Antifungal azoles are widely used for controlling fungal *** response azoles stresses,fungi are able to transcriptionally elevate many genes to reduce toxic effects of azoles. However,the regulatory mechanisms of azole responses in filamentous fungi are poorly understood In this study,we found that deletion of transcription factor gene CCG-8,which was previously known as a clock-controlled gene,rendered *** hypersensitive to antifungal *** comparative analysis of genome-wide transcriptional responses to ketoconazole between the wild type and the ccg-8 mutant revealed that,among 489 transcriptionally up-regulated genes and 431 transcriptionally down-regulated genes in the wild type,transcriptional responses of 80 genes to ketoconazole were regulated by *** azole sensitivity test of all available knockout mutants for CCG-8-regulated genes,we found that CCG-8 contributes to azole resistance by regulating azole responses of many genes,including target gene ergll,the azole transporter gene cdr4,stress response genes aha1(NCU04087) and NCU06317,transcription factor gene NCU01386,hexose transporter HXT13 gene NCU01633,4 enzyme encoding genes and 6 unknown-function genes. Similar to its homolog Opilp in ***,CCG-8 also regulates phospholipid synthesis. However,there is no cross-talk between phospholipid synthesis and azole resistance in ***. CCG-8 homologues are conserved and widely present in filamentous *** of CCG-8 homologue encoding genefvccg-8 in Fusarium verticillioides also made this fungus hypersensitive to antifungal azoles.

Ranavirus cause disease in fish,***,many virulent ranavirus have been isolated and characterized from fish,reptiles,amphibians,and *** of them was the Chinese giant salamander iridovirus(CGSIV) isolated from diseased farmed Chinese giant salamanders(Andrias davidianus) at different areas in *** of habitat loss,pollution, and overharvesting,the population of wild Chinese giant salamanders has dropped ***1V cause serious systemic diseases and high mortality rates in tanned Chinese giant salamanders. Despite the economic and ecological significance of CGSIV,very little is currently known about its molecular *** far,the complete sequence and proteomic information of CGSIV has not been *** aid in the understanding of the molecular mechanisms of amphibian iridovirus pathogenesis and its evolutionary status,the complete genomic sequence and virion proteome of CGSIV should be determined and *** identify the virion proteins of CGSIV,the CGSIV virion proteins were analyzed by proteomic approach,based on a combination of one-dimensional SDS-PAGE and *** data obtained were analyzed by searches against the CGSIV and other iridoviral ORF *** of the CGSIV proteome revealed 13 proteins in viral particles:003L,010L,019L,021L,022R,058R,060R,069L,082L,089L,096L, 096L,*** gain additional information about the expression of the viral genes,RT-PCR was used to detect the existence of mRNAs of the identified *** mRNA expression of the 13 ORFs was examined by RT-PCR after 48 h of *** the genes were completely amplified. The identification of allopolyploidization events normally requires molecular dating and divergence assessments of progenitor *** on gene duplications only,either orthologs or paralogs,provides incomplete *** of mitochondrial DNA yield insights into matrilineal history,which may differ from patrilineal *** important food and pet fishes,the common carp and goldfish,appear to have experienced allotetraploidization sometime from 12 to 20 million years ago(Ma).However,much work is necessary to detail the initial polyploidization ***,use this group of fish as a model system to investigate competing scenarios for *** analyze the growth hormone gene(GH), recombination activating protein 1 gene(RAG1),and the 12 mitochondrial protein-coding genes combined in nineteen species of cyprinids,two species in Balitoridae as *** analyses clarify the phylogenetic position of the paternal and maternal ancestors for the common carp and *** estimation of matrilineal divergence(10.71～12.42 Mya) is significantly younger than that obtained by nuclear genes(17.11～19.64 Mya).Based on analyses of both genomes,the alloployploidization event common to common carp and goldfish dates to about I0.71～12.42 Mya. The matrilineal common ancestor of species in tribe Cyprinini might have doubled its genome by mating with a paternal ancestor in the subfamily Cyprininae,which was a sister-group that diverged around 4.69-8.93 *** work documents the necessity of considering both matrilineal and patrilineal histories when investigating alloployploidization.

Cuticular proteins(CPs) are crucial components of the insect *** numerous genes encoding cuticular proteins have been identified in known insect genomes to date, their functions in maintaining insect body shape and adaptability remain largely *** the current study,positional cloning led to the identification of a gene encoding a RRl-type cuticular protein,BmorCPR2,highly expressed in larval chitin-rich tissues and at the mulberry leaf-eating stages,which is responsible for the silkworm stony *** the Dazao-stony strain,the BmorCPR2 allele is a null mutation with significantly lower expression,compared to the wild-type Dazao *** BmorCPR2 in the stony mutant lost chitin binding ability,leading to reduced chitin content in larval cuticle,limitation of cuticle extension,abatement of cuticle tensile properties,and aberrant ratio between internodes and intersegmental *** variations induce a significant decrease in cuticle capacity to hold the growing internal organs in the larval development process,resulting in whole body stiffness,tightness and hardness,bulging intersegmental folds,and serious defects in larval *** our knowledge,this is the first study to report the corresponding phenotype of stony in insects caused by mutation of RR1-type cuticular *** findings collectively shed light on the specific role of cuticular proteins in maintaining normal larval body shape,and will aid in the development of pest control strategies for the management of Lepidoptera.

Previous studies have implicated that 2 at risk haplotypes(HapA and HapB) of gene encoding 5-1ipoxygenase-activating protein(ALOX5AP) were significantly associated with stroke The aim of this study was to explore the association between haplotypes of ALOX5AP gene and risk of ischemic stroke(IS) in Chinese Han population.A total of 246 patients with IS and 245 matched control subjects were *** ALOX5AP SNPs(SG13S377,SG 13Sll4,SG13S41, SG13S89,SG13S32 and SG13S35) were genotyped by SNaPshot minisequence technique.A common genetic variant SG13S114/AA in the ALOX5AP gene was associated with IS in this Chinese cohort(OR=3.060,95%CI=1.503～6.228).HapA(TGA) and HapB(AAAG) had no significant difference in the patients(36.3%,18.5%) and controls(37.6%,16.3%)(P=0.631, P=0.375).But the frequency of Hap(AGA) and Hap(GAAG) were significantly higher in the patients than in controls(P=0.028,P=0.006).While Hap(TGC) and Hap(GTAG) of ALOX5AP were significantly lower in the patients than in controls(P=0.025,P=0.026).To conclude, SGI3S114/AA of ALOX5AP gene was associated with an increased risk for *** haplotypes, Hap(AGA) and Hap(GAAG) were genetic risk factor of IS while Hap(TGC) and Hap(GTAG) were protective factor of IS in this Chinese population.

Rice is the most important staple food for a large part of the world's human population and also a key model organism for biological studies of crops as well as other related plants. However,most rice-related biological databases are based on expert curation;with the exponentially exploding volume of rice knowledge and other relevant data,expert curation becomes increasingly laborious and time-consuming to keep knowledge up-to-date,accurate and comprehensive,struggling with the flood of data and inevitably requiring a large number of people getting involved in rice knowledge curation. Here we present RiceWiki(http://***),a wiki-based,publicly editable,and open-content platform for community curation of rice *** extant relevant databases, RiceWiki features(1) harnessing collective intelligence in community curation of rice genes,(2) quantifying users' contributions in each curated gene,and(3) providing explicit authorship for each contributor in any given gene,with the aim to exploit the full potential of the scientific community for rice knowledge curation.A live demo for illustrating these features is the rice semidwarfing gene,which was curated collaboratively by multiple users(http://***/***/ Os01g0883800). Due to significant importance of rice,RiceWiki serves as a critical community-curated knowledgebase for the rice research *** the growing volume of rice-related data and contrastingly the small number of expert curators working on rice,RiceWiki bears the potential to make it possible to build a rice encyclopedia by and for the scientific community, which harnesses collective intelligence for collaborative knowledge curation,covers all aspects of biological knowledge,and keeps evolving with novel knowledge.

早期进化分析表明消化酶基因一胰核糖核酸酶基因(RNASE1)重复与消化系统的功能适应紧密相关,是研究基因进化以及新功能产生的一个重要基因家族。本研究中我们对消化系统简单的食肉目物种的RNASE1基因进行研究,结果在犬型超科中的鼬科,浣熊科,小熊猫和臭鼬科的四个科物种中发现了基因重复和假基因化,而且基因重复在四个科独立发生。并提出正选择作用是多个RNASE1基因出现的主要驱动力。此外,通过组织特异性表达研究,发现新产生的基因拷贝都在除肠道以外的组织中表达,包括肺,肌肉和脾脏。这些结果表明该基因在这四个科中中可能产生了新的功能。

基因组多倍化(whole-genome duplication,polyploidy)是生物界中的一个普遍现象,重复基因对(gene pair)的进化命运是生物进化的研究热点。阐明重复基因的进化历程及其在应对新环境中的响应机制对于生物体适应性研究具有重要意义。大豆基因组经历过两轮大的多倍化事件,由此产生了全基因组水平的基因重复,我们的研究表明,部分重复基因在功能上已经产生了分化,而有些重复基因在进化中还存在功能上的冗余。揭示大豆基因组重复基因对的分化机制及其序列结构上的变异,对于大豆基因组进化历程研究具有重要意义。本研究通过基因组学和转录组学的分析方法在全基因组水平上对大豆重复基因对的序列变化和转录水平进行系统分析,发现基因组特征变异导致的表达模式变化在重复基因功能分化中起到重要作用。

微生物是次级代谢产物的主要来源之一。植物内生真菌由于所处的环境特殊,同时与宿主植物之间存在复杂的相互作用,因而能够产生结构多样、活性广泛的次级代谢产物。这些次级代谢产物可能作为信号分子、抗氧化剂等参与内的生真菌与其宿主植物的相互作用。由于内生真菌难于培养、生长缓慢以及缺乏相应的遗传操作系统,对内生真菌次级代谢产物的生物合成与调控的研究相对较少。近年来,许多具有应用前景的新型化合物不断从内生真菌中发现,但内生真菌产生的次级代谢产物一般存在量低、产生不稳定等缺点,限制了这些化合物的进一步研究和利用。揭示内生真菌次级代谢产物生物合成与调控是解决上述问题的最好方式。
