入探讨多糖类中药促肿瘤细胞凋亡的分子机制及其基因的表达的研究对进一步加深理解肿瘤细胞凋亡的分子机制具有重要意义,并有可能为多糖类中药治疗肿瘤提供理论根据。

Although the male and female pronuclei stay in a common zygotic cytoplasm, they are different in many ways, such as transcriptional activity, volume of pronuclei etc. This study was to investigate whether the differences between parental genomes could be erased by the first cell cycle. In order to inhibit the combination of parental genomes during the metaphase of the first cell cycle, we developed a new protocal to produce so called "PZM" embryos. PZM embryo occurred after inhibiting the parental genomes mingling and cytokinesis during the first cell cycle to form a single cell that contained male and female nuclei in a common cytoplasm. In PZM embryos the volume and transcriptional activity of male and female nuclei become equal. Our result suggested that the asymmetries of pronuclear volume and transcriptional activity between male and female genomes were erased by chromatin condensation of the first cell cycle.

Survivin是IAP家族中的一个特定成员,它特异性地在肿瘤细胞中高度表达,具有阻止细胞凋亡和调控细胞周期的功能。但到目前为止,关于Survivin的细胞定位以及抗凋亡机理还处在争论之中。Smac/DIABLO是最近发现的一个重要促凋亡蛋白,它主要通过与IAP(细胞凋亡抑制蛋白)的结合而间接促进细胞凋亡。目前尚无一个直接证据表明Survivin与某个凋亡基因相关。而我们成功地用GST-Pull Down以及免疫共沉淀的方法首次验证Survivin能与Smac/DIABLO直接结合,并且发现Smac/DIABLO的N端对于结合Survivin是必需的。另外,Survivin的突变体Surv-D71R和Surv-BIR(resides 1-98)均失去结合Smac/DIABLO的能力。通过免疫荧光分析以及激光共聚焦技术显示Survivin与Smac/DIABLO在细胞间期共定位于胞质中。此外,我们首次报道在Taxol(紫杉醇)诱导的HeLa细胞调亡过程中,Smac/DIABLO与细胞色素C一同从线粒体释放到细胞质中,且在HeLa细胞中高表达Smac/DIABLO可以促进Taxol诱导的细胞凋亡。为了阐明Survivin的抗凋亡机制,我们首先验证了Survivin不能与Caspases(半胱氨酸蛋白酶)结合,且发现Survivin的点突变体Sury-D71R(不结合Smac)使Survivin从抗Taxol诱导HeLa细胞凋亡转变为促进Taxol诱导的凋亡,Survivin的另一个突变体Surv-BIR(不结合Smac)也丧失了野生型Survivin的抗凋亡功能,因此我们提出了一个假想的Survivin抗凋亡途径:Smac/DIABLO是促凋亡因子,Survivin通过与Smac/DIABLO结合拮抗Smac/DIABLO的促凋亡活性,从而得到抗凋亡功能。为了进一步验证这个设想,我们在体外建立了一个CellFree体系:在这个体系中,Survivin本身不能够抑制Caspases激活,只有当Smac/DIABLO和XIAP共存时,Survivin才具有阻止Caspases活性的能力。这个机制第一次成功地阐明了Survivin的抗凋亡机理,对于研究Survivin的功能特点以及其在肿瘤治疗中的作用具有非常重要的价值。
