Phytotoxic aluminum(Al) is one of the most serious limiting factors for crop production in acidic *** the function of biological N fixation,the soybean could be considered as an excellent rotation and intercropping crop for improving soil fertility and *** biological nitrogen fixation is carried out by prokaryotic organisms harboring the nitrogenase enzyme encoded by the nif,anf or vnf *** nifH gene encodes the iron protein subunit of nitrogenase,and a good correlation between the nifH gene and 16S rRNA gene phylogeny has been found for diazotrophic ***,using the nifH gene as a molecular marker in natural environments can be seen as an advantage,as it provides evidence for potential nitrogen *** mechanism of plant Al tolerance has been generally studied in relation to the exudation of organic compounds chelating Al into non-rhizotoxic complexes in apoplast and *** contrasting soybean genotypes[BX10(Al-tolerant) and BD2(Al-sensitive)]were previously characterized for root exudation and formation of oxalate,malate and citrate and their related enzyme activities in response to -P,+Al,or -P and +Al *** characterize the responses of the diazotrophic community to soybean genotypes under Al toxicity, the diversity and abundance of nifH gene were assessed in the rhizosphere soils of BX10 and BD2,at different growth *** a result,changes of the community structure,determined by denaturing gradient gel electrophoresis(DGGE),were associated with genotypes at day 40 and the presence of soybean at days 76 and *** analyses indicated two and twelve nonidentical sequences were responsive to BX10 and BD2 during the whole growth periods,respectively,and some of sequences in BD2 affiliated with Rhizobium *** Azospirillum brasilens with potential in promoting plant *** addition,dominant diazotrophs varied with the soybean *** gene abundance determined by real-time PCR was higher in BX1

As the fourth most common cancer and the second cause of death worldwide,gastric cancer(GC) causes nearly 1000,000 new cases and over 850,000 deaths each *** infection of Helicobacter pylori(H. pylori) is a common event and is shown to induce aberrant methylation in gastric ***-Barr(EB) virus infection is considered as another important carcinogen in gastric ***1,a member of the evolutionary highly conserved HIT protein super family,is ubiquitously expressed in diverse species including mammalian *** previous studies,Hint1 deleted mice had a marked increase in susceptibility to chemical carcinogen induced gastric tumors,mammary tumors and ovarian *** evidences show that HINT1 is a haploinsufficient tumor *** the present study,we explored possible correlations between the expression level of HINT1 and clinicopathological features in tissues from GC *** mRNA expression levels of Hint1 were detected by qPCR in 34 GCs(tumor and paired adjacent non-tumor tissues specimens) and analyzed the relationship with environmental factors *** and EB *** blot was used to detect the HINT1 protein levels in 51 GCs,and IHC was used to show the immunostaining in gastric mucosal cells,respectively.

Diacylglycerol Acyltransferase（DGAT） plays a critical role in the synthesis of *** this study,PCR-SSCP and DNA sequencing methods were employed to screen the genetic variation of DGAT-2 gene in 299 goats from three breeds（Boer goat,Chinese Xuhuai white goat and Chinese Haimen goat）.Three fragments of DGAT-2 gene were investigated,only exon3 of DGAT-2 gene showed *** alignment between nucleotide sequences of NM05793.2 in GenBank and the sequencing results of three PCR products with different patterns revealed that there was one mutation（A→G） in exon3 of DGAT-2 gene,which constructed two genotypes（AA,AB）.The frequencies of allele A and genotype AA were dominant in all three *** there was no significant difference from one another among the three *** genotype distributions were in good agreement with Hardy-Weinberg equilibrium（P>0.05） in each *** statistical differences were only found in withers heights（P<0.05） in Xuhuai goat between *** was indicated that individuals with genotype AA were significantly higher than those of individuals with genotype AB in withers height（P<0.05）.No polymorphism was detected in the intron3,exon8 and 3' flanking region.

主要组织相容性复合体(major histocompatibility complex,MHC),是脊椎动物某一染色体上(人为6号,小鼠为17号)编码主要组织相容性抗原,控制细胞间相互识别,调节免疫应答的一组紧密连锁的基因群。人类MHC又称HLA,小鼠MHC又称H-2。MHCI类基因在免疫系统中具有重要的作用,MHCI类基因编码的分子是免疫细胞发育、免疫突触的形成、控制T细胞以及NK细胞介导免疫应答的关键分子。而近年来发现MHCI类分子在中枢神经系统中也具有极其重要的作用。既往认为中枢神经系统是免疫豁免区,正常的未受感染的神经元并不表达MHCⅠ类分子,只在某些特殊情况下才高度表达,例如轴突横断,腹侧角根扯离,病毒或者寄生虫感染,暴露于细胞因子及电激活的药理学处理等。但是最近Carla ***教授等科学家研究发现即使是正常的,没有受损的神经元也同样可以表达MHCⅠ类分子,并且具有时空特异性和可调控性。如今已经在胚胎期以及出生后多个时期小鼠的大脑皮层,海马,背外侧膝状体,腹外侧膝状体等多个部位发现了MHCⅠ类分子的表达。MHCⅠ类分子在中枢神经系统中的受体及相关的信号转导机制研究已经起步,用小鼠作为模式动物,提示CD3ζ,Ly49分子,KIR样受体,PirB受体可能是中枢神经系统中假定的MHCⅠ类分子受体,并且推断MHCⅠ类分子以及其假定的受体在中枢神经系统中可能存在着完全不同于免疫系统的功能。已有证据初步推测MHCI类分子不仅发挥免疫功能,它对于正常脑的发育,神经元的分化,突触可塑性及记忆行为都具有重要的作用。MHC在中枢神经系统的表达及功能的研究是近年的研究热点,但该高度多态性的分子在小鼠中枢神经系统不同时期的表达模式未见系统性报道,其功能的研究处于起步阶段。本研究旨在研究MHCI类分子在正常发育的小鼠中枢神经系统中的表达模式,为进一步研究其在小鼠中枢神经系统发育过程中的作用提供实验基础。

利用重组杆状病毒表达系统在昆虫细胞中表达PCV2-ORF2基因,并以构建的重组杆状病毒为免疫原,通过杂交瘤技术,研制针对PCV2的单克隆抗体,为建立准确快速的PCV2诊断方法奠定基础。首先将PCV2-ORF2基因克隆到杆状病毒的转移载体pFastBacTM1中,然后将其转化入大肠杆菌感受态细胞DH10Bac中,与DH10Bac中的穿梭载体Bacmid发生转座,通过抗性和蓝白斑筛选,得到重组杆状病毒质粒reBacmid-ORF2,通过脂质体介导reBacmid-ORF2转染Sf9细胞,成功获得了表达PCV2-ORF2基因的P1代重组杆状病毒。将所获得的重组杆状病毒经Vivaflow200浓缩后,免疫6～8周龄BALB/c小鼠,取其脾脏与Sp2/0细胞融合,以IFA进行筛选,经多次亚克隆后得到3株能稳定分泌抗体的杂交瘤细胞,分别命名为5H6、4E2、5F7。通过IFA、IPMA及Western-blot试验对3株单抗特异性进行分析鉴定,结果显示,3株单抗均能与PCV2-Cap蛋白产生特异性的反应,并利用流式细胞术初步建立了对PCV2毒株感染PK15细胞的检测方法,从而为快速检测PCV2病毒奠定了基础。

为了提高传染性法氏囊病病毒(IBDV)VP2基因核酸疫苗的免疫效力,根据已发表的鸡源C3d序列,设计并合成在5′端添加连接肽(Gly4Ser)2序列的C3d基因。用同尾酶BglII和BamHI构建含有3拷贝C3d与VP2的的重组表达质粒pcDNA-VP2-3C3d。用脂质体法转染BHK21细胞,48h后,Western blot分析表明,表达的重组蛋白为162ku;间接免疫荧光试验检测转染细胞中具有特异性荧光。用重组表达质粒pcDNA-VP2-3C3d、与前期构建的真核表达质粒pcDNA-VP2免疫2周龄SPF鸡,二免14d后,间接ELISA法检测IBDV抗体效价,pcDNA-VP2-3C3d组抗体水平显著高于pcDNA-VP2组,MTT法检测鸡脾淋巴细胞增殖活性,pcDNA-VP2-3C3d组免疫诱导的特异性淋巴细胞增殖活性显著高于pcDNA-VP2组(P<0.05)。本研究表明C3d可增强VP2基因免疫诱导的IBDV特异性体液和细胞免疫应答。

Chinese wheat landrace Huangfangzhu(HFZ) had a high level of resistance to Fusarium head blight(FHB).To identify chromosome regions that were responsible for FHB resistance in HFZ,the mapping population of 102 recombinant inbred lines(RIL,F8) were developed by crossing HFZ and Wheaton,a US hard spring wheat from ***Ⅱresistance / FHB severity was evaluated using single floret inoculation method both in greenhouse and field *** chromosomal regions were associated with FHB resistance in *** major QTL were identified using composite interval mapping:one coincided with the known QTL Fhb1 on 3BS and explained 35.4%of phenotypic variances for greenhouse-field combined analysis;another was assigned to 7AL,which explained 18.0%of phenotypic variances for greenhouse-field combined *** minor QTL on chromosomes of 1AS,1BS,3AS and 5AS were identified using single marker regression model,and each QTL explained≤11.0%of phenotypic variances over single experiments and greenhouse-field combined *** the QTL identified here had positive additive effects and HFZ contributed the favorable *** the QTL on 3BS combined with the QTL on 7AL plus additional minor QTL at one to four loci,the RIL displayed significantly lower PSS than when they presented alone or than the RIL carrying none of *** positive QTL-QTL interactions were detected between the QTL on 1B and the QTL on 3A over three greenhouse experiments(F=28.36,P = 1.96e-7),and between the QTL on 1A and the QTL on 3A in field experiment(F= 11.55,P<0.05).

Rhizosphere microbial communities play key roles in the decomposition of organic matter, carbon sequestration and the promotion of plant *** is well known that microbes colonizing in rhizosphere soil assist with plant uptake of nutrient such as phosphorous,potassium and *** the exudation of roots,plants may regulate the rhizosphere microbial community,encourage beneficial symbioses,and change the chemical and physical properties of *** to the importance of plant-microorganism interactions in the rhizosphere for ecosystem function in natural ecosystems,it is crucial to understand plants influences on bacterium in this *** objective of this study was to investigate differences in organic acids secreted from two soybeans genotypes,BX10[Aluminum(Al)-tolerant]and BD2(Al-sensitive),and the rhizosphere microbial communities in response to soybean genotypes at different growth stages in acidic *** results showed that the organic acid species and quantities secreted by BX10 and BD2 were significantly different at their growth ***,distinct changes stimulated by two soybean genotypes were observed in the ratio of fungi/bacteria and Gram-negative bacteria(GN)/Gram-positive bacteria(GP) at different growing periods. Compared to BD2,BX10 improved in Shannon diversity from PLFA(phospholipid fatty acid) at the seedling and flowering stages,but decreased at the pod-setting *** and DGGE(denaturing gradient gel electrophoresis) fingerprint analysis showed that rhizosphere bacterium differed in the relative abundance of ribotypes,and that the increased abundance of certain microbes in the root vicinities was *** results indicated that influences of soybean on rhizosphere microbes depended not only on soybean genotypes,but also on their growth *** complementary set of methods monitoring soil microbial ecology between soybean genotypes may provide a useful practice conductive to agricultural effic

为分析2012年河南省猪流行性腹泻病毒(PEDV)的遗传变异情况,本研究设计合成2对扩增S1基因的特异性引物,利用RT-PCR方法,对2012年分离的5株PEDV毒株的S1基因进行扩增、克隆和序列测定,并对其进行遗传进化分析。结果表明:5株PEDVS1基因核苷酸同源性为98.3%～99.5%,氨基酸同源性为97.1%～98.9%。与2012年国内登陆的PEDV流行株相比较核苷酸同源性为97.8%～99.2%,氨基酸同源性为97.4%～99.1%。与参考毒株CV777相比较核苷酸同源性为88.7%～89.1%,氨基酸同源性为87.3%～88.4%。同源性分析结果表明:5株分离株S1基因变异情况基本一致,存在相同的插入和缺失,并且与国内2012年登陆的毒株相比没有突变倾向;但与参考毒株CV777相比较,在第163bp～166bp处有3个核苷酸插入,在173bp～174bp之间有9个核苷酸插入,在405bp～406bp之间有3个核苷酸插入,在463bp～464bp之间有3个核苷酸缺失,这些位点核苷酸的插入和缺失导致了其编码的氨基酸相应改变。PEDVS1基因在我国只有一个血清型,但PEDV分3个群,系统进化树分析表明5株分离株S1基因与2012年国内登陆的毒株均属于Ⅲ群,与2011年国内登陆的PEDVⅠ群流行毒株相比较核苷酸同源性为88.6%～89.3%,氨基酸同源性为85.3%～86.9%,而参考毒株CV777属于Ⅱ群。目前我国流行的PEDVS1基因有PEDV群和Ⅲ群的存在,但这些毒株的致病性和抗原性差异仍有待进一步研究。
