Objective To investigate the influence ofβ-elemene on the secretion of ANGⅡand the expression of ATR in hepatic stellate ***:In vitro,HSC-T6 were cultured for 24 hours and then treated with different dose ofβ-elemene(2.5mg/L,5mg/L and 10mg/Lβ-elemene).The control group was *** secretion of ANGⅡin the supernatant was detected by *** mRNA expression of AT1R for 4h,12h and 24h was detected by RT-PCR *** protein expression of AT1R was detected by ***:On the time point of 4h,the inhibition of ANGⅡsecretion in supernatant by 10 mg·Lβ-elemene was obvious vs the control group(P<0.05),while 5.0mg·L and 2.5 mg·Lβ-elemene had no inhibitory effect on the secretion of ANGⅡ(P>0.05).On the time point of 12h,the secretion of ANGⅡin supernatant by 10 mg·L,5.0mg·Lβ-elemene was markedly lower than the control(P<0.01,P<0.05).Following treatment with 5.0mg·L,2.5mg·Lβ-elemene for 24h,significant inhibition of ANGⅡsecretion was observed(P<0.05),but 10mg·Lβ-elemene had no such effect.β-elemene significantly reduced the abundance of AT1RmRNA in hepatic stellate cells after the treatment for 4,12,and 24 h in HSC in a dose-dependent *** expression of AT1R protein also decreased after the treatment withβ-elemene for 24 ***:β-Elemene can inhibited the secretion of ANGⅡand the gene and protein expression of *** may be the mechanism ofβ-elemene prevent the progress of hepatic fibrosis.
