AngiotensinⅡ（AngⅡ） plays a key role in the regulation of blood pressure and the process of cerebral *** have previously demonstrated that pinocembrin（5,7-dihydroxyflavanone）,one of the main flavonoids in propolis,improves cerebral blood flow in focal or global cerebral ischemia-reperfusion ***,we investigate the vasorelaxant effect of pinocembrin on AngⅡ- induced vasocontraction and its molecular mechanism of *** show that pretreatment with pinocembrin inhibited AngⅡ-induced vascular contractions in both endothelium-intact and endothelium-denuded rat aortic *** inhibition was associated with a significant attenuation of the myosin light chain 2 phosphorylation at Ser19 that result from AngⅡ*** rat vascular smooth muscle cells,pinocembrin significantly suppressed AngⅡ-induced Ca *** together,our results demonstrate that pinocembrin inhibits AngⅡ-induced rat aortic rings contraction,most likely through the inhibition of AngⅡ-induced Ca influx.

AIM Ciwujia is a traditional herb that has been widely used in China,and also named 'Acanthopanax senticosus'.It is an important Chinese folk medicine for the treatment of *** this study,the mechanism of Ciwujia on improving sleep in drosophila was studied using the microarray of whole drosophila *** The flies models of sleep-deprivation was maked by the alternative light-dark stimulating at *** then,the effects of Ciwujia on improving sleep of normal flies and flies sleep-derprivation models was determined by using the Drosophila activity monitoring *** this basis,the acting mechanism was explored by using the microarray technology of whole drosophila and bioinformatics data analysis *** Ciwujia can increase sleeping time of the male and female flies significantly at the concentration of 0.125%to 1%,and the sleeping consolidation index was increased;Ciwujia can increase the sleeping time and sleeping consolidation index significantly at the concentration of 0.25%.In flies head Ciwujia can changed expression of 194 genes,night light-dark stimulus can changed expression of 184 genes,the interaction of Ciwujia and night light-dark stimulating can changed expression of 685 genes in flies ***,dco,slo,DopR2 and Nos were screened out as sleep related *** Ciwujia can improve the sleep-wake cycle of fruit flies and sleep deprivation model of fruit flies,the mechanism involves co-regulation of multi-gene.

目的观察番茄红素对糖尿病性肾病的治疗作用及其可能的作用机制。方法采用高糖高脂饮食加链脲佐菌素法制备2型糖尿病大鼠模型。2型糖尿病大鼠分别ig给予番茄红素5,10和20 mg·kg每天1次,连续8周。8周后取尾尖血用罗氏血糖仪测定空腹血糖（FBG）含量,腹主动脉采血后分离血清和血浆,用全自动生化分析仪测定血清肌酐（Scr）、尿素氮（BUN）和24 h尿蛋白（UP）含量,放免法检测血浆胰岛素（Ins）、血管紧张素Ⅱ（AngⅡ）和肾组织AngⅡ含量。HE染色观察肾组织病理形态的变化,RT-PCR法检测肾组织血管紧张素转换酶（ACE）和ACE2 mRNA的表达,Western印迹法检测肾组织ACE2蛋白的表达。结果与正常对照组相比,模型对照组FBG,Ins,Scr,BUN,AngⅡ和24 h UP含量均明显升高（P<0.05）;与模型对照组相比,番茄红素10和20 mg·kg组Scr,BUN,AngⅡ和24 h UP含量均显著降低,Scr由（54.2±4.3）μmol·L分别降低到40.3±2.0和（34.4±3.8）μmol·L,BUN由（17.7±2.3）mmol·L分别降低到15.5±1.2和（13.1±0.5）mmol·L,血浆中AngⅡ由（858±56）ng·L分别降低到680±62和（623±64）ng·L,肾组织AngⅡ由（19.8±2.9）ng·g分别降低到16.7±2.6和（13.6±2.4）ng·g蛋白,24 h UP由（16.4±3.5）mg分别降低到13.7±1.9和（9.9±2.3）mg（P<0.05）。与正常对照组相比,模型对照组ACE2,ACE mRNA和ACE2蛋白表达水平显著降低（P<0.01）,与模型对照组相比,番茄红素5,10和20 mg·kg组ACE2 mRNA和蛋白表达水平则显著升高（P<0.05）,其中番茄红素20 mg·kg组效果更为明显（P<0.01）;ACE mRNA表达水平与模型对照组无明显差异。结论番茄红素能降低糖尿病模型大鼠蛋白尿,改善肾功能,减轻肾组织病理变化,增强肾组织ACE2 mRNA和蛋白的表达,提示番茄红素对糖尿病性肾病的治疗作用可能与抑制肾局部肾素-血管紧张素系统的途径有关。

AIM Our previous study has demonstrated the therapeutic potential of bone marrow derived-neural stem cells （BM-NSCs） in CNS disorders;however,the beneficial effects are modest duo to the poor survival and low neural differentiation *** this study,a new pharmacologic strategy targeted at anti-inflammation and anti-oxidation to promote the survival and differentiation of BM-NSCs thereby enhancing their therapeutic efficacy was *** We firstly generated and identified the BM-NSCs,then investigated the influnence of Sal B on proliferation and differentiation of the cells in the non-permissive circumstances by immunocytochemical staining,flow cytometry and ELISA *** We demonstrate that Sal B possesses the ability to promote BM-NSCs proliferation in a dose dependent manner;While in differentiation medium,Sal B promoted nestin BM-NSCs differentiated into greater numbers of NeuN and NF-M neurons;upon exposure to HO,Sal B exerted neuroprotective effects against HO-induced cytotoxicity,facilitating the cells survival in oxidative stress,inhibiting the apoptosis and LDH leakage, displaying a dose-dependent protective effects on *** of the above activities may be associated with the induction of a crucial NSC-related growth factor,brain-derived neurotrophic factor expression in *** Sal B may be a potential drug to upgrade the therapeutic efficiency of BM-NSCs in CNS diseases.

AIM To investigate whether Salvianolic acid A（SAA） exhibits its protective effects in rats through epoxide hydrolase （sEH） and Epoxyeicosatrienoic acids（EETs）.METHODS Heart and cerebral ischemia in rats were established through the occlusion of left anterior descending coronary artery（LAD） and middle cerebral artery occlusion（MCAO） *** 0.3,1 and 3 mg·kg was administrated before and after operation *** SAA treatment reduced infarct volume in heart and brain,improved cardiac function,and ameliorated neurological ***,the beneficial effects of SAA were attenuated by co-administration of 14,15-EEZE,a putative selective EETs ***,SAA increased the 14,15-EET levels in the blood, heart and brain of sham and ischemic *** for hydrolase activity showed that SAA 1 and 3 mg·kg significantly diminished heart and brain sEH activity of ischemic rats.A fluorescent assay in vitro indicated that SAA could inhibit recombinant human sEH activity in a concentration-dependent manner(IC_(50 = 1.62μmol·L ).Immunohistochemical analysis showed that SAA 1 and 3 mg·kg significantly decreased sEH protein expression in heart and hippocampus CA1 region of ischemic *** Cardiovascular protection of SAA is mediated at least in part via inhibiting sEH to increase EETs levels.

AIM To investigate the effect and pharmacological mechanism of icarrin（ICA）,which is the main component extracted from a traditional Chinese epimedium herb,on Aβproduction in AD-like APP transgenic *** PDAPPV717I transgenic （Tg） mice were randomly divided into model group and ICA treated(at doses 30 and 100μmol·kg·d) *** was orally administered to Tg mice with an age range 4-10 *** burden of Aβwas measured by ELISA and *** amyloid senile plaques were detected by Congo red staining and Bielschowsky silver *** expression of APP and BACE-1 were measured by immunohistochemistry and Western *** co-expression of Aβwith amyloid fibers was detected by applying double labeled *** Orally administered ICA decreased the number of amyloid senile plaques in hippocampus of Tg *** immunohistochemical examination of brain sections stained with polyclonal anti-Aβantibody showed reduced Aβburden, and Aβlevels were also decreased in the insoluble fractions of brain homogenates,as determined by *** expression of APP and BACE-1 in hippocampus was significantly decreased in ICA treated *** ICA could reduce the Aβburden and plaque deposits in the hippocampus of APP transgenic mice through depressing the expression of APP and *** may have a promising application prospect in treatment of AD.

AIM To identify salvianolic acid L（SAL） metabolites in *** Bile duct of wistar rats（n =6,male） was cannulated with PE tubing after *** bile samples were collected for 24 h after intravenous injection of 20 mg·kg *** group of 6 male Wistar rats were kept in metabolism cage after intravenous injection of 20 mg·kg SAL,urine samples were collected for 24 h and blood samples were drawn from ophthalmic veins of the rats by sterile capillary *** samples were acidized by HCl 1 mol·*** extraction of SAL and its metabolites were conducted with ethyl *** removal of organic solvent by nitrogen gas blowing,samples were reconstituted in mobile phase solvent before subjected to liquid chromatography-mass spectrometry *** About 60%of SAL and its metabolites was excreted from *** monomethylation metabolites, four dimethylation metabolites,1 monoglucuronidation metabolites,monoglucuronidation-monmethylation metabolites and monoglucuronidation -dimethylation metabolites were discovered in bile of wistar rats,and the metabolites were also detected in plasma and urine. CONCLUSION The main metabolic pathway of SAL in Wistar rats was methylation and *** excretion was the main excretion pathway,indicating that liver played a very important role in the metabolism of SAL.
