Abstract Zinc is one of the demand essential elements and its deficiency is a critical nutritional and health problem in the *** organic forms of trace element are considered more bioavailable than the inorganic *** is a very important attempt and choice to obtain bound organic zinc by microorganism ***,twenty-two strains which tolerated high zinc were screened and isolated from natural environment,and only one strain ***-1108 grew well with 10,000μg/mL zinc(II).According to morphological,18S rDNA sequence analysis,biophysical and biochemical characteristics,***-1108 is identified as Fusarium *** cells zinc content in ***-1108 was 6.7mg/g dry *** bioavailability in rats confirmed that ***-1108 had higher absorption efficiency than gluconate *** study would bring new ideas for finding new zinc supplements from natural environment.

Using the 16S rDNA clone library technique,a priliminary study was conducted on the community diversity of endophytic bacteria in seeds of Xisha wild *** results indicated that the endophytic bacterial communities contained 42 Operational Taxonomic Units(OTUs ) in their clone libraries respectively,involving 33 genus from 4 groups of Proteobacteria,Firmicutes,Bacteroidetes and *** dominant bacterium(abundance) were Enterobacter sp.(17.55%),Bacillus sp. (16.79%o),Acinetobacter sp.(6.11%),Pseudomonas sp.(6.11%) and Piscinibacter sp.(6.11%).To our knowledge,this was the first report of the study on endophytic bacterial communities in the Noni seeds with culture-independent method.

Chlorella vulgaris,which could serve as a potential source of food and energy because of its photosynthetic efficiency,is a genus of single-celled green algae,belonging to the phylum *** our study,a pathogenic organism targeting *** was discovered.18S rRNA gene sequence analysis revealed that it shared 99.0%homology with Pseudobodo *** electron microscope analysis showed that Pseudobodo sp cells were approximately 4-5 microns long,biflagellate with an anterior collar around the anterior part of the cell in unstressed feeding *** predatory method was direct attack on *** because the crack activity was lost after its lysates were filtered through 0.22um *** the initial host,Pseudobodo sp KD51 could also kill other algae,indicating its relatively wide predatory *** stability,pH and salinity tolerance were tested to understand its predatory activities,and the results showed that it was heat-sensitive,and pH and salinity tolerant.

Maltoporin(LamB) is a family of outer membrane *** has been no report of immunological characteristics of LamB in Vibrio species so *** this study,lamB genes from eight Vibrio strains were cloned and *** bioinformatics analysis indicated that sequence similarities of LamB proteins were ranged from 46.7%to 81.1%.Further,the result showed that their antigenic epitopes were highly conserved implying that LamB might be a shared antigen among *** blot of rabbit serum against recombinant LamB from ***(ATCC 33787) with cell lysates of 18 Vibrio strains showed *** observed on cell lysates of Vibrio strains immunoblotted with the anti-LamB serum ranged between 40 and 49 *** whole-cell ELISA assay further confirmed that the antiserum of recombinant LamB recognized the tested Vibrio ***,the cross-protective property of recombinant LamB was evaluated through vaccination and subsequent challenge with heterogeneous virulent Vibrio strains in *** relative percent survival(RPS) of the vaccinated group varied from 54.1%to 77.8%,showing that zebrafish were protected from Vibrio infection after immunization with LamB *** cumulative evidences in this study suggested that LamB was a conserved antigen among tested Vibrio species and might be a potentially versatile vaccine candidate for the prevention of vibriosis.

The traditional apple plant regions in China have entered the renewal period while the replant problems restrict the development of apple ***,it is urgent to disclose the bacteria community both in replant problem apple orchard soil and in high yield apple orchard soil and furthermore, reveal the relationship between bacteria community and apple replant problems.T-RFLP(Terminal Restriction Fragment Length Polymorphism) was adopted in this paper,and the soil samples covered 9 counties in Bohai Bay Zone at 0-20cm and 20-40cm horizon *** results showed that there was obvious difference of bacteria community between replant problem apple orchard soil and high yield apple orchard soil,and the latter owned higher index value as for the bacteria diversity and *** apple orchard soil in Bohai Bay Zone had formed the steady native bacteria structure and become the predominance community in the adaption to the *** addition,the amounts and the proportion of Firmicutes,Y-proteobacteria,Actinobacteria in apple orchard soil might be one of the most important reasons for apple replant problem in this region.

The glucoamylase gene(amy A) was amplified from Rhizopus oryzae 3.866 total DNA extracts. The result showed that the sequence of amyA was consisted of 2049 bp,and encoded 604 amino *** xylanase gene(xynA) was amplified from Paenibacillus sp.H10-3 total DNA *** result showed that the mature peptide sequence of xynA was consisted of 636bp,and encoded 211 amino ***,the mature peptide sequence of amyA gene,sequence of linker and sequence of xyn A gene were spliced by overlap extension PCR(SOE-PCR),which amyA-l-xynA was *** fusion gene was then inserted into a Pichia pastoris expression vector pPIC9 to produce the recombinant expression plasmid p?*** using electric shocks,we successfully transformed the recombinant pP\C9-amyA-l-xynA into Pichia pastoris GS115,which AX 11 was obtained..The maximum yield of the recombinant glucoamylase and xylanase in AX11 culture medium were 5.8U/mL and 32.3U/mL, respectively.

A novel red-pink pigmented strain,designated R2-4,was isolated from a glacial front soil near Ny-Alesund,Svalbard Archipelago,*** cells were Gram-staining-negative and rod-shaped. Growth occurred at 4-30℃（optimum,20-22℃）,at pH 6.0-9.0（optimum,pH 7.0） and with 0%Nacl. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain R2-4 belonged to the genus *** 16S rRNA gene sequence similarity between strain R2-4 and type strains of related species ranged from 94.51 to 96.08%.Strain R2-4 contained iso- and anteiso-branched C15 fatty acids, summed feature 3（16:1ω6c and/orω7c ）,summed feature 4（17:1 anteiso Band/or iso I） and the unsaturated fatty acid 16:1ω5c as the major cellular fatty acids,MK-7 as the major respiratory quinine, phosphatidylethanolamine（PE） as the main polar *** DNA G+C content of strain R2-4 was 61.6mol%.On the basis of phylogenetic,physiological and chemotaxonomic data,strain R2-4 is considered to represent a novel species of the genus Hymenobacter,for which the name Hymenobacter articus sp.,*** proposed,The type strain is R2-4（CCTCC AB 2012104=KACC 16881）.

A novel aerobic bacterium,designated strain LAM612,was isolated from saline and alkaline soil in Shandong Province,*** of strain LAM612 were Gram-positive,endospore-forming,motile and *** range of temperature,pH and NaCl concentration for growth were 15℃to 55°C,6.0 to 7.0, and 0 to 10%*** optimal conditions were 30℃to 35℃,6.0 to 7.0 and 3%of NaCl, *** analysis of the 16S rRNA gene sequence revealed that strain LAM612 was closely related to *** KCTC 13296,*** KCTC 13178,*** KCTC 13423,and *** LMG 18474,with 98.0%,97.4%,97.2%,and 97.0%similarity,respectively. The major fatty acids of strain LAM0725 were iso-C（35.2%）,iso-C（17.3%）,anteiso-C（9.3%） and iso-C（5.0%）.The predominant menaquinones of strain LAM612 was menaquinone 7（MK-7）.The main polar lipids of strain LAM612T were diphosphatidylglycerol,phosphatidylglycerol, phosphatidylethanolamine,and six unknown *** genomic G+C content was 36.4 mol%as detected by Tm *** on the results of DNA-DNA hybridization and differential phenotypic properties,strain LAM612 could be distinguished from recognized species of the genus Lysinibacillus and was suggested to represent a novel species,for which the name Lysinibacillus huakuii *** proposed. The type strain is LAM612（=ACCC0718）.

The diversity of fungal communities associated with biological soil crusts in arid and semiarid regions are one of the least explored *** particular,the Tegger desert in China remains mycologically poorly *** this study,we investigate the diversity of fungal communities associated with different crusts(i.e.,cyanobacterial crusts,moss crusts and lichen crusts) at five locations in Shapotou Region using cultivation and 454 pyrosequencing.A total of 300 isolates were recovered and 141 isolates were phylogenetically affiliated to 84 different taxa within 34 different genera with >50%of the total isolates belonging to Alternaria,Aspergillus,Chaetomium,Cladosporium,Coniothyrium, Cryptococcus,Emericella,Fusarium,Penicillium and Phoma.A total of 50,223 sequence reads were obtained with Ascomycota,Basidiomycota and Chytridiomycota encompassing >96%of the total sequences. Fungal communities showed high diversity;the 50,223 sequence reads belonged to 1362 *** these, 938 belonged to the phylum Ascomycota,121 to the phylum Basidiomycota,2 to the phylum Zygomycota, 9 to the phylum Chytridiomycota,and 292 to unknown *** detected fungi confirmed earlier trends in fungal diversity in other habitats,quite a number of sequences representing novel taxa were *** conclude that desert crusts in the Shapotou Region harbor a large diversity of fungal communities that varies with crust type and desert locations.

Members of the Achromobacter genus are Gram-negative bacteria including both environmental and clinical isolates,which are increasingly recovered from patients with cystic fibrosis(CF) as the emerging *** better understand the features of the genus and its potential pathogenic mechanisms,6 available Achromobacter genomes were compared in this *** results revealed that:i) Achromobacter had a pan-genome size of 10,750 genes with 3,398 core genes,and a similar global classification of protein functions;ii) the Achromobacter genomes underwent a relatively low recombination that introduced nearly twice nucleotide substitutions less than the point mutation in genome evolution;iii) phylogenomic analysis based on 436 conserved proteins and average nucleotide identity (ANI) both indicated that the Achromobacter genus had the closest relationship to the human/animal pathogen Bordetella rather than to *** entire group of Achromobacter clustered with Bordetella in phylogeny,strongly suggesting a common origin,which therefore highlighted the potentially pathogenic nature of Achromobacter from the phylogenetic perspective;and iv) the CF clinical isolate possessed markedly unique genomic features discriminated from the environmental isolate,and was equipped with numerous factors that facilitate its adaptation to a pathogenic lifestyle,such as a typeⅢsecretion system,a 'polysaccharide island'(36.0 kb) of capsular/cellulose synthesis,adhesion related proteins,alcaligin biogenesis and several putative *** study provided the first comprehensive genomic comparative analysis for Achromobacter,revealed information to better understand this far less known genus on the genomic scale,and,importantly,identified potential virulence factors of the Achromobacter pathogen.
