AIM Our previous study has demonstrated the therapeutic potential of bone marrow derived-neural stem cells (BM-NSCs) in CNS disorders; however, the beneficial effects are modest duo to the poor survival and low neural differentiation *** this study, a new pharmacologic strategy targeted at anti-inflammation and anti-oxidation to promote the survival and differentiation of BM-NSCs thereby enhancing their therapeutic efficacy was *** We firstly generated and identified the BM-NSCs, then investigated the influnence of Sal B on proliferation and differentiation of the cells in the non-permissive circumstances by immunocytochemical staining, flow cytometry and ELISA *** We demonstrate that Sal B possesses the ability to promote BM-NSCs proliferation in a dose dependent manner; While in differentiation medium, Sal B promoted nestin+BM-NSCs differentiated into greater numbers of NeuN+ and NF-M + neurons; upon exposure to H2O2, Sol B exerted neuroprotective effects against H2O2-induced cytotoxicity, facilitating the cells survival in oxidative stress, inhibiting the apoptosis and LDH leakage,displaying a dose-dependent protective effects on *** of the above activities may be associated with the induction of a crucial NSC-related growth factor, brain-derived neurotrophic factor expression in *** Sol B may be a potential drug to upgrade the therapeutic efficiency of BM-NSCs in CNS diseases.

AIM To evaluate clinical efficacy and safety of AOBO-001 oral capsule for the treatment of Urinary incontinence (UI).METHODS The Phase Ⅱ a clinical trial was a randomized, double-blind, placebo-controlled, parallel-group study in adult patients with symptoms related to OAB, and was conducted at five centers in the United ***-six subjects who successfully completed a 2-week placebo run-in period were randomized to three treatment groups for 12 weeks in a 1: 1:1 ratio of AOBO-001 1.6 g *** 3.2 g b.i.d or matching *** primary endpoints for the efficacy assessment were "Change in Average Number of Micturitions per 24 Hours" and "Change in Average Number of Urge Urinary Incontinence Episodes per 24 Hours" based upon the FDA approval standards for the "overactive bladder" treatments such as Fesoterodine Fumarate (Toviaz).RESULTS Average age was 54.5 years (range : 23 to 83 years) and 80% were *** in micturitions per 24 h : the average reductions from baseline micturitions were 1.8 for the both active treatment groups, compared with-1.2 for *** in urge incontinence per 24 h: the average reductions in urge incontinence were-2.09 and-3.39 for 1.6 g and 3.2 g (BID) doses, compared with-1.56 for placebo among subjects with baseline number of micturitions > 8 per 24 h and baseline number of urge urinary incontinence episodes > 3 per 24 *** results of our analyses are similar to the reported treatment effects observed in Toviaz pivotal *** The observed decrease in disease activity scores was clinical significant and relevant in AOBO-001 active *** confirmation is indicated in a larger efficacy study.

AIM To investigate the effects of ganoderma lucidum spores on caspase 8 and nestin in brain tissue of rats with epilepsy induced by pentylenetetrazol (PTZ), and to further study the pathogenesis of epilepsy and the anti-epilepsy mechanism of ganoderma lucidum *** Thirty-six male Sprague-Dawley rats were divided into control, PTZ epilepsy and ganoderma lucidum spores (GLS) *** The results showed that caspase 8-positive cell in rats of PIZ model group were higher than normal control and GLS group, while the content of caspase 8-positive cell were decreased in the GLS group compared with the epileptic model *** content of nestin were higher in PTZ model group than those of control group and GLS group at in cortex and hippocampus,also The content of nestin in GLS group were higher than control *** Ganoderma lucidum spores can effectively reduce the expression of apoptosis cell and inhibit the caspase 8 activations in the epileptic rat's brain tissue, strengthen the function of anti-oxidize in brain tissue, and can supply recovering of epilepsy nerve cell with important basis by reducing the content of nestin neural internality protect mechanism can be strengthened to reduce the damage on nerve system induced by falling *** lucidum spores is an assistant medicine to cure epilepsy, it can be very useful in clinic application.

AIM Interactions of tumor cells with endothelial cells (EC), for example tumor cell adhesion and migration through the EC layer, are essential components of tumor *** calcium regulation and cytoskeletal proteins in both tumor cells and EC provide important signaling mechanisms for the growth, angiogenesis and metastasis of tumor *** the present study,we investigated the influence of tumor cell-EC interactions on the intracellular calcium ion concentration ([Ca2+] i) and vimentin in *** [Ca2 +] i was measured by confocal microscopy in human umbilical vein endothehal cells (HUVEC) before and after the addition of PC-3M prostate carcinoma *** expression and structural organization of vimentin in HUVEC was analyzed by immunofluorescence *** effect of heparin and nifidipine on EC [Ca2+]i and vimentin were ***, a known disruptor of intermediate filaments, was used to induce the collapse of vimentin filaments, and the distribution of calcium ions in ECs was *** PC-3M cells induced a rapid [Ca2+]i elevation and lead to altered vimentin distribution in EC at the sites of tumor cell-EC *** cell-induced [Ca2 +] i elevation was significantly inhibited by heparin and nifidipine; however,heparin and nifidipine had no obvious effects on the expression and organization of ***-induced collapse of vimentin filaments shifted the distribution of calcium ions from the cytoplasm to the nucleus and perinuclear field in *** Rapid [Ca2 +] i elevation and alterations in vimentin distribution occur in EC at the sites of tumor cell-EC contact; the structural integrity of vimentin is partly related to the intracellular calcium ion distribution and concentration.

AIM Oridonin, a natural tetracycline diterpenoid isolated from Chinese herb Rabdosia rubescens, has been reported to be a potent cytotoxic agent against various ***, the effect of oridonin on highly metastatic breast cancer cells has not been *** AND RESLUTS In this study, we examined the anti-proliferation and anti-metastasis effects of oridonin in breast cancer cells MCF-7 and MDA-MB-231, and found it could induce a potent growth inhibition on them in a time-dependent and dose-dependent *** on the flow cytometric analysis, oridonin suppressed MCF-7 cells growth by cell cycle arrest at the G2/M phase and caused accumulation of MDA-MB-231 cells in sub-G1 *** induced apoptosis effect of oridonin was further confirmed by morphologic characteristics assay and TUNEL *** analysis showed that γH2AX-containing nuclear foci were significant in oridonin-treated MDA-MB-231 ***, oridonin significantly suppressed cell migration and invasion,as well as decreases MMP-2/MMP-9 activation and inhibited the expression of Integrin β1 and FAK in breast cancer *** Oridonin inhibited the growth and induced apoptosis in breast cancer cells, which might be related to DNA damage and activation of intrinsic or extrinsic apoptotic ***, oridonin inhibited tumor invasion and metastasis in vitro possibly via decreasing the expression of MMPs and inhibiting the Integrinβ1/FAK pathway in MDA-MB-231 cells.

AIM To study correlation of medicine ingredients and neurotransmitters in brain interstitial fluid (BIF) of cerebral ischemic *** Sprague-Dawley rats (n =12) were randomized into two *** in SLT (Sailuotong capsule) group were administrated with SLT(150 mg·kg-1) by garage and model rats were administrated with distilled water for 7 d before middle carotid artery occlusion (MCAO).Rats' BIF samples were collected by microdialysis and rats suffered from permanent cerebral ischemia during sampling under isoflurane ***-MS/MS was employed for medicine ingredients detection while HPLC-ECD system for neurotransmitter *** study was pursued for the correlation of certain endogenous and exogenous substances in rats'*** Three medicine ingredients from SLT were detected by LC-MS/MS, namely bilobalide (BB), crocin and ginkgolide A (GA).These ingredients comes from Folium Ginkgo and Stigma Croci, which are two herbs composition of *** neurotransmitters were determined by HPLC-ECD system, namely serotonin (5-HT), dopamine (DA), glutamate (Glu), γ-amino butyric acid (GABA).Neurotransmitter levels in rats BIF of SLT group were significantly lower than that of model *** correlation analysis showed that dynamic change of BB and GA were highly correlated with four neurotransmitter level *** SLT down-regulate both monoamines and amino acids levels, which increase dramatically after rats subjected to *** suggests, maybe, SLT protect ischemic brain by down regulating over-released *** aided correlation study suggests BB and GA are the most likely effective ingredients for regulating the neurotransmitters.

AIM To investigate the protective effect of carnosic acid（CA） on the acute oxidative liver injury induced by thioacetamide （TAA） and explore its molecular mechanism from the pathway of PI3K/*** Forty eight KM male mice were divided into six groups（n =8）:（A） control group;（B） carnosic acid group(130 mg·kg);（C） TAA group(50 mg·kg);（D） carnosic acid pretreatment group plus TAA;（E） D plus LY294002 treatment group(7 mg·kg );（F） D plus ZnPP treatment group (25 mg·kg ).CA was administered orally to mice once a day for three consecutive days while the control group and TAA group received *** hour after the last treatment of CA,The mouse in group C,D,E,F were given intraperitoneal injection of TAA, group A and B were given vehicle;1 hr later,the group of E and F were given intraperitoneal injection of LY294002 and ZnPP respectively. 18 hrs later,blood and liver tissue samples were *** evaluation includes the activities of serum transaminase, liver GSH,*** blot analysis was employed to determine the expressions of HO-1,nuclear Nrf2,Keap1,Akt, Bcl-xL and ***-PCR analysis was employed to determine the gene expressions of HO-1,Nrf2,Bcl-xL and caspase-3. RESULTS TAA caused sever liver damage,while CA pretreatment provided hepatoprotection against TAA-induced liver injury, which could be observed from an improved liver histological structure,significant decrease of serum transaminase activities and inhibition of lipid *** also enhanced GSH-Px activities and attenuated glutathione depletion by TAA in *** addition,CA increased the nuclear translocation and expressions of Nrf2,HO-1 and p-Akt,while markedly reduced the level of Keapl *** also increased the expression level of Bcl-xL and reduced the level of ***294002 and ZnPP decreased the protective effect of *** CA increases the expression of HO-1 through PI3K/Akt-Nrl2 pathway and protects hepatocytes against the damage caused by TA

AIM To find out the effects of Morinda Officinalis capsule on the bone mineral density and metabolism in ovariectomized *** Rats were ovariectomized and an osteoporotic animal model was built,by which the organ indexes,the difference of the bone mineral density,bone minerals,tartrate-resistant acid phosphatase（TRAP）,alkaline phosphatase（AKP）, osteocalcin（OC）,calcium（Ca）,phosphorus（P）,1,25-dihydroxyvitaminD(1,25（OH）VD),parathyroid hormone（PTH） and osteogenic growth peptide（OGP） concentration in the blood,as well as pyridinium（PYD） concentration in the urine before and after administration was to be found out,bone's physical,biomechanical and morphological indexes were measured and the relevant parameters were calculated so as to observe the effects of the *** Compared with model Group,the 3-month administration of Morinda Officinalis capsule via oral gavage led to a significant increase（P<0.05 ） in serm OC,Ca and Zn content,vertebral maximum load,a highly significant increase（P<0.01 ） in vertebral bone mineral density（BMD）,serm OGP,Percentage of Tibial Cortical Bone Area（%***） and decrease（P<0.01） in urine PYD,serm AKP and TRAP,percentage of bone marrow cavity area （%***） and a significant increase（P<0.05 ） in were observed at high,medium and low doses of Morinda Officinalis capsule, which were 810,270 and 90 mg·kg respectively;a highly significant increase in serum 1,25（OH）VD（P<0.01） and a significant increase（P<0.05） in femoral ash weight,femoral maximum load and modulus,percentage of vertebral trabecular bone area（% ***） were observed at high and medium doses of Morinda Officinalis capsule,which were 810 and 270 mg·kg,respectively;a significant increase in the femoral mineral density and vertebral modulus（P<0.01） as well as in the iron,copper,calcium and magnesium concentrations,the displacement of femur and vertebra,area of cortical bone（***）,trabecular thickness（***） and trabecular number（Tb.N）(P<0.05) and a significant decrease（P

AIM To compare the anti-cancer effects of furanodiene andβ-elemene in human lung cancer cells and to explore the effects and mechanisms when combined treatment of furanodiene and *** MTT assay and flow cytometry were used to detect the anti-proliferative effects and cell cycle distribution,***-time PCR was used to show the mRNA levels of the related *** blot assay and immunocytochemieal labeling were used to detect the expression and localization of proteins,*** Furanodiene concentration-dependently inhibited the cell proliferation of A549,NIH-H1299,and 95-D lung cancer cells.p-Elemene,another terpenoid isolated from Rhizoma Curcumae,exhibited weaker anti-proliferative effects in A549 and NIH-H1299 cells and activities similar to furanodiene in 95-D *** treatment of furanodiene with paclitaxel showed synergetic activity in NIH-H1299 and 95-D *** with the treatment of paclitaxel alone in 95-D cells,the combined treatment reduced the mitotic phase cells while increased G1 phase *** the group of combined treatment,the protein levels of cyclin D,full-PARP,pro-caspase 3,pro-caspase 7 and Bcl-2 were down-regulated while those of p-p53 and Bax were up-regulated as compared with the treatment of furanodiene or paclitaxel ***,combined treatment did not increase the protein levels of endoplasmic reticulum stress-related proteins,such as binding immunoglobulin protein（BIP） and C/EBP homologous protein （CHOP）,though the mRNA and protein levels of these molecules and the accumulation of CHOP in the nucleus were significantly up-regulated after furanodiene ***,the protein levels of integrinβ4,paxillin and FAK were also obviously down-regulated in the combined treatment *** Furanodiene has considerable anti-cancer proliferation capacity compared with that ofβ-elemene in *** treatment of furanodiene with paclitaxel showed synergetic anti-c
