研究Pax7在奶山羊睾丸组织中的表达及分布规律,探索Pax7在奶山羊精原细胞自我更新维持的调控状况.本实验应用实时定量PCR技术及组织切片免疫荧光染色技术,检测Pax7在3、6.5、10、12、18月龄的不同发育阶段奶山羊睾丸组织中的表达水平.通过PCR扩增目的基因、构建pMcherry-N1-PAX7真核表达载体,在永生化精原干细胞系(Immortal male germlien stem cells of dairy goat, GmGSCs-I-SB)中过表达,检测Pax7及自我更新相关的标记.Pax7的相对表达量在3月龄时最高,之后随着年龄的增长逐渐下降.Pax7的表达量在各发育阶段与GFRα1、PLZF的表达量相比均是处于一个较低水平,表达Pax7的细胞数量比GFRα1+细胞数量要少.在组织切片中可见到Pax7蛋白定位于细胞核中,Pax7+细胞数量较少,均位于曲细精管基底膜处.pMcherry-N1-Pax7转染细胞后经mRNA,蛋白水平检测,基因过表达成功.得到了Pax7的体内雄性生殖器官中的表达规律,且检测到过表达PAX7基因后雄性生殖干细胞自我更新相关标记的上调.
