Aim of the study The present study examines the anti-hyperalgesia,anti-allodynic and synergistic effects of total glucosides of paeony(TGP),extracted from shaoyao as the root of the Paeonia lactiflora Pall,and total bioactive components(TBC),extracted from gancao as the root of the Glycyrrhiza uralensis in the rats chronic constriction injury(CCI) *** and methods Experiments were conduced 14 days after CCI *** and TBC were orally administered and the paw withdrawal latency(PWL) and paw withdrawal threshold(PWT) were determined for 1h,2h and up to 4h after drug *** and TBC were administered at fixed dose ratios of 1:3,1:1 and 3:1 and PWL and PWT were determined for 2h after drug administration.A dose-response curve for each fixed dose ratio of the combination was performed and compared with the theoretical additive *** Rats with CCI had a significantly decreased PWL and *** and TBC significantly inhibited hyperalgesia and *** interaction was observed after administration of a TGP and TBC combination at fixed dose ratios of 1:1,1: 3 and 3:1;the dose-response curves shifted leftward,respectively,from the theoretical additive values. Conclusions These results indicate the analgesia and synergistic interaction between TGP and TBC in this animal model of neuropathic pain.
