猪胰腺干细胞是糖尿病细胞治疗的种子细胞,可解决临床上供体细胞不足的问题。自噬在维持机体稳态和生理平衡功能中扮演重要角色,为探究自噬对猪胰腺干细胞的作用,本研究首先利用慢病毒系统将自噬标记基因LC3转导到猪胰腺干细胞,PCR、 Western blotting、免疫荧光和流式细胞术分析证明,LC3在猪胰腺干细胞中稳定表达,且过表达LC3的猪胰腺干细胞仍表达增殖和发育相关标记。血清饥饿、添加氯喹、血清饥饿并添加氯喹和正常培养四种条件分别处理稳定表达Lc3的猪胰腺干细胞4h后,激光共聚焦的结果显示饥饿处理组细胞中的自噬颗粒明显比正常组多,Western blotting可以看出饥饿处理后细胞LC3Ⅱ/LC3Ⅰ明显高于正常处理组。流式细胞术和Brdu分析发现诱导自噬4h后猪胰腺干细胞中处于S期细胞的比例和Brdu阳性细胞比率明显上升,而抑制自噬后细胞周期中S期比例和BrdU标记阳性率下降。本研究建立了稳定表达LC3的猪胰腺干细胞,证明一定程度诱导自噬能促进猪胰腺干细胞的增殖,同时为进一步研究自噬调控猪胰腺干细胞自我更新和发育分化机制提供了理想细胞模型和技术平台。
