2004年以来,在理论和应用方面对水稻分子育种进行了进一步研究。结果表明外源 DNA 片段导入水稻后,不仅能在杂交后代中稳定遗传,而且已能在生产中应用。在慈利玉米 DNA 导入水稻湘早籼8号的后代中, 在第21代(慈选4号)中仍可检测出供体玉米 DNA 特异片段,并且由其配置的杂交水稻培两优慈4(矮培64s×慈选4号)中亦可检测出供体玉米 DNA 特异片段。对杂交水稻培两优慈4进行田间活体光合作用测定,结果表明, 杂交水稻培两优慈4和父本慈选4号的光合作用均高于母本不育系矮培64s。将玉米 DNA 或高梁 DNA 导入水稻后,在后代选育出了富碘水稻和富含铁、钙、锌、V、V,水稻并推广应用。

通过花粉管通道法将菜豆几丁质酶基因(chitinase,ch)转化5个玉米自交系,D0代获得35株卡那霉素阳性植株,其中13株 PCR 阳性植株,田间抗病性鉴定结果表明,转化植株的抗玉米大小斑病能力有所提高,3株表现高抗。研究发现 DNA 溶液浓度影响转化率,当浓度为100μg/ml 时,转化率最高,为2.61%。DNA 导入时间距人工授粉时间的延长,结实率升高,但转化率显著下降,人工授粉后1 d 导入 DNA 产生的转化株最多,为19株,平均转化率为1.35%。

将高梁、玉米、谷子、小麦、长穗偃麦草、大麦、硬粒小麦等33份材料的基因组 DNA 通过花粉管通道法转入36份普通小麦,创造了大量具有利用价值的变异材料,同时选育出一系列的小麦新品系,其中将高粱的 DNA 导入小麦陇春13,成功选育出了高产、抗逆、耐盐碱小麦新品系89122,较原受体(陇春13)增产21.06%;在盐碱地比当地主栽品种 V26增产10.5%,比原受体增产22.5%,光合效率明显提高。通过 RAPD 鉴定表明,高粱基因组部分片断已经整合到了小麦染色体中。经过表型鉴定、生化分析和分子等水平的综合研究表明,利用花粉管通道法创造新品系和中间变异材料是行之有效的。
