AIM Cryptotanshinone (CTN), one of the major constituents of tanshinones, was investigated for hepatoprotective effects in D-galactosamine (D-GalN)/lipopolysaccharide (LPS)-induced fulminant hepatic failure mouse *** CTN (20 or 40 mg·kg-1) was orally administered 12 and 1 h prior to D-GaiN (700 mg·kg-1)/LPS (10 μg·kg-1) *** Pretreatment with CTN markedly protected mice from lethal liver injury, which has known to be associated with a massive hepatocyte ***, CTN significantly blocked the elevation of TNF-αt and alanine aminotransferase (ALT) and aspartate aminotransferase (AST) as *** also remarkably reduced DNA fragmentation in the liver, which correlated with blockade of caspase 3,caspase 8, and caspase 9 ***, increased cytosolic cytochrome c protein was reduced by *** addition, CTN significantly suppressed the activation of JNK, ERK and p38 induced by D-GalN/LPS, and phosphorylation of TAK1 as ***, CTN significantly inhibited the activation of nuclear factor (NF)-κB and suppressed production of inflammatory *** These findings suggested that CTN prevents D-GaiN/LPS-induced fulminant hepatic failure, and this protection is likely associated with its anti-apoptotic activity and the down-regulation of MAPK activity associated at least in part with suppressing NF-κB.

AIM To investigated the protective effects of the extract of *** against Aβ1-42 induced injury in vivo by determination of the learning and memory ability as well as expressions of activated cAMP response element binding protein by phosphoration (pCREB) and *** 40 SD rats were randomly divided into 4 groups : control, model and treatment groups with donepezil hydrochloride and extract with *** by intragastric *** Alzheimer's disease (AD) model was established by injection of agglomerated Aβ1-42 in rat's *** treating 28 d, the Y-maze was used to test the learning and memory ability, the histomorphological changes of neurons were observed by Nissl's staining, immunohistochemistry and Western blotting were used to test the expression changes of pCREB and *** The results of Y-maze showed the total reaction time and error number of the treatment group with C Songaricum [(23.5 ± 3.35) min, (10.1 ± 2.46) rain, respectively] were significantly lower than model group [(31.44 ±: 5.93) min, (16.11 ± 3.74) min].Histomorphological observation showed the neurons in CA1 region of hippocampal in model rats were obviously damaged, which were manifested by severely decrease of Nissl body and cytoplasm vacuolar degeneration, while this pathological changes could be proved after *** *** and Westera blotting results showed the the extract of *** increased the expressions of pCREB and NGF comparing with the model *** The extract of the *** might improve the learning and memory ability as well as neurons histomorphology in AD rats induced by Aβ1-42 partly dues to regulate the expression of pCREB and its downstream target NGF.

AIM To investigate the effects of curcumin on neonatal rat cardiac myocytes exposure to high-glucose and the mechanisms *** Neonatal rat cardiomyocytes were cultured in normal-glucose (NG, 5.5 mmol-L-1) or high-glucose (HG, 30 mmol·L-1) *** HG-cultured cardiomyocytes were treated with the concentration of 10 μg·L-1 *** to and after the HG injury the following were determined: the cell viability was assayed by CCK8, LDH activity and ROS generation was examined by biochemical tests, cardiomyocyte death and apoptosis were detected by TUNEL and *** the interrelated proteins were measured by Western blot to investigate the involved signaling *** Compared with NG-cultured cardiomyocytes, the LDH level and ROS generation were increased significantly in HG-cultured ***, HG induced apoptotic cell *** could significantly increased the cell viability , decrease the activity of LDH and inhibit the ROS ***, curcumin decreased HG-indnced apoptosis through upregulating Bcl-2 and downregulating Bax proteins *** Collectively, these results strongly suggest that curcumin could reduce cardiac myocytes apoptosis exposure to high-glucose by scavenging ROS and attenuating cell death.

AIM The present study was designed to determine whether epimedium flavonoids (EF) had effect on the development of experimental autoimmune encephalomyelitis (EAE) in rats and to elucidate its underlying *** EAE was induced by irnmunization of adult female Lewis rats with partially purified myelin basic protein (MBP) prepared from guinea-pig spinal cord *** was administrated intragastrically once a day after immunization until day 14 post *** staining, enzyme-linked immunosorbent assay (ELISA), biochemical methods and western blotting approaches were used to evaluate the disease incidence and severity, neuroinflannnatory and neurotrophie response in the central nervous *** Intragastrical administration of EF (20 and 60 mg· kg-1) significantly reduced clinical score of neurological deficit in EAE rats;alleviated demyelination and inflammatory infiltration ; and inhibited astrocytes activation, production of proinflammatory molecules such as interleukin-lβ (IL-13), tumour necrosis factor-α, nitric oxide and nuclear transcription factor in the spinal cord of EAE *** with EF also enhanced the expression of 2', 3'-cyclic nucleotide 3'-phosphohydrolase and nerve growth factor, increased the number of oligodendrocytes and protected the ultrastructure of myelin sheaths and axons in the spinal cord of EAE *** Our results showed that EF inhibited the development of partial MBP-induced EAE in *** effect involved reducing neuroinflammation and enhancing myelination and neurotrophins and our findings suggest that EF may be useful for the treatment of multiple sclerosis.
