Drug discovery calls for faster method development and high-throughput analysis in supporting drug metabolism and pharmacokinetic（PK）studies,whereas the rapid, sensitive,and accurate analysis of biological samples re- mains a significant *** analysis of complex bi- omatrices（***）,liquid chromatography（LC） interfaced to mass spectrometry（MS）or tandem mass spectrometry（MS/MS）has been hampered by adverse ma- trix *** rapid assay development,it would be ben- eficial to improve the time-consuming method comparison and optimization steps by using generic procedures that work for a variety of ***,injudiciously combining the generic procedures including protein precip- itation for sample clean-up and electrospray ionization （ESI）for detection,as well as using conventional short- time isocratic or gradient LC,yield fast assay develop- ment,but often at the cost of decreased assay accuracy and increased risk of assay failure. We previously reported that the use of a mobile phase containing an extremely low concentration of ammonium formate（HCOONH）or formic acid（HCOOH）increased analyte ESi response and controlled against matrix effects. We designated these favorable effects‘LC-electrolyte ef- fects’.These favorable effects can be achieved in either the positive or the negative ion ESI mode,but not for at- mospheric-pressure chemical ionization（APCI）.The mag- nitude of the LC-electroiyte effect on the analyte response depends on both the concentration of the electrolyte modifi- er added into the mobile phase and its identity,which is also *** addition,LC is often optimized with more emphasis on improving the analytical sensitivity by concentrating the analyte on the LC column leading to a narrow and symmetric band and achieving sufficient sepa- ration between analytes and polar matrix components to avoid adverse ion suppression or enhancement of MS-MS *** these reasons,we proposed the so-called ‘pulse gradient sy

AIM:To provide primarily pharmacokinetic profiles and reveal potentially pharmacological components after oral administration of the extractum for Compund Danshen Dripping Pill(CDDP)in rats,which was mainly com- posed of protocatechuic aldehyde(PAL),Danshensu (DSS),lithospermic acid A,lithospermic acid B,salvi- anolic acid D,rosmarinic acid,salvianolic acid B and salvianolic acid A. METHODS:After orally administration of 200 mg CDDP extractum for 1 h,the portal vein blood,the femoral artery blood,the bile and urine samples were separately collect- *** gastrointestinal(GI)contents and mucosa were washed with physiological saline and the washing solution was *** whole GI tract was removed and homoge- nized for *** constituents and concentrations of 'all above samples were determined by the LC-MS analysis combined with the β-glucuronidase and sulfatase treatment. RESULTS:The investigation after oral administration of CDDP extractum for I h in rats indicated that:(1)Most of polyphenolic acids have been degraded into caffeic acid (CA)and DSS in the GI ***,CA could be easily absorbed into blood circulation and a trace of DSS was also transported into the portal *** remains of undegrad- ed polyphenolic acids and Danshensu were detained in the GI tract lumen.(2)Part of PAL was firstly oxidized into PAC in GI epithelial cells before absorption into the portal ***,PAC and the rest PAL were further transport- ed into blood circulation.(3)PAC and PAL were distrib- uted in blood circulation in free and conjugated forms after being glucuronidated in the liver and the kidney while only free CA and DSS appeared in *** that,all of these metabolites were gradually secreted into bite and urine. CONCLUSION:The results suggest that DSS,PAL, PAC,CA and their conjugated forms may be potential pharmacological components for CDDP extractum.

AIM:Nifedipine and atenolol are the representative drugs for calcium antagonists and β-blockers,their combination use is the highly acclaimed anti-hypertension programs in clinical *** order to assess the interaction of the two drugs,the bioequivalence and pharmacokinetics were investigated in a double layer tablet of compound for- mulation containing a sustained-release（SR）Nifedipine plus an immediate-release（IR）atenolol,compared with atenolol-IR tablet and Nifedipine-IR tablet individually in healthy Chinese subjects. METHODS:There were two stage tests:single-dose and *** single-dose test,two random cross-over studies were performed in 20 young subjects who were giv- en as monotherapy,a compound tablet（tested-drug,con- raining Nifedipine-SR 10 mg and atenolol-IR 25 mg）or two individual tablets（control-drug,a single Nifedipine- IR 10mg and a single atenolol-IR 25 mg）followed by a *** multi-dose test,two cross-over studies were performed in another 18 young subjects who were continuously treated for 7-days by two compound tablets once daily or two individual tablets（a single Nifedipine-IR 10 mg and a single atenolol-IR 25mg）twice *** blood samples were collected at different time points before and after drug *** drug concentrations were assessed by determining atenolol and Nifedipine with a validated HPLC or GC *** and tolerance were evaluated by monitoring adverse events and laboratory parameters. AIM:To evaluate the population pharmcokinetics（PPK） and pharmacodynamics（PD）of norvancomycin in patients. METHODS:We enrolled 146 patients who were con- firmed or suspected with gram-positive bacterial infections in this study.A PPK model was developed and validated by the analysis of NONMEM *** PK/PD profiles from the pharmacokinetic parameters in the patients and from MIC values were *** optimal dose regime was calculated by the analyses of the PK/PD model ac- cording to the clinical outcomes and bacterial *** norvancomycin MICs of clinically isolated swains from the patients population were determined by agar dilution meth- ods. RESULTS:The best model was a two compartment phar- macokinetic model with exponential interindividual error and additive residual error statistical *** finding of the study indicated that the change in Ccr values had impacts on drug clearance（CL）.For example,CL of pa- tients with decreased renal function would be influenced. The increased volume of a peripheral distribution（V） was observed when norvancomycin was co-administered with *** logistic stepwise analyses revealed that ratio of AUC/MIC is a major factor which could signifi- candy determine the clinical outcomes and bacterial eradi- cation for patients. When the ratio of AUC/MIC was more than 579.90 for Staphylococcus aureus infections or it was more than 637.67 for Enterococal infections,approximately 95% of patients could achieve cure clinical outcome. CONCLUSION:AUC/MIC should be a major PK/ PD parameter to determine efficacy of *** optimized regimen of norvancomycin can be simulated and developed for different subgroups of bacterial infected pa- tients with special physiologic and pathologic features.

To study the rational and scientific way of adminis- trating the drugs whose pharmacokinetics conform to three- compartment open *** on the double-bottle ex- ponential decay infusion introduced by Sun Rui-yuan and Song Jian-guo,this manuscript describe the comabined multi-bottle exponential decay infusion（CMI）in achieving optimal plasma drug concentration thai ensure adequate therapeutic effects in the absence of significant side effects and a rapid return of ***'s transformation was applied to verify group of equations listed according to the specific apparatus and the three compartments with elimination occurs mainly through central compartment, which yields the following equations used in calculating the volumes and concentration of bottle A,B and P respective- ly: C=(CssVK/d);Cbo=(CadK/αβ)(α+β-K);Vb =(d/K);Cpo=（Caπ）/10;Vp=(d/K;C（t）=(dCa/KV)(1-e). It is estirnated that the plasma concentration against time curve of the CMI is the function in mathematics of pai（π）, the fast distribution rate constant while in the situation of conventional mono-bottle infusion,the C-T curve is the function in mathematics of beta（β）,the slow distribution rate *** pharmacokinetic parameters were obtained and used for calculating the appropriate volumes and concentrations of each bottle *** has verified by animal experiment that the target plasma propofol levels （measured using HPLC）were rapidly attained and main- mined as desired,thus provided an alternative method of drug delivery.

AIM:hopride,which is a dopamine D2 antagonist with acetylcholinesterase inhibitory actions,is often prescribed for patients with functional *** primary metab- olite in humans is the N-oxide,generated by oxidation of the tertiary amine N-dimethyl *** urinary excre- tions of itopride and its N-oxide were 3.7% and 75.4%, respectively,in healthy subjects after a single oral admin- istration at a therapeutic *** support a predomi- nant role of FMO in the formation of itopride N-oxide in human liver *** addition,itopride appears to be a suitable probe for human liver *** aim was to establish a simple method adopting a one-step liquid-liquid extraction with dichloromethane followed by high-perfor- mance liquid chromatography（HPLC）with positive ion electrospray ionization tandem mass speetrometric（ESI- MS/MS）detection to simultaneously determine itopride and its N-oxide in human plasma and urine for FMO3 pharma- cogenetics study,using sulpiride as an internal standard （is）. METHODS:Acquisition was performed in multiple reac- tion monitoring（MRM）mode,by monitoring the transi- tions:m/z 360.1＞166.4 for itopride,m/z 376.1＞ 165.5 for itopride N-oxide and m/z 342.9＞112.2 for IS,*** were chromatographed on an Intersil ODS-3 reverse-phase chromatographic column （2.1 mm×150 mm,5 μm）by isoeratic elution with water （1000 mL water added 1 mL formic acid）-acetonitrile（60 :40,v/v）,the flow rate Was 0.2 mL/min with a total analysis time of 3 rain per run. RESULTS:The linearity for itopride and its N-oxide were in the range of 0.25-1000 ng/mL and 0.5-1 000 ng/mL,respectively,both in plasma and urine（r= 0.999）.The recovery of itopride was 95.8% for plasma and 101.2% for urine anti that of itopride N-oxide was 90.4% for plasma and 98.4% for urine,The intra- and inter-day RSD were＜15% both in plasma and urine. AIM:This study is aimed to investigate Runx2's regula- tion by phytoestmgens resveratrol and genistein,screen for Runx2 mutation in Chinese eleidocranial dysplasia(CCD) patients,also determine the functional difference between Runx2's two major isoforms-type Ⅰ and type Ⅱ. METHODS:The simian virus 40 large T antigens (SV40LT)gene was used to establish immortalized hBM- SCs and the cell line were then identified by specific sur- face markers confirmation and pro-longed life span and re- newable proliferative capacity *** we tested resveratml and genistein's regulation on the cell line by monitoring several osteoblastic markers and invested the possible involved pathway by western ***- quently,we determined the functional difference of Runx2's two major isoforms-type Ⅰ and type Ⅱ-in osteo- blastic differentiation by *** for the first time we screened for the Runx2 mutations in Chinese CCD patients by DNA analysis. RESULTS:We successfully established a permanent hBMSCs by stable transfection of *** on this cell line platform,we illustrated that phytoestmgens res- veratrol and genistein regulated Runx2's expression in os- teoblastic differentiation and might partly via MAPK path- *** also found that type I and type I1 play a different role in osteoblastic differentiation process,during which type Ⅰ major increased in the early phase and important for the cell proliferation while type Ⅱ predominantly functioned in the late stage 'and necessary for the cell maturation. What' s more,we sequencing coding domains and promot- er domains of Runx2 from 9 patients in 3 different CCD fa- milial cases and found two nucleotide changes. AIM:Previous studies identify that C7-OH epimers of taxoids are the thermodynamically more stable isomers due to the strong hydrogen bonding of the C7α-OH to the C4α- acetate acyl *** order to understand the effects of certain structure modification on taxoids' interactions with human hepatic cytochromes P450 better,the present study attempts to clarify the identity of the CYPs involved in 7- epi-Taxol,7-epi-10-Deacetyl'-Taxol,and their corresipond- ing C7β-*** metabolism and the underlying mech- anisms are also to investigate. METHODS:LC/MS/MS was used to identify the struc- tures of various taxoids metabolites by human liver micro- *** were conducted with CYP isoform spe- cific inhibitors and recombinant human CYP isofonns to as- cribe individual reaction to a single CYP isoform. RESULTS:Two monohydroxylated metabolites（M-1 and M-2）of 7-epi-Taxol were detected by LC/MS,and C3' （M-1）,C6（M-2）were proposed as the possible hydroxy- lation sites.7-epi-10-Deacetyl-Taxol was hydroxylated at C6 by human liver microsomes thus making M-3 as the unique primary *** inhibition studies and assays with recombinant human CYPs indicated that C3' （M-1）was generated predominantly by CYP3A4 and C6 （M-2,M-3）by *** with the formation of C6-OH-majorepi-Taxol decreased significantlympounds among all chemotherapeutic drugs,Paclitaxel,the K for C6-hydroxylation of 7-epi-Taxol decreased significantly（13 versus 3μmol/L,8.5 versus 1.7μmol/L）by human liver microsomes and recombinant human ***, the overall metabolism of 7-epi-10-Deacetyl-Taxol in- creased to 20% in contrast with the low biotransformation rate of 10-deacetyl-Taxol（less than 0.8%）. CONCLUSION:These findings suggest the distinct 3D conformation change of taxoids caused by the intromolecu- lar hydrogen bonding may attribute a major role in the sub- strate recognition by CYP2C8 and lead C-6 of taxoids more accessible to the active site.

AIM:To study the association between haplotypes of the SLCO1B1 and the rifampicin-mediated inducible CYP3A4 activity.A single-point determination of midazolam plasma concentration method was developed to assess the constitu- tive and inducible CYP3A4 activity.A pharmacokinetic study of a single dose of 450 mg rifampicin was conducted to evaluate the mechanism of rifampicin-midazolam interac- tion in different SLCO1B1 genotypic subjects. METHODS:Twenty-three healthy volunteers with differ- ent SLCO1B1 haplotypes（7 for SLCOIBI *1a/*1a,7 for SLCO1B1*1b/*1b,7 for SLCO1B1*1b/*15 and 2 for SLCO1B1*15/*15）were enrolled in this study. AIM:To investigate the pharmacokinetics and safety of gemifloxacin in healthy Chinese subjects and provide some theoretic bases for its reasonable application in clinic. METHODS:（1）12 healthy Chinese male subjects were enrolled in this study with an open label,3-period cross- over oral single dosing pharmacokinetic *** sub- jects sequentially took 3 doses of gemifloxacin（160,320 and 480 mg）according to the randomization schedule. （2）20 subjects were chosen to participate in a random- ized,double blind,multiple dosing pharmacokinetic *** subjects were orally given 320 mg gemifloxacin or matching placebo once daily for 7 consecutive days. Clinical observation and laboratory test were performed during the study for the assessment of adverse *** concentrations of gemifloxacin in serum and urine were de- termined by high performance liquid chromatogram （HPLC）.The pharmacokinefic parameters were analysed by 3P97 analysis software.
