Ca plays an essential role in the competence development of Escherichia coli cells. Using the potassium pyroantimonate method, we compared the Ca localization in competent and non competent E. coli cells. We found that two types of calcium pyroantimonate precipitates, round and acicular, existed on the surface of competent cells, but were not detected on non-competent cells, Though a few round or amorphous precipitates were found in the cytosol, there was no obvious difference in the cytoplasm between competent and non-competent cells. The data indicate that the competence of E. coli cells might correlate with Ca accumulation in the periplasm rather than that in the cytoplasm of E. coli cells.
