Calmodulin(CaM) is a multifunctional Ca-binding protein in eukaryotic cells,it plays important roles in regulating the growth and development,environment adaptation and stress tolerance in plant. A full-length cDNA sequence coding CaM of mulberry tree,which we designated MCaM-3(GeneBank accession No:GQ303247),was cloned based on the ESTs from mulberry *** sequence analysis showed that the MCaM-3 is 951bp in length,it contains a 143bp 5'-UTR(untranslated region) and a 358bp 3'-UTR,an open reading frame(ORF) of 450bp,encoding 149 amino acids with a predicted molecular weight of 16.85kD and an isoelectric point of *** SMART analysis showed that the protein of MCaM-3 has four EFh functional domains,which can bind *** analysis revealed that,CaM genes are highly conservative in mulberry and other species including Arabidopsis,poplar,soybean,wheat,rice and maize. Phylogenetic analysis based on MCaM-3 and CaM genes in other 19 species showed that mulberry tree shares close relationships with castor oil plant,tobacco and so *** expression level of MCaM-3 at different developmental stages of mulberry leaves and flowers and different sites in mulberry were also investigated,the MCaM-3 transcripts can all be detected with the most abundant expression on mature tissues in mulberry,such as mature female flower and climax *** results of semi-quantitative RT-PCR analysis showed that the expression lever of its mRNA could be increased significantly under the conditions of low temperature,drought and salt stress respectively compared to the normal growth *** results validated the proteins encoded by MCaM-3 may be involved in a general response to abiotic stresses in *** research will help to understand the resistance mechanism of functional genes in mulberry in the future.
