杜氏盐藻是迄今为止发现的最耐盐真核生物之一,同属团藻目的雨生红球藻则对盐敏感。以遗传标记改为阿特拉津氯水解酶基因的植物双元表达载体p1301-atzA-22-4为载体,以雨生红球藻细胞为受体,对杜氏盐藻4 M盐度cDNA文库进行了高通量筛选,获得了5条高丰度表达的、盐胁迫上调表达的新cDNA,其GenBank注册号为KC469062-KC469066。该5条中的Ds-26-16长为732 bp,转化烟草、雨生红球藻及大肠杆菌,能显著提高不同受体细胞的耐盐性。由于烟草、雨生红球藻、大肠杆菌分属高等植物、藻类和细菌,亲缘关系远,表明其产生的抗盐性具有广谱性特点。将其唯一阅读框架重组到pET2lb(+)多克隆位点,并转化大肠杆菌,发现工程菌抗盐性提高的因素是其编码的小蛋白,并引起其它蛋白出现了盐胁迫上调/下调现象。blastp分析显示,Ds-26-16编码蛋白的氨基酸序列与非洲爪蟾蜍的Zincfinger BED domain-containing protein 1-like有短区段同源(69%,11/16),与Oreochromisniloticus的一个核酸结合因子(13JOH3RENI)有短区段同源(52%,12/23)。由此推测,Ds-26-16是一个新的盐胁迫反式调控基因。
